The basal vessels of the brain, especially the circle of Willis and basilar artery are of particular interest and importance as they are the main and only source of metabolism to such vital structures as basal ganglia, hypothalamus, midbrain and pons. Many investigators have therefore contributed to the knowledge of the course, tributary area, variations, anomalies etc., from the anatomical, physiological and pathological aspects.

Developmental changes in the human growth hormone receptor and its signal transduction pathways

Although tissue human growth hormone receptors (GHRs) have been identified from as early as the first trimester of fetal life, little is known about their functional role. We recently reported that fetal hGHRs appear to be relatively immature or have adapted to the in utero environment [194]. To further characterize these age-related differences, we have examined certain aspects of the hGHR structure as well as its signalling pathways. Initial cDNA sequencing and immunoblotting analyses showed no difference in hGHR molecular size. We then used RT-PCR to investigate levels of an alternatively spliced mRNA for a truncated (1--279) hGHR (T) which acts as a dominant negative to the full-length (FL) receptor; however, fetal hepatocytes showed T/FL ratios similar to postnatal liver (0.07 +/- 0.02 vs. 0.09 +/- 0.01, M +/- SEM). Western blotting was used to assess developmental differences in the relative abundance of downstream signalling factors. Stimulatory Jak and Stat proteins were detected in all fetal hepatocytes and postnatal livers tested (13 weeks fetal age to 62 years). (Abstract shortened by UMI.)

Identifying protein interaction partners of the Pitx2c N-terminus during embryogenesis

