Cento e setenta perus de corte fêmeas foram distribuídas, aleatoriamente, em cinco tratamentos com quatro repetições de oito aves cada. No terceiro e no décimo dia de vida, os animais foram submetidos ao desafio oral de SH. No sétimo, 19º e 35º de vida as aves foram eutanasiadas (n=10 por tratamento) e colhido amostras de segmentos do inglúvio, duodeno, jejuno, íleo e ceco para quantificação de SH, quantificação relativa do microrganismo Faecalibacterium prausnitzii e análise morfométrica. No decorrer do período experimental foram realizadas colheitas de suabes cloacais para pesquisa de SH. A administração do probiótico e produto de EC foi capaz de reduzir a incidência e a colonização de SH no inglúvio. Nos cecos os tratamentos reduziram a colonização de SH somente aos 19 dias de idade. Somente a dieta suplementada com AOs influenciou positivamente na quantidade de F. prausnitizii no ceco (19 e 35 dias) e não houve correlação entre a quantidade relativa desse microrganismo e o número de UFCs de SH. A excreção fecal de SH, foi influenciada pelos tratamentos a partir dos 26 dias de vida das aves, aos 34 dias todos os tratamentos reduziram a excreção fecal. A morfometria intestinal foi influenciada pelos tratamentos somente aos sete dias de vida, onde os animais que receberam AOs apresentaram maiores alturas de vilosidades de jejuno quando comparado com o grupo controle positivo. A administração dos aditivos via ração demonstrou eficácia no controle e na persistência da infecção por SH. No entanto, o uso de AOs foi mais eficaz para modular a microbiota cecal, provando que a composição da dieta pode modular populações de bactérias benéficas, mas tais microrganismos podem não estar correlacionadas com a redução da colonização cecal por SH. / The present study evaluated the use of organic acids (AOs), probiotic and competitive exclusion (CE) product administered continuously in the feed on the control Salmonella Heidelberg (SH) and action microbiota and morphometry intestinal. One hundred and seventy poults females were randomly distributed in five treatments with four replicates of eight birds each. In the 3rd and 10th day of life, the animals were submitted to the challenge with SH. In the 7th, 19th and 35th of life the birds were euthanized (n = 10) and samples of the crop, duodenum, jejunum, ileum and caeca were removed to quantification of SH, quantification relative of the Faecalibacterium prausnitzii and morphometric analysis. During the experimental period were perfomerd clocal swabs for SH research. The administration of probiotic and CE product was able to reduce the incidence and colonization of SH in the crop. In the caecum the treatments reduced SH only 19 days of age. Only the diet supplemented with AOs influenced positively on amount of F. prausnitizii in the cecum (19 and 35 days) and there was no correlation between the amount on the microorganism and the number of UFCs of SH. Fecal excretion of SH, was influenced by the treatments from 26 days of life, at 34 days all treatments reduced faecal excretion. The intestinal morphometry was influenced by the treatments only to seven days of age, where the animals that received AOs had higher jejunal villus heights of villi when compared with the positive control. The feed additives administration demonstrated effectiveness in controlling and in the persistence of the infection by SH. However, the use of AOs was most effective to modulate the cecal microbiota, proving that the composition of the diet can modulate populations of beneficial bacteria, but such microorganisms may not be correlated with the reduction of the SH / Processos: FAPESP 2015/16428-5; 2015/18350-3

Avaliação morfológica do intestino e hematológica de aves de corte (Gallus gallus domesticus) infectados experimentalmente por Salmonella enteritidis e submetidos ao tratamento por exclusão competitiva /

