Changes in the intensity of road transportation in CZ - 1995-2010 Abstract This diploma thesis focuses on the development of intensity load of road network between 1995 and 2010. Theoretical part is devoted to general definition of transport and transport systems. The next part is the theoretical concept of research that is focused on the spatial interaction and road transport in terms of spatial structure. The conclusion of theoretical part is devoted to characteristics of the road network and transport resources of Czech Republic. The empirical part of the thesis is describing the methodology of data processing at first. The following is an initially analysis of the evolution of traffic intensity on the motorway and road network in Czechia. Then they are analyzed regional differences in the intensity of road traffic and their evolution over time. Finally, here are three studies. The first is focused on the road transport at inputs to districts with the regional centres. The second is focused on the development of transport in the border region and compared with the hinterland. Coming Out As A Political Act In Lgbt Movement In Turkey

This thesis analyzes the coming out action of individuals through perception of political identity. Instead of considering coming out as an individual experience, it is discussed as a political action that effects the others around the individual. This political action is examined from the Queer Theory&rsquo / s perspective of subverting the gender norms. The coming out experience of lesbians, gays and bisexuals not only has an impact of their personal environment, but also effects their relation to the LGBT movement. The importance of coming out in LGBT movement is explained through the interviews with lesbians, gays and bisexuals. Where they place themselves politically in their socialization process is analyzed. LGBT movement&rsquo / s historical background is introduced and compared with the movement in Turkey. The issues originate from Turkish society&rsquo / s social structure is indicated through interviews.

São salmonelas de composição antigênica similar e a distinção de ambas tem sido baseada em provas bioquímicas. Entretanto, este procedimento é laborioso e o surgimento de estirpes com comportamento bioquímico atípico tem estimulado a procura por outros métodos de diagnóstico, como os testes moleculares. No presente estudo, analisou-se o método descrito na literatura envolvendo o gene rfbS em conjunto com testes desenvolvidos com base nos genes speC e speF. O gene rfbS codifica a enzima que atua na parede celular de bactérias e tem sido utilizado para diferenciar SP e SG. Com o propósito de padronizar a PCR para ser utilizada na rotina laboratorial, empregou-se dois métodos de extração de DNA e diferentes concentrações de reagentes. Entretanto, tendo em vista a dificuldade de padronização encontrada desde o início, foi concomitantemente desenvolvida a PCR para os genes speC e speF. Estes dois últimos estão relacionados com a produção da enzima ornitina-descarboxilase, a qual é ativa em SP e inativa em SG. Após testes inicias, foi possível padronizar a PCR do gene speC com posterior utilização da técnica de tratamento enzimático com a enzima de restrição Eco RI. Tanto para o gene rfbS quanto para os genes speC e speF, os resultados encontrados permitiram a diferenciação de 13 amostras de SP e 20 de SG isoladas no Brasil e duas cepas ATCC. Sendo assim, as ações sanitárias e a tomada de decisão no campo podem ser conduzidas de maneira rápida e segura. / Abstract: Salmonella Pullorum (SP) is very similar to Salmonella Gallinarum (SG). They are respectively agents of Pulorosis and Avian Typhoid, both diseases responsible for economic losses to poultry production. Due to the fact that both salmonellas have similar antigenic composition, their differentiation has been based in biochemical tests. However, this fact that this kind of procedure is very troublesome and the occurence of strains showing atypical behavior have stimulated the search for others diagnostic methods, such as molecular tests. In the present study, a method described in the literature using gene rfbS was analyzed together with tests developed based on genes speC and speF. Gene rfbS encodes an enzyme that acts on the cell wall of bacteria and has been used in the differentiation between SP and SG. In order to standardize the PCR procedure to be used in laboratory routine, two methods for DNA extraction were used, as well as different concentrations of the reagents. However, due to the difficulty in the standardization observed from the beginning of the study, a PCR procedure was developed for genes speC and speF. These genes are related to the production of the enzyme ornithine-descarboxilase, active in SP and inactive in SG. After the initial tests, PCR of gene speC was standardized, and later on it was coupled with the enzymatic treatment with restriction enzyme Eco RI. Results obtained for gene rfbS as well as for genes speC and speF enabled differentiation of 13 SP and 20 SG strains isolated in Brazil and two strains ATCC. So, the sanitary actions and the decisions in the livestock can to be conduced with safety and rapidity. / Doutor

So, it is not just about demographic data, which are thus presented with a matter of course, but also the economic a social data, where is this sort often neglected. The subject of the dissertation "Economic and social statistics by gender perspective" is to provide a coherent picture of the need for gender statistics, because the data in this area for men and women are often very different and it is necessary to identify possible causes and options for their removal.

