Clarifying the Role of the CST Complex in DNA Replication and Repair

<p>Ends of linear chromosomes are maintained by specialized structures known as telomeres. These structures are protected by a number of essential protein complexes including the shelterin complex and CST (CTC1 – STN1 – TEN1) complex. CST is an RPA-like ssDNA binding protein that is vital for telomere length maintenance <i>via</i> inhibition of telomerase and stimulation of DNA polymerase α -primase during C-strand fill-in synthesis. CST is also known to possess additional genome-wide roles in regulating DNA replication and repair including helping facilitate replication re-start at stalled forks, activating checkpoint signaling at double-strand breaks, and promoting replication origin firing. Proper and efficient repair of DNA is critical in order to protect the integrity of the genome and prevent extreme mutagenesis. Telomeres have a strong predisposition to oxidative DNA damage in the form of 8-oxoguanine caused by exposure to reactive oxygen species and free radicals. These oxidative lesions are repaired by the base-excision repair (BER) pathway. Previous work has implicated telomeric proteins such as the shelterin complex in mediating BER. Here we show for the first time that the CST complex and individual subunits robustly stimulate a myriad of proteins involved in the BER pathway including Pol β, APE1, FEN1, and LIGI. CST’s ability to augment these BER-associated proteins could be instrumental in promoting efficient DNA repair. Additionally, we find that CTC1 and STN1 are able to significantly enhance the polymerase activity of Pol δ and Pol α on both random-sequence and telomeric-sequence DNA substrates <i>in vitro</i>. What is more, we establish the ability of CST to resolve G4 structure and promote Pol δ synthesis, which we predict is a key feature of CST’s involvement in DNA replication at telomeres, which are known to form replication-inhibiting G4’s. Our results define important mechanistic insight into CST’s role in DNA replication and repair, and provide a strong foundation for future studies relating defective telomere maintenance to aging disorders and cancers which impact human health.</p>

Modelování mozku krysy / Modeling the brain of a rat

This master’s thesis deals with creation and modification of several numerical rat’s brain models which are subsequently used in electromagnetic software CST Studio Suite 2014. The first part of the thesis is focused to insertion of exciting source to the model which depends on the used approach. These sources produce signals on the surface of the rat’s brain model which are then analysed. The experimental part of the thesis dealt with three different procedures of cooking agar jelly, which were used for creation of five plate capacitors, which subsequently served for measure electric properties of phantom. The conclusion of this master’s thesis is focused on comparison between results from simulations and experimental parts with a subsequent analysis of the deviations of results and their possible causes.

The aim of this diploma thesis is to study the possibilities and problems of low-profile antennas and subsequently to design a directional antenna with spherical reflector for millimeter wavelengths. The theoretical part deals with basic information about horn and reflector antennas, and about SIW technology. In addition, the thesis deals with the specific design and modeling of the antenna using CST Microwave Studio. The simulated design achieved a sufficient bandwidth to cover ISM (57 GHz to 64 GHz) with gain 13.6 dBi at 60 GHz frequency. On fabricated antenna, the antenna impedance ratios were worse and the target bandwidth was not reached. Antenna gain was 14.33 dBi at 60 GHz frequency.

This thesis deals with optimization of coaxial cavity filters. It thoroughly describes optimization method Tuning – Space Mapping (TSM) in CST Studio. It focuses on the explanation of the fine and the coarse model and their mutual link. The whole algorithm is illustrated and tested on the third-order coaxial cavity filter working in the band of 880 – 960MHz. In order to check the function of TSM, the algorithm is applied on fourth-order filter, triplet filter and quadruplet filter. Another part of the thesis focuses on the automatization of the whole optimization procedure. Macros for automatic determination of calibration constants and automatic calibration process between the coarse and the fine model are proposed and programmed in CST. The complete optimization procedure is applied on optimization of seventh-order filter with two cross couplings.

In this thesis is discussed a filter design of a band-pass substrate integrated waveguide filter on the frequency of 5 GHz. It also includes design theory, filter synthesis, filter analysis in the program CST Microwave Studio® and the results of measurement on a vector network analyzer. The analysis results are compared with the measured results. Good agreement between simulated and measured results is observed.

Assessing effective medium theories for designing composites for nonlinear transmission lines

<p>Nonlinear transmission lines (NLTLs) are of great interest for high power microwave (HPM) generation because they can sharpen pulses to create an electromagnetic shockwave to produce oscillations from 100 MHz to low GHz. NLTLs provide frequency agility, compactness, durability and reliability, providing a solid-state radiofrequency (RF) source for producing HPM. The essential component of NLTLs is the nonlinear material, typically a dielectric that varies with voltage or a magnetic material whose permeability varies with current, incorporated in the transmission line in various topologies. This thesis presents an alternative approach involving designing composites comprised of nonlinear dielectric inclusions (barium strontium titanate (BST)) and/or nonlinear inductive inclusions (nickel zinc ferrites (NZF)) in a polymer base host material, analogous to electromagnetic interference designs that incorporate stainless steel inclusions of various shapes in a plastic to tune the composite’s electromagnetic properties at GHz. Appropriately designing NLTL composites requires predicting these effective properties both in linear (for a fixed and low voltage and current) and nonlinear regions (permittivity and permeability become voltage dependent and current dependent, respectively) prior to designing HPM systems comprised of them. As a first step, this thesis evaluates and benchmarks composites models in the commercial software CST Microwave Studios (CST MWS) to various effective medium theories (EMTs) to predict the permittivity and permeability of composites of BST and/or NZF inclusions in the linear regime, compared with experimental measurements. The manufacturing and measurement of the nonlinear composites will be briefly discussed with an analysis of the homogeneity of a composite sample using 3D X-ray scan. Long-term application of these approaches to predicting the effective nonlinear composite permittivity and permeability and future work will be discussed.</p>

The main aim of this work is to describe the history and development of Czech Television's regional news coverage in years from 1992 till 2012, using the testimonies of direct TV staff involved in this reporting beyond the available resources, thereby expanding the amount of information on this issue. To achieve this information, the oral history method was used and the subsequent verification of the information obtained was done using the literature, diploma theses, annual reports and other sources in this work specified. The result of this work is the extension and addition of new facts and information about the history of Czech Television's regional news, especially of internal character, not yet obtainable from available professional publications or public reports and data. These include information on the nature of interpersonal relations in regional news, the attempt to influence the outward processing and thematic focus of the editorial offices and the specific technical and structural development of individual regional editorial offices.
