Fylogeneze krvetvorby obratlovců / Origins of vertebrate hematiopoiesis

(ENGLISH) Hematopoiesis is dependent on the actions of hematopoietic stem cells (HSCs). This process is tightly controlled through a complex array of extrinsic and intrinsic factors. Even though the hematopoiesis seems to be well conserved across the disparate vertebrate animals, erythroid and thrombocytic differentiation have changed during the evolution of mammals. Specifically, adult mammalian red blood cells have the unique feature of being enucleated, and mammalian thrombocytes are not individual cells, but fragments of megakaryocytes, instead. It is likely that these enhancements provided a survival advantage to early mammalian species; however, they also bring up the question of evolutionary origin of these cells that studied using zebrafish (Danio rerio) model. First, it was necessary to generate a toolbox of a recombinant cytokines and optimized culture media that allowed us to manipulate zebrafish hematopoietic cells ex vivo in liquid and clonal cultures. Interestingly, teleost species underwent an extra duplication event during their evolution and as a result, two copies (paralogs) of some of the genes are present in zebrafish. Application of supply chain management is still relatively recent practice, but it's importance is growing over time. Today, in the era of customer centricity, in order to become competitive firms shouldn't avoid collaboration with it's supply chain partners. Purpose of my thesis was to find out how supply chain management can contribute to building relationship with customers, who are the basis of any existing company. First part summarizes knowledge about supply chain, compeptitive strategies, customers, types of supply chains and customer relationship management. Particular part is dedicated to customer service and how supply chain management can contribute to it's improving. It describes also what meaning has customer service for future competitiveness. In second part, I presented the case of Zappos.com. E-retailer, which aligning entire functional strategies and whole supply chain to common strategic purpose managed to build excellent relationship with it's customers.

The thesis analyses competitive environment of suppliers of concurrent quality control technology in industrial enterprises and research laboratories. The thesis provides concepts for a creation of competitive advantages and related higher evaluation of costs and diferentiation than competition. Further it compares technologies from various manufacturers, comparing economical and technical parametres. The evaluation of results follows a presentation of approachs to achieve the most efficient competitors fight management. The thesis also include prognosis of future conditions in the business technology sector on Czech and Slovak market and concept of an appropriate format of effective competitive intelligence. In conclusion there is a presentation of a quality and quantity concept evaluation with emphasis on economic gain of presented solution.

The dissertation deals with the topic of how the school management of Czech primary schools assign teachers to classrooms. The aim of the thesis was to define the system of assigning teachers to classrooms at schools applying pupils' differentiation and to clarify which criteria the school management follows in the assignment of teachers to the classrooms. The theoretical part was based on both domestic and foreign sources dealing with the concept of differentiation of pupils in primary education and with the choice of assignment of teachers to classrooms. Differentiation of pupils was presented in terms of the positive effects of effective education of similarly competent pupils and in terms of the view that differentiation supports the unequal distribution of pupils in classes and the subsequent recruitment of teachers applying different demands and curriculum. In addition, a model of pupils' differentiation and models of assignment of teachers to classrooms were presented, and also the theoretical model of teacher's knowledge was introduced, which was further used in the empirical part for the determination of the criteria for assigning teachers to classrooms. Regional nationalism and consequent separatist tendencies have a significant impact on the territorial (dis)integration of many countries across the globe, and Europe is no exception. These processes contributed to changes in the traditional academic understanding of states as uniform, cohesive, and continuous political entities. Nevertheless, research on territorial and societal cohesion and continuity within regional nationalist movements has so far been neglected, even though this aspect seems to be key to the successful mobilization and goal achievement of separatist movements in heterogeneous democratic societies. This thesis aims to examine the limits of uniformity and continuity, that is, a time-space differentiation in political affiliations to secessionist regional nationalism in Catalonia, including the explanation of a time-space nonstationarity in the effects of factors behind the affiliations. Catalonia is an example of region with intense cultural and inclusive-assimilationist nationalism which proliferates within an ethno-culturally heterogeneous and segregated society. Škola, základ života - Soubor školských staveb v Ostravě na Černé louce / School, the Foundation of Life – a Complex of Educational Buildings in Ostrava, Cerna louka

The location of Černá louka is situated between the city-centre of Moravská Ostrava and an area of Slezská Ostrava next to the confluence of two rivers – Ostravice and Lučina. The development at the building site of Černá louka is a result from an international competition 2010, which winner was the project Culture Meadow from studio Maxwan. The main idea of the project is to concentrate cultural and educational buildings around a culture meadow. This design has been used as a default situation for the master´s thesis. The complex of schools is situated at the parcel of existing Miniuni-site between cultural cluster Černá louka and the residential quarter Nová Karolina. The volume of the school with its front-spaces reacts to the surrounding structures. In the middle of the complex arises a half-public open space, where are located the internal entrances. Because of the placing of the school next to the city-centre the multifunctional pitch is situated on the roof of the gym. The design of the school respects principles of the age differentiation just as the horizontal integration. That´s why the substance makes up a symbiosis of 5 parts - common space, gym, first and second level, gymnasium. These parts are connected together and at the same time enable to use individual parts separately. The building of kindergarten with its garden is separated from the school because of differential needs of the smallest children.

(in English) The present thesis explores the issue of teaching heritage language learners and returnee students in English as a foreign language (EFL) classes in Czech secondary schools. The aim of the thesis is to examine the experiences of heritage language learners and returnees in the EFL classroom, their strengths and weaknesses in English, their attitude towards English language learning in terms of potential anxiety and motivation, as well as heritage language learners' wishes in relation to language instruction. These issues are investigated from the perspective of the students themselves. The theoretical part reviews the relevant literature on heritage language acquisition and teaching, and on the effects of experience abroad on language acquisition. Moreover, it is complemented by discussions of differentiated instruction and language education for the gifted, two areas which can prove helpful in terms of finding suitable solutions to the problems encountered by the target population in foreign language classes. The empirical part is qualitative and consists of the analysis of in-depth semi-structured interviews with three participants, gymnázium students from Prague.