Genomic and Peptidomic Characterization of the Developing Avian Brain

<p>Chicken and Japanese quail are commonly used models in developmental and sex specific neuroendocrine research. There is relatively little known about the mechanisms behind their sex specific brain development, especially regarding the impact of the sex chromosomes (male: ZZ, female ZW) in relation to gonadal hormones. This thesis explores several aspects of these processes. Gene expression analysis with cDNA and Affymetrix arrays on brain tissue from both pre-gonadal embryos and embryos with differentiated gonads indicate a strong sex chromosomal presence in sexual dimorphic somatic tissue development in both chicken and Japanese quail. This sex chromosome pattern seems to remain in adult brain tissue. The data demonstrates that chicken males exhibit a significant level of Z-gene dosage compared to females in both somatic and germ line derived embryonic tissues. Several avian sex determination gene candidates (MHM non-coding RNA, DMRT1, HINTW, and HINTZ) were analyzed by real-time PCR. DMRT1 is dosage compensated in male brain tissue, in contrast to its reported gene dosage in male gonads. Early embryonic ethinylestradiol (EE2) exposure did not affect male or female neural gene expression patterns during later development. A peptidomics analysis on quail embryonic day 12 (ed12) and ed17 diencephalon by LC-MS identified over 60 endogenous peptides and analyzed the expression patterns for 38 of them with regard to age, sex and early EE2 exposure. There was a general upregulation between ed12 and ed17, but no clear sex effects were detected. Multivariate analysis indicates that EE2 exposed individuals differ from control individuals in a gender independent manner, and that Gonadotropin-inhibiting hormone related peptide 2 (GnIH-RP2) is a candidate for EE2 induced peptidomic alterations in male embryonic brain.</p>

The aim of this thesis is to examine the practices directed to the flexibilisation of the labor brought together with the means of neoliberal structuring and the trade union crisis appeared as an outcome of these in terms of the Tekel Workers&rsquo / resistance in Ankara. The theoretical frame of this study is created in the content of the discussions that starting from the Marxist approach on trade unions and the critical view within this approach focus on the reasons of the crisis of the trade unions as a result of the applications of the neoliberal ideological structuring that appeared by the crisis of the accumulated capital during the mid-70&rsquo / s. Over this perspective, the analysis concentrates on the one hand employment models&rdquo / being multi-layered and insecure and on the other, the fragmentation of the class and therefore the representation crisis of the trade unions that are the outcomes of the commodification of the labor by the deregulation, privatization and flexibilisation policies applied in the process of neoliberal hegemony. In this context, the resistance of the Tekel workers that continued non-stop for 78 days is argued basing on the assumption that the process which forces the more flexible, insecure working conditions without any attachment to the trade unions via the application of the neoliberal political apparatuses becomes the common platform/destiny of all the parts of society constituted by different identity structures.

Grassroots Resistance Against Urban Renewal: The Case Of Guzeltepe, Istanbul

The aim of this study is to contribute to the urban social movement literature of Turkey which is lacking particularly for &ldquo / gecekondu resistance&rdquo / and to reveal and study the gecekondu resistance as a grassroots movement against the demolition of gecekondus, in the name of urban renewal projects in Istanbul, especially since 2004. It also investigates the &ldquo / urban social movement&rdquo / concept whose meaning is under discussion in the literature due to its usage in different aspects of resistance in the urban area. The literature review focuses mainly on the production of space, focusing particularly on urban renewal, urban resistance and social movements especially reflecting on the theoretical perspectives of prominent scholars like Lefebvre and Castells. The research focuses on neighborhood resistance in gecekondu areas / however in an attempt to make a categorization of ways of urban resistance in Istanbul. This thesis is based upon the field study pursued in the period between January and October 2006, in G&uuml / zeltepe neighborhood, in Ey&uuml / p, being a remarkable example of resistance for various reasons which will be elaborated in the thesis. G&uuml / zeltepe which is a part of the urban renewal project in Istanbul is investigated with participant observation and in-depth interviews comprising people both participating directly in the resistance and &ldquo / ordinary&rdquo / dwellers, who do not have political affiliations. The study is supported by a systematic analysis of representations of gecekondu resistance and its demolition in the Turkish press, from July 2005 until August 2006.
