Use of pulsed-field gel electrophoresis to genotypically characterize salmonellae grouped by serotype

The prevention and control of salmonellae in commercial swine operations are becoming increasingly important. The current approach focuses on identifying sources and/or origins of salmonellae contamination before swine are processed for human consumption. The objective of the current study was to assess strain variability among salmonellae grouped by serotype and to determine common origins of contamination (farm or slaughter plant). Salmonellae were previously collected from swine at slaughter, serotyped by the National Veterinary Services Laboratory and stored at - 70??C. Pulsed-field gel electrophoresis (PFGE) was performed to genotypically characterize serotypic isolates using restriction endonuclease XbaI. Dendrogram comparisons were also used to assess genotypic similarity when multiple genotypes existed. This study found PFGE to be more discriminatory than serotyping indicating that multiple genotypic strains existed among selected serotypes. On the basis of PFGE results alone, origins of contamination could not be determined in this study. It is suggested by the author, that origins of contamination could be further defined pending future research, in which in-depth longitudinal studies are included. When used as an adjunct to conventional typing methods, PFGE may prove to be a substantial subtyping system in epidemiologic investigations to identify point-of-entry contaminants to the food chain.

Design of Safety Device of A Large Lithium Battery Cell

In recent years, LEV and hybrid has been gradually popularized due to the energy crisis and increasing environmental awareness. The prevalence of LEV is to use batteries to replace the power from the gasoline. Secondary lithium battery is a good option for LEV since it has the features of light weights, high power density, long life, low pollution, and works without memory effect. However, the safety is a concern for end-users with more and more recalls of lithium-ion batteries involving the explosions. The purpose of this research is to propose a systematic approach of battery safety device with an engineering design method. The analysis on the factors affecting the safety is prior to the objective of this study as to define the specification of the battery safety device. Then, substance-field analysis and standard is to improve the initial systematic model, and the solution is specified with the synthesis of morphological matrix. Under the safety test for batteries, the effectiveness and feasibility of protected device is eventually verified.

Rethinking Field Studies for Design : An Argument for Using Longer Field Studies within Design

<p>This case study aims at showing how longer field studies can influence the design process. The design case is a redesign of the GUI of software for cell phone network simulation. The study explores the potential of a long field study, presenting it in a way that makes it possible for the reader to follow every step of the process and thus evaluate the merits of this long fieldstudy, and of longer field studies, in general.</p><p>Data for the study was collected and an initial analysis performed during a four weeks field study. Participant observation was the primary method, but other methods were also used. After the field study was completed, further analysis led to the formation of a design rationale, sorted according to three identified usage groups and one general category.</p><p>This study argues that a long field study, with design measurements, informs the design significantly. Week one was needed for creating the necessary rapport with the users/informants, while new information decreased during week four. This confirms the argument of earlier studies that the time span of field studies for focused design cases may be shorter than for more wide-aiming social research. However, one main finding is that the most significant data for the design rationale was unevenly collected, mainly in weeks two and three. Thus, the study argues that design cases may benefit from longer fieldstudies than is the standard within the design community of today.</p>