Orientador: Isabel Cristina Boleli / Banca: Silvana Martinez Baraldi Artoni / Banca: Nilce Maria Soares Queiroz Gama / Resumo: O experimento foi realizado com o objetivo de avaliar os pesos corporal (PC), do fígado (PF) e da bursa de Fabrício (PB), o perfil hematológico (eritrograma, leucograma e glicemia), a integridade e o desenvolvimento da mucosa intestinal de pintos de corte de submetidos ao tratamento por exclusão competitiva (EC) antes e após infecção experimental por Salmonella Enteritidis (SE). Foram utilizados 128 pintos, distribuídos num delineamento ao acaso com esquema fatorial 2 x 4 [2 sexos e 4 tratamentos (NI: não infectados com SE; I: infectados com SE; IEC: infectados com SE e tratados com EC 24 h após infecção; ECI: tratados com EC e infectados com SE 24 h após tratamento)]. Os animais foram sacrificados com 1, 3, 5 e 7 dias pós-infecção (dpi) para obtenção do conteúdo cecal (microbiológico), fígado, bursa de Fabrício e fragmentos do intestino delgado. Os dados demonstram que o uso de EC após infecção não evita a colonização cecal por SE, mas que a mesma é evitada quando a EC é realizada antes da infecção. A partir do 7° dpi as aves infectadas apresentaram os menores valores de PC e PB, e não se evidenciou diferenças na glicemia e no PF. As alterações do eritrograma ficaram restritas aos três primeiros dpi e os resultados do leucograma mostraram uma resposta sexo-específica frente à infecção por SE. Em relação à integridade, o presente trabalho mostrou que os vilos do intestino delgado dos pintos de corte apresentam um grande processo de renovação celular ao final da primeira semana de vida e que EC acelera este processo. O desenvolvimento intestinal variou de acordo com o segmento, o sexo, o tratamento e a idade analisada. / Abstract: The present experiment was carried out with the objective to evaluate the weights corporal 0NC), of the liver 0NF) and bursa of Fabrício (WB), the hematology profile (erytrogram, leucogram and glycemia), the integrity and the development of the intestinal mucosa by young chickens submitted to competitive exclusion treatment (CE) before and after experimental infection with Salmonella Enteritidis (SE). 128 young chickens, distributed in completely randomized design in 4 x 2 factorial arrangement [had been used 2 sex and 4 treatments (NI: not infection with SE - Control; I: infection with SE; ICE: infection with SE and treated with CE 24 h after infection; CEI: treated with CE and infection with SE 24 h after treatment)]. The animais had been sacrificed with 1, 3, 5 and 7 days post infection (dpi) for attainment of the cecal content (microbiological), liver, bursa of Fabrício and fragments of the thin intestine. The data demonstrate that the CE use after infection does not prevent the cecal settling for SE, but that the same one is prevented when the CE is carried through before the infection. From 7° dpi the infections birds had presented the lesser values of WC and WB, and it did not prove differences in the glycemia and the WF. The alterations of the erytrogram had been restricted to three first dpi, and the results of the leucogram had shown to a sex-specific reply front to the infection for SE. In relation to the integrity, the present work showed that the vilos of the thin intestine of young chickens present a great process of cellular renewal to the end of the first week of life and that CE speeds up this processo The intestinal development in accordance with varied the segment, the sex, the treatment and the analyzed age. / Mestre

Statistical validation of limiting similarity and negative co-occurrence null models : Extending the models to gain insights into sub-community patterns of community assembly

Competition between species is believed to lead to patterns of either competitive exclusion or limiting similarity within ecological communities; however, to date the amount of support for either as an outcome has been relatively weak. The two classes of null model commonly used to assess co-occurrence and limiting similarity have both been well studied for statistical performance; however, the methods used to evaluate their performance, particularly in terms of type II statistical errors, may have resulted in the underreporting of both patterns in the communities tested. The overall purpose of this study was to evaluate the efficacy of the negative co-occurrence and limiting similarity null models to detect patterns believed to result from competition between species and to develop an improved method for detecting said patterns. The null models were tested using synthetic but biologically realistic presence-absence matrices for both type I and type II error rate estimations. The effectiveness of the null models was evaluated with respect to community dimension (number of species × number of plots), and amount of pattern within the community. A novel method of subsetting species was developed to assess communities for patterns of co-occurrence and limiting similarity and four methods were assessed for their ability to isolate the species contributing signal to the pattern. Both classes of null model provided acceptable type I and type II error rates when matrices of more than 5 species and more than 5 plots were tested. When patterns of negative co-occurrence or limiting similarity were add to all species both null models were able to detect significant pattern (β > 0.95); however, when pattern was added to only a proportion of species the ability of the null models to detect pattern deteriorated rapidly with proportions of 80% or less. The use of species subsetting was able to detect significant pattern of both co-occurrence and limiting similarity when fewer than 80% of species were contributing signal but was dependent on the metric used for the limiting similarity null model. The ability of frequent pattern mining to isolate the species contributing signal shows promise; however, a more thorough evaluation is required in order to confirm or deny its utility.

