This thesis deals with the principles of ultrasonic perfusion analysis and methods for determining perfusion parameters. It examines Evolutionary algorithms and their ability to optimize the approximation of dilution curves from ultrasond tissue scannig. It compares the optimization performance of three evolutionary algorithms. Continuous genetic algorithm GA, algorithm SOMA and PSO. Methods are evaluated on simulated and clinical data.

Vliv prosklení na vnitřní mikroklima objektů / The influence of microclimate on the inside glass objects

This master thesis deals with the influence of glazing on the microclimate inside buildings. In particular, I focused on the shading coefficient and the various factors that affect it. The thesis also concentrates on the origin and composition of the solar radiation, the formation of thermal stress and refraction. In the next part of the thesis, I focused on designing two variants of shielding factor and its impact on the thermal load of gym, sports hall and the related necessary changes. The last part of the thesis deals with experimental measurements of incident solar radiation passing through the window into real physical model. From the measured values, we determined realistic shading coefficient and then applied it in the previous section of this thesis.

Analýza účinku koncentrace slunečního záření na degradaci tepelných izolací ve skladbách plochých střech / Analysis of effect of solar radiation concentration on the thermal insulation degradation in the flat roof construction

The master´s thesis analyzes influence of direct and reflected solar radiation to heat strain and degradation of thermal insulation in the flat roof construction. In many building was a problem in a significant degradation of thermal insulation made from expanded polystyrene in the vicinity of glass surfaces. To determine the causes of this problem, it was necessary to study and test the properties of building materials and the physical nature of the effect of solar radiation on the building structure. In the introductory section lists the cases of buildings with degradation of thermal insulation and existing knowledge on this issue. The theoretical part describes the technical properties of thermal insulating materials used in the flat roof construction, nature and division of solar radiation, optical laws of light reflection and physical theory to explain the technical thermal behavior of building materials and structures. The last part describes a practical model and analyzing measured data in thermal stress in the flat roof construction. The practical part also determines the importance of different influences configuration structures using to dynamic simulations. At the end of the practical part they are validated theoretical knowledge obtained by analyzing data from a practical model.

A posteriorní odhady chyby pro řešení konvektivně-difusních úloh / A posteriori error estimates for numerical solution of convection-difusion problems

This thesis is concerned with several issues of a posteriori error estimates for linear problems. In its first part error estimates for the heat conduction equation discretized by the backward Euler method in time and discontinuous Galerkin method in space are derived. In the second part guaranteed and locally efficient error estimates involving algebraic error for Poisson equation discretized by the discontinuous Galerkin method are derived. The technique is based on the flux reconstruction where meshes with hanging nodes and variable polynomial degree are allowed. An adaptive strategy combining both adaptive mesh refinement and stopping criteria for iterative algebraic solvers is proposed. In the last part a numerical method for computing guaranteed lower and upper bounds of principal eigenvalues of symmetric linear elliptic differential operators is presented. 1

Diffuse large B-cell lymphoma (DLBCL) is the most common type of non-Hodgkin lymphoma. It is a molecular and prognostic heterogeneous disease. Three main genetic subtypes are called germinal center-like DLBCL (GC-like DLBCL), non-germinal center-like DLBCL (nonGC-like DLBCL) and primary mediastinal B-cell lymphoma (PMBL). These subtypes can be reliably distinguished only with usage of gene expression profiling (GEP). The GEP method can be applied only when fresh frozen tissue is available. The method is technically difficult and expensive. Thus, it is not used routinely. Since the DLBCL subtypes differ in prognosis, it is extremely important to be able to distinguish them. The presented thesis is focused on distinguishing of PMBL diagnosis in the group of DLBCL. Easily stored formalin-fixed, paraffin-embedded tissue (FFPE) and gene expression analysis using real-time quantitative polymerase chain reaction (RTqPCR) are used. In the first step, PMBL and DLBCL cases were distinguished with an internationally accepted clinical-pathological method. The agreement between clinical-pathological diagnosis and GEP is only 76%.
