Wild Music: Ideologies of Exoticism in Two Ukrainian Borderlands

This dissertation presents case studies of two distinct Ukrainian borderland groups: the Crimean Tatars of Crimea, and the Hutsuls of the Carpathian Mountains – two human collectivities that are both, today, Ukrainian by citizenship. Both of these groups also embody dominant stereotypes of otherness in Ukraine – Hutsuls as the ideal Herderian romantic folk, and Crimean Tatars as the menacing, mysterious, “oriental” other. This dissertation traces how historical stereotypes of both of these groups as “wild” have shaped and defined their contemporary expressive cultures, specifically addressing how stereotypes of wildness—or hegemonic conceptions of “otherness”—manifest on the ground within the communities who bear the stigma of such entrenched histories of exoticism. This ethnographic project focuses on music as a medium for challenging and reinforcing ideologies of exoticism, demonstrating how insiders and outsiders in both cases draw upon indigenous musical tropes to express or subvert stereotypes of “wildness.” By analyzing how music energizes social and political agendas for borderland groups such as the Hutsuls and Crimean Tatars, this project emphasizes the copresence of alternate subalterities within the nation-state, demonstrating the degrees to which a post-socialist, diverse and fractured state such as Ukraine is constructed through imaginings of its internal, peripheral Others.

Use of recombinant antigen in the diagnosis of Crimean-Congo haemorrhagic fever virus infection

A dissertation submitted to the Faculty of Health Sciences University of the Witwatersrand, Johannesburg for the degree of Master of Science. / The Special Pathogens Unit (SPU) of the National Institute for Virology has diagnosed a total of 158 cases of Crimean-Congo haemorrhagic fever (CCHF) from the time that the disease was first recognized in South Africa in 1981 up until the end of 2000. The virus has a propensity to cause nosocomial infections, and consequently rapid diagnosis is important for the isolation of the patient and the institution of barrier-nursing to protect medical staff and the community at large. Thus it is essential that the SPU should have the latest diagnostic and research tools available. Diagnosis of CCHF is generally confirmed by isolation of the virus, detection of viral RNA amplified by reverse transcriptase polymerase chain reaction (RT-PCR), demonstration of seroconversion or a >4-fold increase in IgG antibody titre, or detection of specific IgM antibody activity. Virus can be isolated in 1-6 days in cell culture, but the method is less sensitive for the isolation of low concentrations of virus than the use of suckling mice which, however, takes 7-9 days.

Rapid development of optimized recombinant adenoviral vaccines for biosafety level 4 viruses

This thesis describes the production of adenovirus-based vaccines containing codon-optimized genes from Nipah virus and Crimean-Congo Hemorrhagic Fever virus. Genes encoding envelope proteins from Crimean-Congo Hemorrhagic Fever Virus and Nipah Virus were codon-optimized for translation in human cells and constructed using a modified method of non-gapped gene synthesis, while the entire M segment encoding the glycoprotein precursor for Crimean-Congo Hemorrhagic Fever Virus was commercially synthesized. Genes were cloned into recombinant human adenovirus serotype 5 and the resulting viral particles were amplified, titred and analyzed for in vivo efficacy. Results show that a modified method of non-gapped gene synthesis is an effective and efficient method of producing antigen-encoded DNA and at a fraction of the cost and time required for commercial synthesis. Furthermore, adenovirus-based vaccines induce both cellular and humoral immune responses providing for a highly efficacious vaccine during potential disease outbreaks, where time to completion is of utmost importance. This study has shown that recombinant adenoviral vaccines for Crimean-Congo Hemorrhagic Fever virus and Nipah virus can be produced rapidly and efficiently from virtual DNA sequence to optimized recombinant vaccines in just eight months.
