A caracterização molecular de G. duodenalis revolucionou a compreensão da taxonomia, diversidade genética e epidemiologia da giardíase em seres humanos e animais. Em nosso estudo, realizamos a caracterização molecular de G. duodenalis em bezerros bubalinos do Estado de São Paulo, Brasil. Assim, foram colhidas 183 amostras fecais de animais da raça Murrah, com até seis meses de idade. Estas amostras foram examinadas por meio da reação em cadeia pela polimerase tipo para amplificação da subunidade menor do gene do RNA ribossômico, todas as amostras positivas por esse gene, foram caracterizadas para amplificação parcial dos genes beta-giardina, glutamato desidrogenase e triosefosfato isomerase. G. duodenalis foi verificada em 6,56% das amostras fecais e por meio da análise das sequências, verificou-se 100% de similaridade genética com "assemblage" E. Esta foi a primeira detecção de G. duodenalis "assemblage" E em bezerros bubalinos no Brasil. / Abstract: Giardia duodenalis is a flagellated protozoan that colonizes the intestinal tract of vertebrate hosts. A molecular characterization of G. duodenalis revolutionized an understanding of the taxonomy, genetic diversity and epidemiology of giardiasis in humans and animals. In our stud, we performed the molecularly characterization of Giardia duodenalis in buffalo calves from State of São Paulo, Brazil. Then, 183 fecal samples of Murrah buffaloes were collected up to six months of age. These samples were examined by nested polymerase chain reaction for parcial amplification of the small subunit of the ribosomal RNA gene. All G. duodenalis-positive samples were characterized by beta-giardin, glutamate dehydrogenase and triosephosphate isomerase genes. G. duodenalis was detected in 6,56% of the faecal samples, and sequence analysis showed 100% genetic similarity with assemblage E. This was the first detection of G. duodenalis assemblage E in buffalo calves in Brazil. / Doutor

The Phenotypic and Genetic Distribution of Threespine Stickleback that Inhabit the Willamette Basin, Oregon, USACurrey, Mark 17 October 2014 (has links)
A key to understanding the origin and maintenance of the diversity of life is to understand how phenotypic and genetic variation is partitioned within and among populations. I characterize the spatial partitioning of phenotypic and genetic variation in an old Willamette Basin freshwater distribution of threespine stickleback (Gasterosteus aculeatus) and compare these results to younger populations. Phenotypic variation was measured using 14 phenotypic traits, and genetic variation was assessed using RADseq and Stacks software to identify tens of thousands of single nucleotide polymorphisms. The major partitioning of phenotypic and genetic variation in Oregon is along a stereotypical transition from oceanic to freshwater that has been seen in younger systems. Phenotypic and genetic variation is significantly partitioned between basin populations, and the genetic variation is geographically structured. This work suggests that parallel divergence between oceanic and freshwater forms originated before the end of the last glacial maximum.