The vertebrate body is patterned asymmetrically along the left-right axis. Left-right patterning is required to establish correct asymmetric organ formation and positioning, and ultimately it is essential for normal physiological functioning. Misregulation in this process can lead to severe physiological defects in multiple organs, including the heart and gut. Pitx2c is a paired-like homeodomain transcription factor and is crucial for translating left-right signals into asymmetric morphogenesis. It is asymmetrically expressed in the left lateral plate mesoderm and continues to be expressed on the left side of organ primordia that are destined to become asymmetrically formed or positioned organs. Previous work studying Pitx2c has demonstrated its evolutionarily conserved role in left-right patterning. While the homeodomain is important for Pitx2c's function, our lab identified a putative interaction domain in the N-terminus of Pitx2c that is required for its function in left-right patterning. To further characterize the role of Pitx2c, I used a yeast two-hybrid screen to identify candidate protein interaction partners of its N-terminus. Thirty-two candidates were identified from the screen. Candidates that were isolated at least twice were prioritized for further analysis. After eliminating non-coding clones, false positive interactions, and confirming interactions of the candidate putative interaction domains with the Pitx2c N-terminus in yeast, six candidates remained: Anti-silencing function 1 homolog B (S. cerevisiae; Asf1b), Eukaryotic translation initiation factor 3, subunit A (Eif3a), Eukaryotic translation initiation factor 3, subunit M (Eif3m), Niemann-Pick C type 2 (Npc2), Serine/arginine-rich splicing factor 2 (Srsf2), and Serine/arginine-rich splicing factor 15 (Srsf15). mRNA Expression patterns of these six Pitx2c candidate protein interaction partners was analyzed by whole mount in situ hybridization and compared to Pitx2c expression patterns. All candidates were expressed symmetrically in the embryo, including tissues where Pitx2c is asymmetrically expressed. In the chick embryo, Npc2 and Srsf2 were expressed in a tissue-specific manner from HH stage 10 to HH stage 26, while Eif3a and Eif3m were broadly expressed throughout the embryo from HH stage 10 and only became enriched in particular tissues at HH stage 24 to HH stage 26. All candidates were coexpressed with Pitx2c in the looped heart tube. Only Srsf2 was coexpressed with Pitx2c in the lateral plate mesoderm. The known functions of these candidates are consistent with the known role of Pitx2c in transcriptional regulation and suggests new roles for Pitx2c in mRNA processing and translational regulation during left-right patterning. / Le corps des vertébrés est structuré asymétriquement le long de l'axe gauche-droit. La latéralité est requise afin d'établir la formation des organes ainsi que leur positionnement qui, ultimement, sont essentielles pour un fonctionnement physiologique normal des vertébrés. Lors de ce processus, une mauvaise régulation peut causer des troubles physiologiques sévères dans plusieurs organes incluant le cœur et l'intestin.Le facteur de transcription homéodomaine Pitx2c joue un rôle essentiel lors de la traduction des signaux gauche-droit en morphogénèse asymétrique des organes. Pitx2c est exprimé de façon asymétrique dans le mésoderme de la plaque latérale gauche et continu d'être exprimé du côté gauche des futurs organes asymétriques. Quelques études ont démontré que Pitx2c a un rôle qui est conservé lors de l'évolution dans la latéralité. Trente-deux candidats ont été sélectionnés. De ce nombre, les faux positifs ainsi que les candidats qui représentent région la non-codante d'une protéine ont été supprimés ce qui a permis de réduire la liste à six candidats : Anti-silencing function 1 homolog B (S. cerevisiae; Asf1b), Eukaryotic translation initiation factor 3, subunit A (Eif3a), Eukaryotic translation initiation factor 3, subunit M (Eif3m), Niemann-Pick C type 2 (Npc2), Serine/arginine-rich splicing factor 2 (Srsf2), ainsi que Serine/arginine-rich splicing factor 15 (Srsf15).Les patrons d'expression des six protéines candidates potentielles d'interaction avec Pitx2c ont été examinés par hybridation in situ et ont été comparé avec le patron d'expression de Pitx2c. Chez tous les candidats, l'expression asymétrique dans les embryons de poulet a été observée incluant dans les tissus où Pitx2c est aussi exprimé asymétriquement. Npc2 et Srsf2 sont exprimés de façon spécifique dans certains tissus à partir du stade HH10 chez le poulet. Eif3a et Eif3m sont largement exprimés dans les embryons à partir du stade HH10 mais leur expression est enrichie dans certains tissus à partir du stade HH22. L'expression de tous les candidats coïncide avec celle de Pitx2c dans la courbure du tube cardiaque. Seulement l'expression de Srsf2 coïncide avec celle de Pitx2c dans le mésoderme de la plaque latérale gauche. Les fonctions des différents candidats suggèrent que ces partenaires potentiels d'interactions de la partie N-terminale de Pitx2c pourraient jouer un rôle important lors de la régulation de la transcription, la formation de l'ARN messager et la régulation de la traduction lors de la latéralité.

