The aim of the thesis was to focus on the evaluation of ejaculate quality from rams obtained by various methods of sampling and description of ejaculate collection methods in male farm animals with a focus on small ruminants. The ejaculate collection method of rams in the zwartbles breed did not affect ejaculate parameters. A significant difference was found in the volume of ejaculate, and also in other indicators of ejaculate quality, for example in sperm activity, as well as in eddy motion, sperm concentration and percentage of morphologically normal spermatozoa. The most significant difference seems to be in sperm concentration; in other indicators, differences do not have such an impact on the production of insemination doses and practical utility in reproduction. The effect of the ejaculate collection method on rams was also evident in short-term preservation in commercial diluent and pasteurized milk.

The aim of this diploma thesis The effect of stable microclimate on qualitative and quantitative parameters of ejaculates of breeding bulls, where the effect of stable envi-ronment and age on the volume, concentration, activity and changes in morphology of sperm in ejaculates of bulls was evaluated. In the period from 1st March to 31st August 2014, there were done 154 collections of semen from nine bulls of Czech Pied cattle lairaged at the insemination center of bulls operated by a breeder's cooperative Impuls in Bohdalec. Immediately after the collections macroscopic and microscopic examinations have been performed for all samples, which included determining the volume, con-centration and activity of sperm. After evaluation of morphological parameters of bovi-ne sperm the percentage of morphologically normal sperm and pathomorphological da-maged sperm were evaluated. There were observed changes on the head, acrosome, connecting part and flagellum. Our results show that the biggest effect on the qualitative and quantitative parameters of semen for breeding bulls has age, the temperature along with moisture, where the greatest differences in results were found for the extreme va-lues. A value of temperature - humidity index (THI) has negligable effect, where there were the smallest differences in the obtained samples.

Evaluation of semen quality is an important element in the evaluation of stallion fertility. Quality ejaculate produced in sufficient quantities is a prerequisite for successful insemination in horse breeding. To assess the quality of ejaculate is used by many laboratory evaluation methods. Selected methods are also engaged in this thesis. The basic parameters considerating in the ejaculate in this thesis are the evaluation of activity, sperm concentration and morphological sperm analysis. For using ejaculate in producing insemination doses need to be assessed volume of ejaculate too. Minimum volume of the ejaculate of stallions should be 20 ml. The activity of fresh semen should be at least 60 %. Normal sperm concentration value of stallions ejaculate is considered at least 100 × 106 in 1 ml of semen. The minimum amount of morphologically normal stallions sperm is required above the threshold value of 60 %. In this thesis we studied a group of stallions which were stabled in Zemský hřebčinec Tlumačov, s.p. Stallions were included in breeding season for 2015. Evaluation of quantitative and qualitative parameters were performed od 47 samples of fresh semen, which were collected from 12 stallions during the breeding season. Semen collecting was from April to July 2015. Immediately after each semen collecting was measured volume of ejaculate and sperm activity was recorded. To further evaluation were counting sperm concentration and morphology of sperm evaluation. For mophological analysis were preparations stained by two methods: B. T. Farelly staining and method Byghošť. The analysis was determined by the percentage of morphological sperm defects, which were divided into the following categories: acrosome defects, sperm head defects, sperm mid-piece defects, sperm tail defects, abnormal forms of sperm and immature sperm. The research also used a theoretical comparison of the two methods of stainings due to occurence of morphological defects and estimate the correlation relationships between qualitative and quantitative parameters of the ejaculated os stallions.

This diploma thesis deals with the quality of ejaculate of shepherd dog breeds. In the first part we will classify shephed dogs within the FCI organization and we will approach the breeds that we assess in this work. We will describe the anatomical and physiological characteristics of the reproductive organ of male dog, its ejaculate, ejaculation, semen and their development. We will also learn about the possibilities of dog ejaculate collection. In the main section we will deal with the evaluation of the ejaculate and we will focus on the macroscopic and microscopic examination of fresh ejaculate. In the course of the work, 100 ejaculates from 28 dogs belonging to I. FCI group will be evaluated, each dog will be taken twice to four times and one shot will be made from each sampling. The dogs will be divided into groups according to age, breed, stabling and feeding, and the examined parameters of the quality of the ejaculate will be discussed and compared. In terms of the monitored indicators, a highly statistically significant effect (P is less than or equal 0.01) of the order of collection on the volume of ejaculate obtained will be demonstrated. A statistically significant difference (P is less than or equal 0.05) between the first and the second sampling will be found in the evaluation of the effect of the order of collection during one day. The second volume will be found to be lower than the first one. It will be also shown that this value is affected by the age of the dog. According to our research, breed type and age of the dog causes a statistically significant difference (P is less than or equal 0,05) for sperm activity. The order of collection and stabling have a lesser influence on this indicator and the feed represents a negligible factor. Concerning the sperm concentration in the ejaculate, we will demonstrate a statistically significant difference (P is less than or equal 0.01) between the age groups of the dogs and the type of their accommodation. We will find a statistically significant difference (P is less than or equal 0.05) in the order of sampling. The factors of breed type and feed will have minimal influence in this case. We will not prove a statistical difference in the influence of the studied factors on color and odor, and we will be able to assert that these factors have negligible influence on these values. As for the qualitative sperm indicators, we will focus on the percentage of altered heads, connecting parts, flagella, degenerate and immature sperm. Concerning the values, we will conclude that the breed type, the age and, in the case of immature sperm, the order of collection have the most important influence on these parameters.

