In the present thesis called "The modern analytical methods for detection of aminoacids asociated with malignant prostate tumours" sarcosine has been studied as a potential tumor marker which has significance for the diagnosis of aggressive forms of prostate cancer. The effect of addition of sarcosine prostate line PC3 has been also studied as well. Next studied area were the the interactions with amino acid sarcosine, sodium chloride, uric acid and uracil. We have primarily focused also on the development of methods for the detection of sarcosine in urine samples of patients with Cap and prostate cells.

The subject of this thesis is the design of specific LAMP primers and LAMP analysis and its optimization. The purpose of this analysis is to specifically distinguish Xanthomonas gardneri from other pathogenic xanthomonads. Two sets of LAMP primers from the DNA sequences of hrpB and atpD genes were designed. These LAMP primer sets are highly sensitive and the detection limit of LAMP assay was found to be 0.01 l / mg of DNA. The reaction temperature of the LAMP was optimized to 64 °C to reach the maximum of amplification. LAMP can be applied to detection and identification on pathogens in plant tissues. LAMP takes less time than conventional PCR to detect bacteria. This LAMP assay has a great potintial to be applied to detection and identification of Xanthomonas genus bacteria pathogenic for tomato and pepper.

This work also deals with the attachment of per-6-deoxy-per-6-methylsulfanyl-β- cyclodextrin to gold surface of quartz crystal microbalances. Several disease processes taking place in the cells are characterized by an increase of the concentration of nucleic acids, in particular micro RNAs (miRNAs). A detection system that could selectively detect the increased presence of the miRNAs directly in the living cells in real time with nanoresolution is therefore highly desired. Fluorescent nanodiamond particles are considered promising candidates thanks to their biocompatibility, small size, allowing them to penetrate the cell membrane, and stable fluorescent defects in the crystal lattice, namely nitrogen-vacancy (NV) centres. The NV centres are the most studied colour centres of nanodiamonds due to their unique room-temperature optical properties, allowing for highly sensitive detection of changes in the magnetic field (magnetic noise) with quantum sensing techniques. For instance, the length of the T1 relaxation time NV electronic spin is greatly influenced by the presence of paramagnetic species, which causes a shortening of the T1 relaxation time depending on the proximity to the NV centres. However, for selective quantum sensing with nanodiamonds, the use of molecular transducers is necessary to bind targeted molecules with high specificity and allow their detection via the change of the NV spin properties. Part of this paper is design of new method against SEO spam, its implementation and testing performance of this method upon created testing data.

The thesis deals with the presentation of an overview of the methods used for leak detection set by technical standards. Following the analysis of the pressure records from the pipeline, a new method is proposed, which is based on existing methods and uses the determination of the direction of arrival of the wave directly at the measurement station. The proposed concept was subsequently developed in Matlab, where the ability to detect was also verified. The whole algorithm was then implemented in programmable logic controllers and a suitable communication between them was designed. The entire solution was then tested in terms of functionality.

The thesis is focused on the design of a wideband detector used for detection of HIRF field. The detector, which is based on PICA antenna, was deeply examined and its´shape was also modificated for the purpose of achieving better omnidirectional and impedance characteristics. The thesis also deals with the design of cross PICA antenna variations, which purpose is to improve pattern bandwith. The PICA antenna variations mentioned were compared with one another at the end of the work. The end of the thesis also deals with the comparison of measured results gained from the specimen made with the results gained from the simulations.

For the image changes and registrations was made computer programme (Matlab, Graphical User Interface), where the user can try the different methods decribed in teoretical part of thesis.