Réseaux d'interactions, biodiversité et services éco-systémiques en milieu agricole : que nous apprennent les coléoptères carabiques ? / Interaction networks, biodiversity and ecosystem services in agricultural areas : lessons from carabid beetles communities

Le contrôle des ravageurs est l'un des principaux services éco-systémiques rendus par la biodiversité à l'agriculture. Les communautés d'insectes auxiliaires, hébergées par les espaces agricoles présentent des niveaux de diversité, spécifique et fonctionnelle, élevés et leurs biologie et traits de vie sont bien décrits et étudiés. Ces communautés constituent donc un excellent modèle pour aborder des questions d'intérêt à la fois fondamental et appliqué sur les mécanismes à l'origine de la biodiversité et sur l'impact de la biodiversité sur l'approvisionnement en services éco-systémiques. Dans cette thèse, nous développons une combinaison originale d'approches classiques et d'approches moléculaires de pointe pour élucider les relations trophiques au sein de la communauté de coléoptères carabiques en milieu agricole. Les coléoptères carabiques peuvent contribuer de façon significative au service de contrôle biologique mais les mécanismes généraux conditionnant cette contribution restent encore difficiles à évaluer par manque d'approche systémique pour l'analyse de leurs réseaux d'interactions.L'étude sans a priori du régime alimentaire de la communauté de carabes dans un paysage agricole typique révèle un partitionnement de la ressource entre groupes d'espèces. L'étude expérimentale des interactions entre espèces avec un régime alimentaire similaire montre une différentiation des activités spatio-temporelles à fine échelle. D'un point de vue fondamental, ces résultats semblent démontrer la prépondérance des processus déterministes (partage de niche) par rapport aux processus neutres (stochasticité environnementale) pour expliquer la coexistence des espèces. D'un point de vue appliqué, l'importance de la ressource dans la structuration des communautés de carabes fournit un levier d'action potentiel pour l'élaboration de stratégies de gestion afin d’optimiser leur fonction de régulateurs naturels. / Biological control is one of the main ecosystem services provided by biodiversity in agroecosystems. Communities of beneficial insects, hosted by agricultural areas exhibit high levels of species and functional diversity, and their biology and life history traits are well described today. These communities are therefore an excellent model for addressing issues of fundamental and applied interest about mechanisms at the origin of biodiversity and its impacts on the supply of ecosystem services. In this thesis, we develop an original combination of advanced molecular approaches and more traditional methods in order to elucidate trophic interaction network within the community of carabid beetles in agricultural areas. The carabid beetles can significantly contribute to the service of biological control, but their contribution and beneficial conditions are difficult to assess because of their opportunistic and plastic feeding behavior. A without a priori investigation of carabid diet at community level in a typical agricultural landscape reveals a resource partitioning between groups of species. Additional experimental studies in laboratory conditions indicate that interspecific competition could be the mechanism generating this partitioning. From a fundamental point of view, these results suggest a preponderance of deterministic processes (niche partitioning) compared to neutral processes (environmental stochasticity) to explain the coexistence of species. From an applied point of view, the importance of the resource in structuring carabid communities provides a potential lever of action for the development of efficient management strategies optimizing carabid function as crop auxiliaries.

Characterization of the plankton community in the lower Rincon Delta: Investigations regarding new approaches to management

In light of increasing harmful algal blooms and the need to protect human health and aquatic resources, proactive management approaches merit further study. For this purpose I conducted field samplings to characterize plankton community composition and laboratory experiments to test some approaches to new management schemes in the lower Rincon Delta. On site measurements and microscopic analysis showed that environmental parameters and plankton community composition varied considerably among sampling stations and sampling dates. A recent modeling study suggested that manipulation of freshwater inflow to estuaries might prevent phytoplankton blooms and enhance secondary productivity. To test this theory I conducted three semi-continuous design and flow-through incubation design experiments using natural plankton assemblages. I investigated the effect of two different pulsing regimes of inflow and nutrient loading on zooplankton densities, and phytoplankton biomass and diversity. Despite differences in zooplankton structure and phytoplankton community composition between the two experiment designs, the results confirmed that pulsed inflows might alter plankton dynamics. My findings showed that 3-day pulse treatments consistently supported greater zooplankton densities and higher phytoplankton species diversity when compared to 1-day pulse treatments. In addition, accumulation of phytoplankton biovolume remained low during 3-day pulse treatments. Differences in zooplankton performance between 3-day pulse and 1-day pulse inflow treatments were likely due to the ability of phytoplankton to uptake and store greater amounts of nutrients under conditions of 3-day pulse inflow. This resulted in food of higher quality for zooplankton, and might have supported greater zooplankton population growth rates. Additionally, in an attempt to understand the mechanisms leading to high biodiversity in aquatic ecosystems, I built a resource-storage model and studied the effects of resource-storage on competition of multiple phytoplankton species on multiple abiotic resources. I compared this model with a well-established multi-species competition model. My results showed that for certain species combinations a resource-storage-based model can generate dissimilar outcomes when compared to a model without resource-storage.