The influence of prenatal maternal stress on postnatal immunity in offspring

Introduction: Current epidemiological evidence suggests that asthma and allergic conditions have an etiological component rooted in prenatal development. One of the associated risk factors is the exposure of the conceptus to prenatal maternal stress (PNMS). Current information on the effects of PNMS on postnatal immunity is limited to animal studies with scattered methodologies and two human studies with methodological constraints. The present state of knowledge does not allow drawing definitive conclusions and new studies are needed. Objectives: The main goal of the study presented in this thesis is to analyze the influence of PNMS on the immune system, and to determine which factors from the stressor and the stressed subject are relevant for this phenomenon. Methods: A comprehensive review of the animal literature was conducted to determine which aspects of immunity are most affected by PNMS, which factors intervene in this relationship, and what are the potentially global consequences of PNMS-induced changes on postnatal immunity and health. The available studies were analyzed by the timing (by Window of Vulnerability), type and duration/frequency of the stressor, and by the species, sex and age at assessment of the offspring. These conclusions were used to construct the hypotheses to test in Project Ice Storm, a human study that has been assessing prospectively the consequences of PNMS exposure caused by a natural disaster. Immunity was tested in 37 adolescents (average age 13½ years) whose mothers were pregnant at the time of the storm, measuring their major leukocyte populations and cytokine production capacity using flow cytometry. Results: The review of the animal literature indicated that PNMS affects multiple aspects of postnatal immunity. The most replicated findings were changes in the production of immune-inflammatory mediators (cytokines), and alterations in cell populations. Globally, PNMS might induce the development of a T-helper 2 immune phenotype, which potentially explains the epidemiological link between asthma and PNMS. The timing of the exposure to PNMS was determinant for the effect of PNMS on T lymphocyte populations, and the sex of the offspring influenced the manifestation and direction of cytokine production changes. The human study confirmed that PNMS influences postnatal immunity, indicated that PNMS effects depend on the objective exposure to the stressor rather than on maternal distress, showed that there is an enhanced but not exclusive vulnerability of CD4+ cells to PNMS in early pregnancy, and showed that IL-1β, IL-6 and TNF-α were altered, with TNF-alpha showing different levels in males and females. Conclusion: PNMS exposure has long-term consequences on immunity in animals and humans. Its effects are pervasive and determined by factors from the stressor and the subject. Some of the alterations induced by PNMS could be linked to the development of immune-related disorders such as asthma. / Introduction : Les données épidémiologiques actuelles suggèrent que l'asthme et les allergies ont une composante étiologique liée au développement prénatal. Un des facteurs de risque associé à ce lien est l'exposition du fœtus au stress maternel prénatal (SMP). Les effets nocifs du SMP sur la vie postnatale ont été documentés aux niveaux anatomique, physiologique, et comportemental. Cependant, les preuves actuelles sur les effets du SMP sur l'immunité postnatale sont limitées à des études animales avec des méthodologies diverses ainsi qu'à deux études chez l'humain présentant des contraintes méthodologiques. Les connaissances actuelles ne permettent pas de tirer des conclusions définitives; de nouvelles études sont requises. Objectifs : Le but principal de l'étude présentée dans cette thèse est d'analyser l'influence du SMP sur le système immunitaire, et de déterminer quels éléments du facteur de stress et du sujet stressé sont impliqués dans ce phénomène. Cette connaissance permettrait d'améliorer la compréhension des mécanismes sous-jacents responsables de l'influence du SMP sur l'immunité postnatale. Méthode : Une revue de la littérature a été effectuée afin de déterminer quels éléments du système immunitaire sont davantage affectés par le SMP, quels facteurs interviennent dans cette relation, et quelles sont les conséquences potentielles des changements induits par le SMP sur l'immunité postnatale et la santé. Cela a été effectué en analysant les études disponibles en considérant le temps (par Période de vulnérabilité), le type et la durée/fréquence du facteur de stress, ainsi que l'espèce, le sexe et l'âge du rejeton à l'évaluation. Les conclusions tirées de cette revue ont été utilisées pour définir les hypothèses à tester dans le cadre d'une étude humaine qui investigue de façon prospective les conséquences de l'exposition au SMP dû à un désastre naturel : le Projet Verglas. L'immunité a été testée chez 37 adolescents de cette cohorte (âge moyen 13 ans ½) en mesurant leurs populations de leucocytes ainsi que la production de cytokine in vitro en utilisant la cytométrie en flux. Résultats : La revue de la littérature animale indique que le SMP affecte de multiples aspects de l'immunité postnatale. Les découvertes les plus répliquées sont un changement dans la production de cytokines et une altération dans les populations de cellules. Globalement, le SMP peut induire le développement d'un phénotype de réponse immunitaire de type 2, qui pourrait potentiellement expliquer le lien épidémiologique entre l'asthme et le SMP. La période d'exposition au SMP (début de la grossesse) était déterminante pour l'effet du SMP sur les populations de lymphocytes T, et le sexe du rejeton influençait la manifestation et la direction des changements dans la production de cytokines. L'étude humaine confirme que le SMP influence l'immunité postnatale, que les effets du SMP dépendent de l'exposition objective au facteur de stress plutôt que la détresse maternelle, qu'il existe une vulnérabilité accrue mais non exclusive des cellules CD4+ au SMP tôt dans la grossesse, et enfin que les cytokines IL-1β, IL-6, et TNF-α sont altérés. Les niveaux de TNF-alpha étant différents entre les garçons et les filles. Conclusion : L'exposition au SMP a des conséquences à long-terme sur l'immunité animale et humaine. Ses effets sont invasifs et sont déterminés par des caractéristiques du facteur de stress et du sujet. Des altérations induites par le SMP peuvent être liées au développement de troubles immunitaires tel que l'asthme.

