Diploma thesis has special education theme. This thesis deals with the auditory perception in preschool children. The goal of the work is to evaluate the level of auditory perception of children in an ordinary preschool class. We focus on listening, auditory differentiation, short-term auditory memory, auditory analysis and synthesis, and perception and reproduction of rhythm. In order to answer the research questions, we created a test which is designed in accordance to the auditory perception development tables. The results show that on average the children score 82.8 percent in the test. The most difficult part of the test is listening with the average score of 61.0 percent. On the other hand, the most successful part is the perception and reproduction of rhythm with average score of 89.2 percent. The dissertation thesis investigates the relationship between corporate social responsibility (CSR) and financial performance (FP) on the sample of 51 Eurozone banks over the period from 2008 to 2014. The investigation is based on a panel data regression analysing the financial data from Bankscope and the social performance data from CSRHub. Return on assets and the ratio of non-performing loans to total loans represent the measures of financial performance and are used as dependent variables. The results of this model have shown a positive and statistically significant CSR-FP relationship. It is argued that even though the results show statistical significance, they do not necessarily include such a strong informational value. This is caused by methodological limitations, such as potentially biased data on CSR, as well as by the theoretical ones. The main theoretical concern, detected in the dissertation thesis, is a need for redefinition of the banks' driving motives of engaging in CSR activities. Banks engaging in CSR activities for merely strategic reasons should be analysed separately on a firm-level as they may otherwise bias the empirical results. Another important aspect of the work was an argument that banks benefit from CSR mainly through the product differentiation. This could not have been tested empirically, but it is assumed that the product differentiation, for example through reputation enhancement, may play a significant role in boosting bank's profits.

Ray-finned fishes (Actinopterygii) exhibit the greatest biodiversity among vertebrates. The vast majority of extant actinopterygian fish species belong to clade Teleostei - a lineage whose significant evolutionary success might have resulted from a teleost specific whole- genome duplication (TSGD) that occurred at the onset of this group, subsequent to its divergence from the rest of actinopterygian lineages. Despite the growing body of sequenced fish genomes and analyses of their transcriptomes, the largest contribution to understanding fish genomes comes from analyses of DNA content and from cytogenetics. Genomes of ray-finned fishes and especially those of Teleostei exhibit vast diversity and rapid dynamics of repetitive DNA sequences whose variability is reflected in a wide range of fish genome sizes and in the dynamics behind karyotype differentiation. Therefore, ray-finned fishes offer a unique opportunity to study genome variability as a driving force underlying morphological and ecological diversification, evolution and adaptation. The canonical Wnt/β-catenin signalling pathway plays an important role in proliferation and differentiation of neural progenitors during embryogenesis as well as postnatally. In the present study, the effect of the Wnt signalling pathway on the differentiation potential of neonatal and adult neural stem cells (NS/PCs) isolated from subventricular zone (SVZ) of lateral ventricles and their membrane properties were studied eight days after the onset of in vitro differentiation. To manipulate Wnt signalling at different cellular levels, three transgenic mouse strains were used, which enabled inhibition or activation of the pathway using the Cre- loxP system. We showed that the activation of the Wnt signalling pathway leads to higher expression of β-catenin in both postnatal as well as adult NS/PCs, while Wnt signalling inhibition results in the opposite effect. To follow the fate of NS/PCs, the patch-clamp technique, immunocytochemistry, and Western blot were employed. After eight days of NS/PCs differentiation we identified three electrophysiologically and immunocytochemically distinct cell types of which incidence was significantly affected by the canonical Wnt signalling pathway, only in differentiated neonatal NS/PCs. Mucopolysaccharidoses are a group of diseases that belong to lysosomal storage disorders. A common sign of these monogenic multisystem diseases is a gene mutation leading to a deficiency of the lysosomal enzyme participating in glycosaminoglycan degradation. It results to their accumulation in the tissues and organs, where they cause a progressive damage. There is no efficient treatment available for most mucopolysaccharidoses. Moreover, the research is complicated because of the low prevalence and type of affected tissues. Animal models of these human diseases are used for an evaluation of newly developed therapeutic approaches. However, they also have many limitations due to the different pathogenesis and catabolic pathways of the accumulated substrates between humans and animals. Therefore, animal models are replaced by human cell models. In this thesis, the development of four mucopolysaccharidoses human cell models is reported (MPS IIID, MPS IVA, MPS IVB, MPS VI). Corresponding genes (GNS, GALNS, GLB1, ARSB) were inactivated using CRISPR/Cas9 technology, where plasmids containing specific inserts are delivered to the target human induced pluripotent stem cells (iPSC), using electroporation. Saccharomyces cerevisiae is a unicellular eukaryotic organism capable of forming organized multicellular communities - colonies and biofilms. During development, colonies of laboratory strains differentiate into specifically localized cell subpopulations - U and L cells, located in the upper and lower part of the colony, respectively. The U and L subpopulations of cells vary in morphology, metabolic processes and stress resistance. Protein granules are membrane-less "organelles" found in both unicellular and multicellular eukaryotic organisms. The formation of protein granules is related to the physiological state of the cell (e.g. chronological and replicative aging), but also to changing environmental conditions and to cellular responses to stress factors. A relatively large fraction of proteins relocalizes to some type of protein granule during the lifespan of the cell. Granule formation can increase fitness of cells, help them to cope with limiting energy resources, and plays a crucial role in the adaptation of cells to stress conditions. Localization of many proteins in the cell varies depending on its physiology.
