Analýza podjednotkového složení a funkce mitochondriální FoF1 ATP syntázy u modelů deficience strukturních podjednotek / Structural composition and functional properties of mitochondrial FoF1 ATP synthase on models of specific subunits deficiencies

Efimova, Iuliia January 2018 (has links)
Mitochondrial ATP synthase represents the final complex of oxidative phosphorylation (OXPHOS) system located in the inner mitochondrial membrane. Its primary role is to utilize mitochondrial membrane potential (Δψm) generated by respiratory chain complexes to produce energy in the form of ATP. Mammalian ATP synthase comprises of 17 different subunits organized into membranous Fo and matrix-oriented F1 domains. Defects of complex V and their manifestation have been studied on mitochondrial, cellular, tissue and organism levels using different models, including human cell lines and cell lines derived from patient tissues. In many cases mitochondrial diseases display threshold behaviour, when genetic defect is phenotypically manifested only bellow certain threshold in particular enzyme complex activity and/or content. This work was aimed at elucidation of functional consequences of ATP synthase deficiency in HEK293 cell lines with suppressed gene expression of γ, δ or ε subunits of ATP synthase central stalk. We have analysed range of clones with respective subunits knockdown and found varying decrease in assembled ATP synthase content, which was mirrored by the decrease in individual ATP synthase subunits. The only exception was subunit Fo-c, whose levels remained unchanged or even increased. ATP...

Problematika energetické bilance a faktory, které ji ovlivňují / Energy balance and its influential factors

Staufčíková, Tereza January 2018 (has links)
The theoretical part of the thesis deals with the issue of energy balance. Principles of its regulation and factors that affect it through energy intake or energy output are described in the thesis. It also describes methods of measuring and calculating energy output for proper adjustment of energy needs and achieving energy balance. In addition, the thesis presents energy needs in in specific life stages and pathological conditions that may occur during prolonged disruption of the energy balance. Both physiological and pharmacological factors which may impact energy metabolism are mentioned. The main objective of the practical part was to analyze freely available articles on the Internet providing different ways of weight reduction. The articles were subjected to a critical assessment from the point of view of a nutritional therapist and compared with expert studies. The secondary objective was to evaluate the questionnaire which also dealt with weight reduction. The individual responses were focused on the respondents' personal experience with weight loss. The results showed that up to 90 % of respondents attempted to reduce weight and that freely available articles, which do not always provide relevant information, were often used for this purpose. Keywords: energy balance, basal energy...

Monitorování příjmu sacharidů ve výživě sportovců / Monitoring of carbohydrate income in sportsmen's diet

Pourová, Veronika January 2017 (has links)
The topic of my master thesis is monitoring of carbohydrate intake in sportsmen's diet. In the thesis, intake of carbohydrates as well as other macronutrients is monitored in order to evaluate, if athletes prefer other energy sources to carbohydrates and if their total energy intake is sufficient. If athletes prefer easily digestible carbohydrates or if they follow recommendation for healthy diet and increase their intake of whole foods, legumes, fruits and vegetables can be evaluated by monitoring fiber and sugar intake. All the monitored data are compared with non-athletes in order to see, in which field their diets vary. In the theoretical part, principles of sports nutrition are defined, basics of population recommendations are mentioned and methods of nutritional assessment and energy expenditure estimation are explained. In the practical part, five-day food-diaries of athletes and non-athletes are evaluated by NutriPro Expert software. Five-day physical activity diaries recorded during the same days are evaluated by using the Compendium of Physical Activities. All the studied values (energy intake, energy expenditure, carbohydrate, fat, protein, sugar, fiber and alcohol intake) were compared to recommended values for athletes and non- athletes and further compared among these two groups. The...

Stanovení klidového energetického výdeje u osob s obezitou / Assessment of the Resting Energy Expenditure in Obese Individuals

Sadílková, Aneta January 2017 (has links)
Introduction: Weight loss programs are based on inducing a negative energy balance by decreasing energy intake while increasing energy expenditure. Energy needs of obese patients are difficult to determine. The value of resting energy expenditure, as the largest component of the energy needs, is usually calculated to asses an adequate energy intake for obese patient. In clinical practice, predictive equations are most commonly used for this purpose, because of their simplicity, although many clinical studies have shown that in obese individuals they may generate errors large enough to impact the outcome of treatment. It leads to the prescription of an inadequate diet that does not meet the criterion of long-term sustainability. Objective: The aim of the thesis is to compare the values of resting energy expenditure (REE) measured by indirect calorimetry with values calculated with Harris-Benedict predictive equation in a selected group of obese individuals. Methods: The sample includes 38 subjects, 28 women and 10 men, patients of the General University Hospital in Prague. The average age is 48 ± 11,71 years, the mean BMI is 42,88 ± 9,09 kg/m2 . For each subject, resting energy expenditure was measured by indirect calorimetry and calculated by the Harris-Benedict predictive equation. Indirect...

Energy Performance Contracting / Energy Performance Contracting

Čtrnáctý, Jaromír January 2009 (has links)
Energy Performance Contracting is an energy saving method, when the supplier guarantees a certain minimal level of energy savings to the customer, as well as non-rising energy costs. The method was developed at the turn of 70's and 80's in the USA and has spread to dozens of countries around the world till today, although the rate of the EPC market development differs from country to country. The method is useful mostly for heating, electricity and gas cost savings by the end-users. In the Czech Republic, the method has been used since 1993. Until now, several dozens, or even hundreds of projects have been realized, mostly for public sector customers. This paper analyzes the current state of the EPC method usage around the world as well as in the Czech Republic, the key part comprises of the EPC principles description, based on data from real EPC contracts. The work is concluded by a SWOT analysis of the method and by a discussion of the following question: "Can EPC projects blockade the overall saving potential in some cases?" The key information source for most of the work were answers to questions sent to eight Czech ESCO companies representatives, as well as the outputs from guided interviews with four of them, which were taking place between January and May 2009.