Examination of the concurrent validity of the neurobehavioral assessment for preterm infants

The Neurobehavioral Assessment for Preterm Infants (NAPI) is an instrument that was developed to measure the progression of neurobehavioral development in preterm infants born from 32 weeks conceptional age to term. This study examined the concurrent validity of the NAPI against another neonatal criterion instrument, the Einstein Neonatal Neurobehavioral Assessment Scale (ENNAS), which measures similar constructs and has demonstrated excellent reliability and validity. The results indicated that there was good correlation between many similar items and clusters found on both the NAPI and ENNAS. There was also a good correlation between NAPI cluster scores and performance and the ENNAS total deviant score, and the NAPI was sensitive to differences between infants classified as normal or abnormal by the ENNAS. Clinicians and researchers require instruments with psychometrically sound properties so that accurate tests results are obtained and this study has contributed to strengthening the validity of the NAPI.

Talk or text to tell? How young people in Canada and South Africa would prefer to self-disclose their STI status to their romantic partners, friends, and parents

Text messaging or texting has become a popular way among emerging adults to maintain their interpersonal relationships, and has recently become regarded by health professionals as a tool for communicating with young people about sexual health. To determine whether university students can be invited to receive information, and disclose their newly realized STI status to their romantic partners, their friends, and their parents, to help curb the spread of HIV, Herpes, or Hepatitis, 303 students (183 from McGill University, and 120 from The University of Kwa-Zulu Natal) (M=21 years) indicated on a survey (a) how they would prefer to self-disclose their STI status to receive care and support from others: Talking face-to-face; a mobile phone call; an SMS text message; or other modes of computer-mediated communication. Students also indicated (b) how they would prefer to receive their STI test results from a doctor or nurse, and (c) receive new information about an STI relevant to their own health. Contrary to expectations from previous literature, almost all students indicated that they would prefer to self-disclose their STI status, as well as to receive results and new information by talking face-to-face with their partners, parents, and doctors, rather than through a text message, a mobile phone call, or any mode of computer-mediated communication. Male students were two times more likely than female students to prefer to self-disclose their STI status to their romantic partner by talking with them rather than phoning or texting them. McGill students were two times more likely to talk face-to-face with their parents about their STI status than were students at UKZN. Finally, two times the number of students at UKZN, and four times more female than male students at UKZN indicated that they would prefer to receive their STI test results from a health care professional by talking with them at a health clinic rather than communicating via a mobile phone call or a text message. Sex differences in testosterone-related patterns of cortical maturation across childhood and adolescence