Vliv selenu, zinku, vitamínu C a E na potlačení tepelného stresu u kanců

The aim of the study was influence of the complex of two elements (zinc, selenium) and two vitamins (vit C, vit E) on the organism of pigs under heat stress. Twenty boars of the Duroc breed were included in the experiment. The boars were approximately the same age and weight, housed individually. They were divided into 2 groups. The control group (n = 6) was fed only to a basic feed mixture containing 0.02 mg Se; 21.5 mg Zn; 9.9 mg of vitamin E and 16.0 mg of vitamin C. In addition, 0.5 mg of selenomethionine, 70 mg of alpha-tocopherol, 70 mg of zinc oxide and 350 mg of ascorbic acid were added to the diet (n = 6). The experiment took place from June to September at the insemination station in Velké Meziříčí. The control group showed a slight increase in ejaculate volume (by 19 %), which is common in the summer months. At the same time, the sperm concentration was unstable, it increased by 15 % in August, but fell below the baseline by 3.5 % in September. Motility was around 69 % during the experiment. Statistically significant was an increase (by 15 %) of morphologically abnormal sperm in the ejaculate (P < 0.05). The experimental group had higher ejaculate from the beginning. However, the volume curve was almost identical to the experimental group. The sperm concentration increased by 23 % in the trial group after adding antioxidants. During the experiment the concentration only slightly decreased. Motility in the experimental group averaged 70 % and did not change significantly during the experiment. Even in the control group there was a statistically significant increase in morphologically abnormal sperm (by 12.3 %), (P <0.05). From the results it can be concluded that the increased amount of selected antioxidants did not have a demonstrable effect on the improvement of the ejaculate quality, but it can be assumed that this quantity has ensured the stability of the seed parameters.

The aim of the thesis "Evaluation of qualitative parameters of rams ejaculates" was to compare techniques of ejaculate collection of rams of combined breed. The sampling technique for the artificial vagina was compared to that of the electro-ejaculator. The quantitative and qualitative indicators of ejaculates of rams were evaluated in the thesis. The parameters evaluated were: ejaculate volume, sperm movement, sperm initial aktivity, sperm concentration in ejaculate and sperm morphology. Samples were taken at four days. A total of 24 samples were obtained from 10 rams of the combined breed, of which 12 were sampled by artificial vagina sampling by stimulation with fixed sheep and 12 samples by electro-ejaculator. The electro-ejaculator collection technique was found to have lower sperm concentration (P≤0.01) and lower initial activity (P≤0.05) compared to the artificial vagina collection technique. The difference in ejaculate volume and sperm movement in the compared methods was not statistically demonstrated. No significant differences between sperm collection techniques in sperm abnormalities were observed or statistically demonstrated. The results of the diploma thesis showed that the better technique of ejaculate collection is taking on the artificial vagina, because it leads to higher initial activity of sperm (P≤0.05) and higher concentration of sperm (P≤0.01), which can be used mainly in the preparation of insemination doses.

Aspekty spermatologického vyšetření čerstvého ejakulátu beranů

The diploma thesis deals with the ram genital organs, hormonal regulation, sperm production and ejaculate. Practically were made preparations of rams before collecting semen, semen collection and ways of obtain semen. Further were evaluated quality native ram semen by macroscopically and microscopically methods. Evaluated material was obtained from zwartbles rams breed owned by Ing. Martin Hošek, Ph.D. All the ejaculate was immediately after collection examined macroscopically and microscopically. The volume of ejaculate, wave movement and activity of sperm were evaluated. Subsequently ejaculate transported was to the laboratory department of livestock reproduction of the Department of Animal Breeding Faculty of Agronomy, Mendel University in Brno. There was also assessed the concentration of sperm was made chill survival test when ejaculate was stored at 3 °C and the results were evaluated after 24 and 48 hours. There were also evaluated morphological coatings, which monitored defects of the head, acrosome defects, defects in the flagellum, immature sperm cells, sperm cells degeneration and normal sperm. The results show that the volume of the obtained sperm has an effect on genetic line, season, frequency of colection and age groups. For semen quality were proven decisive sampling sequence which had the effect of sperm concentration and survivability.

Topic of this work was evaluation of spermatozoa of shepherd dog breeds, where we took a closer look on evaluation of dog ejaculate quality from FCI I. group. In this work the basic methods of ejaculate evaluation were mentioned as well as special evaluation methods, with focus on durability tests for needs of fresh semen insemination with use of special solutions. For our purposes were chosen 7 dogs, 5 of them were Czechoslo-vakian shepherd of one owner and 2 dogs were German shepherd breed of the second owner. For the best elimination of environmental influences on results these dogs were masturbated in home environment. As solutions we used UHT milk, dry milk, Androhep, Androstar, stallion solution and distilled water. We used standard mathematical-statistic methods for data evaluation.
