The aim of thesis is acquaint with possibilities of fixation biological tissues. Choose a suitable tissue and fix it with formaldehyde and another fixator. Last but not least make measurements which determine effects of formaldehyde on the relaxation time of biological tissues. Mice brains were chosen as a suitable for tissues fixation. Measuring effects of formaldehyde and another fixator to relaxation time carried out at the Institute of Scientific Instuments Academy of Sciences of the Czech Republic.

The diploma thesis theme, which I have chosen, Addiction to the nasal use of pervitin, is based on my long-term interest in this subject.One of the factors that may be important in the study of any drug problem is the ritual. Rituals here may appear on several levels - it may contribute to the formation and maintenance of a dependency, in the application itself (creation of "lines" on the mirror, the use of identity cards, sniffing with a rolled banknote, etc.) and can take various forms (e.g., group or individual) (Holzbachova, 1999). For my thesis, I have chosen the method of qualitative research. The basic approach of qualitative research includes the grounded theory. The aim of the thesis is to analyze and describe the different aspects of ritual behavior of nasal drug users. The research question was to discover, which aspects of ritual occur in nasal drug users. As it is described in literature, all drug users ritualize the drug application to some extent. This represents stylized and routine behavior that occurs during drug use. There are situations, like the procurement of drugs, methods of taking, activities following the use of drugs and the ways to prevent the negative effects of drugs (Grund, 1993). During an interview, two of the interviewees said the word "ritual". Cyril considers it a ritual and the activities precede sniffing. These are activities associated with the hunt for the drug, the journey to the drug and the nervousness "before trip", which according to him is stronger than the feeling of drug application. Dana describes the application procedure as a ritual. However, she describes as ritualistic her behavior during the purchase of a sufficient supply of cigarettes for intoxication or when applying the drug on the toilet seat, wiping the toilet boards before the actual preparation of the drug for application. Although the other two interviewees did not use the word ritual in their answers directly, some ritualistic elements signaled a routine behavior in conducting the drugs process by them as well. In general, we assume that the thesis can contribute to the discussion about the ritual aspects of drug users. This work brings in new information about the rituals of nasal drug users. We anticipate publication in a professional periodical.

Vliv fixačních činidel na kvalitu a kvantitu nukleových kyselin v archivovaných vzorcích tkání / The effect of fixatives on DNA quality and quantity in archival tissue specimen

Formaldehyde is widely used fixative. Its advantages are low cost, simplicity of use and good fixation traits, which are fast tissue penetration, good preservation of morphological structures and compatibility with downstream histological applications. Formaldehyde disadvantages are negative effects on nucleic acids. Formaldehyde solutions modify primary structure of deoxyribonucleic acid (DNA), fragment DNA and create protein- DNA covalent bonds that hinder DNA isolation procedures. Level of negative effects of formaldehyde is dependent on many factors. Effect of formaldehyde chemical composition (formaldehyde dilution, presence of buffer or formic acid) and effect of fixation length were studied in this work. On DNA extracted from fixed tissues, DNA quantity and level of DNA fragmentation were studied by quantitative polymerase chain reaction, fluorescence assay for DNA quantification and by on-chip electrophoresis on bioanalyzer Agilent 2100. Quality and quantity of acquired DNA were tested by DNA profile determination for identification purposes using STR (short tandem repeats) analysis. Results show that of all tested fixatives, buffered 4% formaldehyde is the most suited solution in regards of sufficient amount of DNA and sufficient DNA quality. This diploma thesis deals with common parasites of game animals, animals and human occurring in the territory of the Czech Republic. It focuses on their fixation and general preparation of such specimens for usage in teaching of Science. The first part is devoted to the phenomenon of parasitism, and the history and definition of this concept. Systematic hierarchy of various unrelated parasitic groups is clarified. The second part provides the list of parasites, from the evolutionary original to the highly derived groups. We give an overview of their morphology and anatomy with the emphasis on their life cycle. The detailed knowledge of life cycle is essential for obtaining specimens. In each group, we indicate methods of collection and fixation in detail. The co called smartmicroscope represent the main point of the third part of proposed diploma thesis. Text of the thesis is supplemented with pictures taken by smart microscope, as well as by common microscope. Species Lupinus polyphyllus is one of the most hazardous invasive species in the Czech Republic and its increasing occurence represents a threat of native biodiversity not only in the flora of conservation areas. Influence of management experiments on invaded communities was studied on permanent plots founded on three localities in the Šumava National Park. The role of nitrogen fixed in this invasion was explored by analyzing of soil samples. Influence of allelopathic inhibition of invasive species on some coexisting native species was tested in the laboratory experiment.

This diploma thesis is focusing on mortage loans in the teritory of Czech Republic. Thesis is divided into two parts. In the first oneit brings theoretical recourse od mortages market in Czech Republic, including history of these loans. It divides those loans by typology, and defines providers of these servicer. In the practical part, there is developer analysis of the interest rates of individual banks at mortage market in Czech Republic. Analysis is made at different parameters of mortage loans. For this interes rate, there i salso calculated monthly payment. By comparison of each offer there is a possibility to choose the best offer for every case. For every potencial consumer is possible to compare profitability of financing own housing by mortage or by housing by rent.

Diploma thesis with the theme "Mortgage market in Czech Republic" has its objective in characterizing and analyzing the development of mortgage market from its beginning to the present situation. There is described the history and common characteristic of mortgage banking in the first part. There are also stated all related legal standards. There is described further procedure and particular phases which has to be completed by each mortgage loan applicant in the next chapter. Next chapter is focused on description of mortgage loan drawdown and its repayment. In the end of this chapter there is comparison of interest rate development and the mortgage loan volume according to the fixture time. Concurrently there is analyzed influence of interest rate to the volume of mortgage loans. Last but on chapter analyzes all relevant cost related to a mortgage loan application. Everything coming out from the previous analysis is put into the context with the results of my survey. Based on that the conclusions about impact of costs on number of mortgage loan applicatons are made. Final chapter compares individual bank offers and analyzes all possible factors which impacts final results.