Background: Neuroendocrine theories of hemispheric lateralization hold testosterone as the predominant factor mediating sex-specific patterns of cortical growth and the ensuing lateralization of cognitive functions. However, to date attention has been mostly focused on prenatal testosterone levels as opposed to peripubertal changes in testosterone. Yet puberty coincides with both a significant rise in testosterone levels and increased differences between sexes in specific cognitive skills. Currently, direct examinations of relationships between testosterone and cortical grey matter remain limited, particularly in child or adolescent subjects. Weaknesses of the literature include small sample sizes, cross-sectional designs, uncorrected multiple comparisons, and the examination of restricted age spans. As a result, findings have been conflicting and vary across studies. Thus, the exact nature of testosterone-related sex-specific trajectories in cortical growth, timing of these effects throughout adolescence and brain regions most sensitive to testosterone, remain unclear. Objective: Examine testosterone-related sex differences in cortical thickness in a large, representative, longitudinal sample of healthy children/adolescents. Methods: Data from the NIH MRI Study of Normal Brain Development (n=282 subjects, 4-22 yo, 469 magnetic resonance imaging (MRI) scans obtained longitudinally over 4 years) were analyzed using linear mixed effects models to examine the association between salivary testosterone levels and cortical thickness while controlling for age, sex, total brain volume, and collection time. Results: We found significant and complex 'age by testosterone' and 'sex by testosterone' interactions on cortical thickness of widespread regions of both hemispheres, with striking differences between males and females as well as between younger pre-pubertal and older post-pubertal subjects. When males and females were tested separately, more pronounced regional effects emerged. Males showed a negative association between cortical thickness and testosterone levels exclusively in the left hemisphere, specifically in the posterior cingulate cortex starting at age 14, progressing as children age to include the precuneus (ages 16-19), dorsolateral prefrontal cortex (ages 16-21) and anterior cingulate cortex (ages 20-22). Females showed fewer areas of testosterone effects, but the associations exclusively involved the right hemisphere, specifically a positive association between cortical thickness and testosterone levels in the right somatosensory cortex, which was only apparent in the 5-8 years old age epoch, and later reversed, to a negative association in the same area for subjects ages 20-22. Conclusions: Significant sex- and age-specific associations exist between testosterone and cortical thickness in the developing brain but appear more complex than previously reported. While sex-specific testosterone-related cortical maturation appears to affect widespread areas in both hemispheres, more pronounced regional effects were observed in this study in key cortical regions known to be implicated in visuospatial and sensorimotor skills as well as cognitive control and executive functions. Females also show early positive associations between testosterone and cortical thickness consistent with known protective effects of androgens on neurons. In addition, the male-specific association between testosterone and cortical thickness in the left hemisphere and the female-specific association with the right hemisphere support the view that sex differences in hemispheric lateralization progress in a dynamic fashion throughout puberty. Whether the observed testosterone-cortical thickness associations underpin age- and sex-related differentiation in behavior, psychopathology, or specific cognitive abilities during childhood and adolescence requires additional study. / Contexte:Les théories de latéralisation hémisphérique postulent que la testostérone serait le facteur principal à l'origine des différences de développement cortical entre les sexes. Conclusions: Il y a des interactions significatives entre l'âge et la testostérone et le sexe et la testostérone affectant l'épaisseur corticale. Bien que le développement cortical spécifique à chaque sexe semble affecter des régions étendues dans les deux hémisphères, des effets localisés plus prononcés ont été observés dans cette étude dans des régions corticales connues pour être associées au contrôle cognitif et aux habiletés visuo-spatiales et sensorimotrices. Les femmes ont aussi démontré des associations positives avec le cortex somatosensitif qui sont consistantes avec les effets neuroprotecteurs des androgènes précédemment rapportés dans la littérature. De plus, la relation spécifique entre la testostérone et l'épaisseur corticale de l'hémisphère gauche chez les hommes et l'hémisphère droit chez les femmes supportent le concept de latéralisation hémisphérique comme un processus dynamique qui évolue pendant la puberté. D'autres études seront nécessaires pour confirmer si les associations observées entre la testostérone et l'épaisseur corticale sont vraiment reliés à une différentiation entre les sexes en termes de comportementset d'habiletés cognitives spécifiques au cours de la puberté.

