This work deals with the current topic of carrier systems. Since the biggest problem is the passage of hydrophobic drugs through the bloodstream, or through universal body barriers (eg blood-brain), it is necessary to chemically modify these carriers in order to be able to administer hydrophobic substances effectively. Based on a literature search, several systems are designed and subsequently studied, in which there is a presumption of possible use for carrier systems and at the same time they have biosurfactants incorporated in them due to their ability to solubilize hydrophobic molecules. The theoretical part of this work will describe the individual biosurfactants, the process of their production, their physicochemical properties, and the possibility of their use in carrier systems. At the same time, the individual carrier systems, the procedure of their preparation, the possibilities of their use are described, and their advantages and disadvantages are also compared. In the practical part, the screening of both individual substances and their mutual interactions, as well as methods used to study the emerging structures is then performed. The study of molecular interactions is primarily performed using the technique of dynamic light scattering. The next part describes the optimization of hydrogel formation with incorporated biosurfactants in their structure and then the formed gels are subjected to rheological and solubilization tests. The study of the internal structure of these gels is performed using a scanning electron microscope.

Studium interakcí tenzidové složky přípravku Septonex s vybranými proteiny / Study of interactions of the surfactant component of Septonex with selected proteins

The study of protein and surfactant interactions is of great significance in a number of applications, such as the cosmetic, food or pharmaceutical industries and many others. However, they require further study due to their compositional complexity and the limitations of current analytical approaches. In this thesis, the cationic surfactant septonex in combination with two differently charged proteins lysozyme and bovine serum albumin under different physiological conditions (temperature, surfactant concentration, environment and others) was selected to study the interactions. Characterization of protein-surfactant interactions is a very important but challenging task, therefore it is essential to use appropriate approaches to explore the nature of these interactions. In order to unify the information to provide rational models, calorimetric methods (DSC, ITC) and dynamic light scattering were used. Isothermal titration calorimetry monitors the evidence for the formation of the system of the mentioned substances and information on aggregation behavior, differential scanning calorimetry characterizes the thermal stability of proteins and dynamic light scattering made it possible to monitor changes in particle size. Both proteins have been proven to interact with positive septonex, even if the lysozyme molecule is also positively charged. However, significant differences were found between the two proteins. From the obtained results it is evident that the identical charge of the protein with the surfactant has an effect on the intensity of the measurement, although all measured interactions showed an endothermic character.

This diploma thesis deals with the preparation and characterization of stealth liposomes and their combination with trimethylchitosan (TMC). This complex could find application in the field of inhalation administration. Stealth liposomes were prepared from neutral phophatidylcholine, negatively charged fosfatidic acid and polyethyleneglycol bounded to phosphatidylethanolamine. We have managed to prepare stealth liposomes with suitable properties that should guarantee passive targeting without evocation an immune response, despite the content of the negative component. We also found a suitable method of preparation for stealth liposome–TMC complex, where the change of size and zeta potential confirmed the non–covalent bound between two components despite the content of the polyethyleneglycol.

Generace kovových nanočástic v nízkoteplotním plazmatu v kapalině / Generation of metallic nanoparticles by non-thermal plasma in liquids

This thesis focuses on the process of nanoparticle generation using new source of nonthermal plasma combining corona and pin-hole discharge in liquids. The theoretical part is focused on generation of metallic nanoparticles using various types of plasma discharge, the properties of metallic nanoparticles, their preparation by other methods and methods of characterization of nanoparticles. The experimental part deals with the preparation of copper, silver and gold nanoparticles from solutions of their precursors. The influence of experimental conditions, such as the influence of voltage polarity, effect of precursor concentration, effect of added electrolyte or reducing agent were investigated. All samples were analyzed by UV-VIS spectroscopy. Dynamic light scattering was used to determine the sice of nanoparticles. To confirm the presence of nanoparticles, samples were analyzed using scanning microscope with and energy dispersion spectrometer for elemental analysis.

Vliv velikosti částic kosmetických pigmentů na vlastnosti finálního výrobku / Influence of particle size of cosmetic pigments on final properties product

This diploma thesis is focused on the study of the influence of the of size cosmetics pigments on the properties of the final product. This effect was studied on samples of make-up given by industry partner. Make-up samples were measured by the rheology method and the yield stress of all samples was determined. Scanning electron microscopy was used to determine the size and shape of the pigment particles. Particle size was also measured by dynamic light scattering, but the results were influenced by forming aggregates. To easier determination of the types of pigments in the sample was determined elemental composition of the pigments (Ti, Fe, O, Al) by EDS detector. Due to the unknown composition of make-up, the information was supplemented by X-ray powder diffraction (XRD) analysis, and it was possible to determine the compounds TiO2 (anatase and rutile), FeOOH and Fe2O3. The results obtained by these methods showed what pigments can be found in make-up samples, what their sizes and shapes are. It has been shown that the smaller the pigments are, the higher the viscosity and yield stress of the final product. The higher these quantities, the harder it is to handle the product.

