Hodnocení kvalitativních parametrů ejakulátu psů

The dog has been accompanying men for eons, according to some literary sources the dog is the very first domesticated animal. My study included 152 dogs of 43 breeds. n total we had 408 ejaculates. We evaluated the proces of collection. Each ejaculate was evaluated on content, sperm count and motility of semen and morphological examination of semen.The evaluation of content and quality of ejaculate brought following average results. Time of ejaculation was 413,16 +- 168,66 seconds; the volume of ejaculate was 8,49 +- 6,51 ml; motility of semen 71,2 +- 16,8 %. There was approximately 69,1 +- 15,7 % of morphologically normal semen in the evaluated ejaculates. The results show, that you can expect quality ejaculate from middle sized sporting dogs. The dogs should be fed with a complete food mixture. The dog should be used for stud from the age of 2 years and in ideal conditions should not mate more often than once a week.

The theoretical part of the thesis on "Evaluation of semen quality selected breeds of dogs", provides a brief summary of available information obtained from literature dealing with dogs reproduction, anatomy male genital tract of the dog and basic laboratory techniques of animal semen analyzes. Practical work was focused on evaluating quantitative and qualitative parameters of ejaculate different breeds of dogs from the kennel Bohemia Alké. Was investigated primarily affect the order of collection, ages, body frame, time of day and length of the sampling collection itself on ejaculate volume, activity concentration, total sperm count and percentage of occurrence patomorfologických defects. Subscriptions ejaculate, macroscopic and microscopic evaluation was conducted in field conditions in the home where the dogs lived. To evaluate the percentage of damaged sperm pathomorphological were made morphological coatings, which were transported to the laboratory department reproduction of farm animals and the Institute of Animal Breeding, Faculty of Agronomy, Mendel University in Brno.

Hánlová, P: Effect of chromium on semen quality and carbohydrate metabolism in bre-eding boars, Diploma Thesis MENDELU, Brno, 2015, s. 59 The aim of this thesis was to develop a literature search and experimentally de-termine the influence of mineral nutrition on boar semen quality. The thesis is divided into two parts. The first assesses the impact of the addition of chromium on semen qua-lity of breeding boars. The second part examines the influence of the addition of chro-mium on glucose and insulin levels in the blood of breeding boars. The experiment was conducted in Velké Meziříčí at the insemination center boars. The experiment consisted of 28 Duroc boars breed for 14 weeks. These boars were divided into 4 groups according to age balanced. In the control (n = 7) group (K), was chromium receiving 205 ug/kg/day. In the 1st group (n = 7) animals (CR1) was dose chromium even increased by 60.6 ug/kg compound feed for the second group (n = 7) boars (Cr2) of 121.2 ug/kg and 3. group (n = 7) boars (Cr3) was added chromi-um in an amount of 181.8 ug/kg compound feed. The addition of chromium to the semen quality of breeding boars too had a signifi-cant impact. During the first months of feeding chromium occurred in the control group to a decrease in ejaculate volume of 29 ml. In the group S3 during the experiment is bound to increase sperm concentration, but in the fourth taking under review decreased significantly compared to previous months. The difference between the third and fourth taking was 131.5 thousand/mm3. This difference is statistically insignificant, and could be caused by external influences or errors in nutrition. Sperm motility during certain months was below 70%. This ejaculate should not be used for insemination. In the second part of the experiment, the addition of the chromium significantly reduced glukose levels and insulin activity. The obtained results can not recommend feeding chromium breeding boars.

The aim of this thesis was to investigate the issues of ejaculate evaluation and analyse the quality of ejaculates via macroscopic and microscopic methods. From 1st January to 31st March 2015, 69 ejaculates were collected from 7 rams of the dual-purpose Zwartbles breed owned by Ing. Martin Hoška, PhD, 4 rams of the Suffolk breed and 1 ram of the Charollais breed from the School Agricultural Facility Žabčice of Mendel University in Brno. Macroscopic and microscopic analyses were performed immediately after sample collection, including the detection of the ejaculate volume as well as the sperm activity and concentration. For the evaluation of morphological parameters of the ram ejaculates, morphological smears were performed, which were transported to a laboratory of the Department of Animal Breeding, Faculty of Agronomy, Mendel University in Brno. Mainly changes in the sperm head, acrosome, midpiece and tail (torsion, DAG effect) were monitored. Further, the presence of immature and degenerated sperm was also monitored. The results show that the quality of ejaculate is influenced by the age of the rams and particularly by the frequency of the sampling and the size of the testicles whereas the difference between the breeds of distinct purpose was not significant.

Vliv parametrů ekvilibrace a mrazení ejakulátu býků na motilitu spermií po rozmrazení / Effect of equilibration and freezing parameters on bull sperm motility after thawing

The hypothesis assume the existence differences in bulls spermatozoa viability after thawing, characterized by higher values of motility and higher proportion of live spermatozoa, in case of prolonged equilibration and lower temperature freezing gradient. The aim of this thesis was to find the influence of different equilibration and freezing curve conditions to on spermatozoa viability after thawing. The theoretical part of the thesis summarizes available information about the bulls semen parameters, methods for its evaluation and especially the processing and long-term preservation. Within the case study part of the thesis, during the years 2013-2015 the semen of the pre-selected group of 5 breeding bulls was being collected at the Sires insemination center. The group was uniform as to age, breed and housing and treatment conditions. After initial assessment the collected semen was divided into parts, which were then processed in different wals. A part of straws was processed by standard protocol, the other part was processed using different length of equilibration and freezing curve. First part of the straws was equilibrated in a standard protocol for 120 minutes; subsequently half of the equilibrated straws was standardly frozen by the Direct Freezing method based on 3-phase freezing curve while the other half was frozen using the different 2-phase freezing curve. Second part of the straws was initially equilibrated for 240 minutes with the subsequent freezing, similarly as in the previous case, used 3-phase and 2-phase freezing curve. Frozen straws were then stored in liquid nitrogen container at -196 °C. Spermatozoa motility was evaluated using CASA. The semen was further evaluated on the proportion of live and dead spermatozoa, using coloration of Eosin and Nigrosine, and on the sensitivity of the spermatozoa membranes, using HOS test, which detects the reaction of spermatozoa tail membrane to hypoosmotic conditions. The obtained results indicated higher progressive spermatozoa motility (+5.57 %) and higher percentage of live spermatozoa (+4.47 %) following the application of the prolonged equilibration (240 min). When applying the length of equilibration 240 minutes, the values of total spermatozoa motility and HOS test were higher as well, though the differences were not statistically significant. When evaluate the effect of using different freezing curves on the parameters of the spermatozoa after thawing, higher values (average + 1.6 %) at almost all of these parameters using 3-phase freezing curve were found, the diferrences were not statistically significant. Statistically significant differences among the bulls (P < 0.01) were found in total as well as progressive spermatozoa motility, in the proportion of living spermatozoa and in value of the HOS test - the individuality among the bulls is therefore evident. While evaluating the characteristics of spermatozoa movement by CASA, statistically significant differences among the bulls were confirmed at VCL, VSL, VAP, LIN, STR, WOB (P < 0.01), also between the diferrent length of equilibration (P < 0.01), but were not statistically significant when using diferrent freezing curves.