Effects of maternal diabetes on fetal development in rats

The mechanisms underlying the high incidence of fetal abnormalities including fetal lung immaturity during maternal diabetes are not fully understood. Utilizing streptozotocin-diabetic rats as the model, I have examined the role of fetal hyperglycemia and hyperinsulinemia and other factors on fetal adrenal and lung functions in culture. Insulin and glucose did not alter fetal adrenal and lung cell proliferation and adrenal corticosterone output. On the other hand, a novel protein-bound, low molecular weight non-proteinaceous cytotoxic factor was detected in the serum of diabetic animals. In addition, a novel protein with cytostatic activity was found in fetal lungs, the concentration of which increased during diabetes. Partial amino acid sequence and Western Blot analysis revealed this protein to be similar to histone H2B. An extra-nuclear role is suggested for this protein because it appears to be present in the microsomal fraction of fetal lungs. It is concluded that fetal lung immaturity during diabetes may be contributed by cytotoxic and cytostatic factors contained in the serum and fetal lungs, respectively.

Reflex modulations with sensori-motor context could rely on premotor gain modulation: Implications for the VOR system

Gain modulation is believed to be a common integration mechanism employed by neurons to combine information from various sources and modalities. Gain fields have been shown to widely exist in cortical and subcortical areas of the brain and are believed to be the basis for fundamental brain functions such as coordinate transformation. Nevertheless the existence of gain fields has not been explored in lower brain areas like the brainstem. In this thesis, a physiologically relevant control system model is developed for both the slow and quick phases of the angular horizontal vestibulo-ocular reflex (VOR). This model is then used as a framework to show that if nonlinear gain fields are embedded in the response of VOR premotor neurons (in contrast to generally assumed linear neurons) viewing-context-dependent behavior of the VOR results automatically. This shows that, in principle, reflex modulations with sensori-motor context could be the outcome of local premotor nonlinearities, and not complex cortical computations. Interestingly, similar nonlinearities have been previously discovered in the spinal circuits. Simulations of this hybrid nonlinear model predicts that disconjugate eye movements during the VOR would be an inevitable consequence of the existence of such gain fields in the central bilateral VOR pathway. Behavioral data collected from normal human subjects during the angular VOR in darkness confirm the presence of large disconjugate components which are fully compatible with the model predictions. These theoretical and experimental results have implications for the design of clinical protocols to test reflexes. / La modulation du gain est consideree comme un mecanisme general d'integration dans le cerveau, employee par les neurones pour combiner des informations provenant de diverses sources et modalites. Ces characteristiques de champs de gain se sont montrees souvent, repandues dans le cortex et sous-regions du cerveau. Elles sont soupconnees detre a la base de fonctions fondamentales comme la transformation de coordonnees spatiales. Neanmoins, l'existence de champs de gain n'a pas ete exploree dans les regions inferieures du cerveau, comme le tronc cerebral.Dans cette these, un modele du systeme de controle des yeux est developpe a la fois pour les phases lentes et rapides du reflexe vestibulo-oculaire (VOR) angulaire, tout en soutenant la compatibilite avec les donnees physiologiques pertinentes. Ce modele est ensuite utilise pour demontrer que si de tels champs de gain nonlineaires sont ancres dans le circuit du VOR sur les neurones premoteures (contrairement aux presumes neurones lineaires), le comportement et les aspects dynamiques du VOR dependraientt automatiquement du contexte binoculaire sans recours a d'autres calculs exterieurs. Ceci demontre que, en principe, les modulations du VOR et de plusieurs autres reflexes avec le contexte sensori-moteur pourrait etre le resultat de nonlinearites locales. Notons que des nonlinearites semblables ont deja ete decouvertes dans les circuits de la moelle epiniere. Des simulations de ce modele hybride non-lineaire predisent que les mouvements oculaires seraient disjoints pendant le VOR, en consequence inevitable de l'existence de tels champs de gain dans les centres premoteurs bilateraux. Les donnees experimentales recueillies ici chez l'homme et le singe soutiennent ces predictions: pendant le VOR angulaire dans l'obscurite, nous avons observe une importante reponse en vergence dans les movements binoculaires que ce soit en phase lentes ou rapides.