Možnosti enkapsulace přírodních antioxidantů / Encapsulation of natural antioxidants

In theoretical part of this diploma thesis the basic properties of antioxidants, especially anthocyanins and phenolic compounds are described, as well as the basic features and principles of free radicals formation. The theoretical part also describes some possible ways of encapsulation of antioxidants into polysaccharide and lipid particles. In the experimental part basic characteristics of extracts from selected lyophilized fruits and vegetables (carrots, apples and mixed berries) are described. As group parameters of plant extracts the total antioxidant activity, content of flavonoids and phenolics, carotenoids, anthocyanins and ascorbic acid were determined. In experimental part also various encapsulation techniques were tested, encapsulation effectiveness of each technique was evaluated and the stability and size of the created particles were determined. As the best encapsulation method in terms of encapsulation efficiency in most of samples/parameters ethanol injection was found. On the other hand the particles prepared by ethanol injection are relatively unstable in terms of zeta potential, which is followed by their tendency to aggregate. As the most stable particles prepared by thin layer evaporation (TLE) and reverse phase thin layer evaporation (RP-TLE) were evaluated. Particles prepared by TLE, RP-TLE and chitosan-alginate particles exhibited a negative charge, while particles prepared by ethanol injection stayed uncharged and chitosan particles have a positive charge.

Interakce tenzidů se směsí hyaluronanů o různé molekulové hmotnosti / Interaction between surfactants and hyaluronan with different molecular weight.

The behavior of the system consisted by mixture of two different molecular weight hyaluronates and surfactant was investigated. Mixtures were 17 kDa hyaluronate with 1,46 MDa, 73 kDa with 1,46 MDa, 300 kDa with 1,46 MDa, 806 kDa with 1,46 MDa and 1800 kDa with 1,46 MDa. These compounds were always mixed in the weight ratios 70:30, 50:50 and 30:70. As the surfactant cetrimonium bromide and TWEEN 20 were used. Interactions were studied in aqueous solution with different ionic strength. Sudan red was used as hydrophobic dye. In all experimental series with cetrimonium bromide was observed phenomenon of discontinuous separated phases, described as pearls. Samples containing pearls were tested on stability, were dried and rehydrated back, as were also heated in solution. Subsequently, the particle size was measured in the remaining sample after pearls were filtrated. Mixtures of hyaluronate were characterized by measuring the viscosity using rheology microrheology. It was found that these compounds are heterogeneous and each sample point is not the same viscosity.

Dynamic Light Scattering for the Characterization of Polydisperse Fractal Systems by the Example of Pyrogenic Silica

Dynamic light scattering (DLS) is a method to size submicron particles by measuring their thermal motion (diffusion) in suspensions and emulsions. However, the validity of the Stokes-Einstein equation that relates the diffusion coefficient and the particle size is limited to spherical particles and very low concentrations. Within this thesis, DLS is used for the characterization of suspensions of pyrogenic silica which consists of fractal-like aggregates composed of sintered spherical primary particles. These structural features clearly complicate the understanding of DLS experiments and have been a severe obstacle to employing DLS as routine standard tool for the characterization of pyrogenic silica. The main objective of this thesis is therefore to evaluate the application of DLS in product development and quality assurance of pyrogenic silica industry, what essentially means to identify those structural properties of fractal aggregates which are measurable with DLS and to quantify the method’s sensitivity to changes in these properties. The investigations presented here are split up into four parts, simulations that establish a relation between structural and hydrodynamic properties, experiments validating the simulation results, the characterization of concentrated suspensions and the application-oriented analysis of DLS data for specific industrially relevant measurement tasks. / Die Dynamische Lichtstreuung (DLS) ist eine Messmethode zur Größenbestimmung submikroner Partikel. Dabei wird primär die stochastische Bewegung der Teilchen (Diffusion) in Suspensionen und Emulsionen bewertet. Die Stokes-Einstein Gleichung, die das Verhältnis zwischen gemessenem Diffusionskoeffizienten und Partikelgröße wiedergibt, ist jedoch nur für kugelförmige Teilchen, die in sehr niedriger Konzentration vorliegen, gültig. In der vorliegenden Arbeit wird die dynamische Lichtstreuung zur Charakterisierung von Suspensionen pyrogener Kieselsäure eingesetzt. Diese besteht aus fraktalen Aggregaten, die wiederum aus versinterten aber meist kugelförmigen Primärpartikeln zusammengesetzt sind. Diese strukturellen Eigenschaften erschweren die Anwendbarkeit der DLS bzw. die Interpretation der Messergebnisse und verhinderten bisher den Einsatz der DLS als Routinemethode zur Charakterisierung pyrogener Kieselsäuren. Das Hauptziel dieser Arbeit ist daher eine Bewertung der Möglichkeiten der DLS für die Produktentwicklung und Qualitätssicherung in der Herstellung pyrogener Kieselsäuren. Das bedeutet im Besonderen, dass sowohl die messbaren granulometrischen Eigenschaften als auch die Sensitivität der Methode bei Eigenschaftsänderungen ermittelt werden müssen. Die hier durchgeführten Arbeiten sind in vier Teile gegliedert: Simulationen, die eine Beziehung zwischen strukturellen und hydrodynamischen Eigenschaften herstellen, Experimente zur Validierung der Simulationsergebnisse, die Charakterisierung konzentrierter Suspensionen und die anwendungsorientierte Auswertung von DLS-Daten für spezifische industrierelevante Messaufgaben.

Stabilita systémů pro řízené uvolňování léčiv na bázi plastifikovaného škrobu / Stability of controlled drug release systems based on plasticized starch

The thesis is focused on the research of stability of controlled drug release systems based on a blend of plasticized starch/polycaprolactone (TPS/PCL) that served as a carrier. Antibiotic vancomycin was used as a model drug, and its release from TPS/PCL pellets into aqueous environment was followed by UV-spectroscopy and the obtained time dependences were treated by a simple kinetic model. Moreover, the simultaneous release of starch particles to the surrounding liquid phase was studied by static and dynamic light scattering as well as transmission electron microscopy (TEM) in order to obtain information on the stability of biodegradable matrix and on the structure of the products of the pellet decomposition on a nanoscale level.
