Lectina da esponja marinha Tedania ignis: purifica??o, caracteriza??o e intera??o com leishmanias

The purification procedure was carried out by crude extract precipitation with ammonium sulfate 30% (FI). The precipitated was resuspended in the same buffer and fractionated with acetone 1.0 volume (F1.0). A lectin was purified from this specific fraction by using an affinity chromatography Sepharose 6B. This lectin preferentially agglutinated human erythrocytes from B type previously treated with papain enzyme. The hemagglutinating activity lectin was dependent of divalent Mn2+ cation and was inhibited by the carbohydrates galactose, xylose and fructose. SDS-PAGE analysis indicated a molecular mass of the lectin around 45 kDa. This protein showed stability until 40?C for 1 h. Further, it showed activity between pH 2.5 and 11.5, with an enhanced activity at pH 7.5. Leishmania chagasi promastigotes stained with Coomassie brilliant blue R-250 were agglutinated by F1,0 and in the presence of galactose this interaction was abolished. These results show that this lectin could be implicated in defense procedures and it will can be used as biological tools in studies with this protozoon / Uma lectina foi purificada e caracterizada da esponja marinha Tedania ignis. O procedimento de purifica??o foi conduzido a partir do extrato bruto obtido por solubiliza??o em tamp?o B?rax 50mM, pH 7,5 e precipita??o prot?ica com 30% de sulfato de am?nio (FI). O precipitado foi dissolvido no tamp?o de extra??o e fracionado com 1,0 volume de acetona (F1,0). A lectina foi purificada desta fra??o por meio de cromatografia de afinidade em Sepharose 6B. A prote?na apresentou uma massa molecular de 45 kDa, conforme determinada por eletroforese em poliacrilamida em SDS, e se mostrou est?vel at? a temperatura de 40?C, por 1 hora. Uma curva de pH foi conduzida entre os valores de pH 2,5 a 11,5, onde observou-se uma maior atividade em pH 7,5. Dentre os eritr?citos humanos testados ela aglutinou preferencialmente os do tipo B tratados com papa?na. A atividade hemaglutinante da lectina foi dependente do c?tion bivalente Mn+2 e foi inibida pelos carboidratos galactose, xilose e frutose. Formas promastigotas de Leishmania chagasi coradas com Coomassie blue R-250 foram aglutinadas por F1,0 e na presen?a de galactose essa intera??o n?o foi observada. Rhythmic structure in the music of Jean Guillou: "Agni-Ignis" from Hyperion (1988).

In 1988, Jean Guillou composed Hyperion on a commission from the French oil company, Société Nationale Elf Aquitaine, to celebrate its discovery of oil. He developed this theme of fire using his imagination, European philosophical concepts, and various myths. As with Messiaen, rhythm becomes a significant element in Guillou's organ music to convey a heightened dramatic story of an exaltation of fire and to represent the fourth movement of "Agni-Ignis." For aspects of rhythmic structure, I developed new methodology to analyze rhythm in six sections of "Agni-Ignis." Guillou uses experimental rhythmic techniques such as rhythmic subdivision, cycling, rhythmic ostinato, durational contrast, and rhythmic crescendo to build the musical structure of the piece. Among them, the primary subdivision of 16th-note groups organizes throughout the piece as a cyclic theme to convey the powerful and vivid mood of fire. This rhythmic group creates many pitch patterns as thematic transformation to provide both rhythmic and harmonic complexities. The two types of rhythmic ostinato, which is variable and invariable type, juxtapose below the manual's skillful rhythm to provide variety and unity. The other notable features of rhythm appear at the border of each section, such as rhythmic crescendo, durational contrast, 32nd-note groupings, rest and fermata to build tension and relaxation. The rhetorical figure of pitch D, which is another "fire theme" in the pitch aspect dominates the core section which has a much slower rhythm with sustained notes. In general, Guillou has been influenced by his predecessors such as his teacher, Messiaen, and Stravinsky. However, he is uniquely suited to explore the limitless possibilities of the organ in a more secular and avant-garde style. The purpose of this study is to give the performer new insight to guide his or her performance.

The area is located in the vicinity of the Brno reservoir, specifically from the shipping dock to the bay Rakovec. The project addresses the links between these two points by pedestrian, bicycle and wheel-promenade, which runs both on shore and on the water. The first part is the area of pedestrian zone with buildings in the form of waves, which offers various services such as restaurant equipment, information center and a music bar and more others. Buildings protrude from the terrain field and they are designed as a reinforced concrete structure with steel beams and their roofs are walkable. In addition, walking and biking trails continue along the shore and they are separated by terrain elevation. The complex of cultural, social and sports center is located in the ness of existing beach Kozí Horka. There is a proposed cultural center facilities, stage and auditorium, café & snack bar, showers and public toilets, first aid, bike rental and sports facilities. These functions are grouped into four objects solved as a timber (light wood frame, sandwich external walls with ventilated facade). The buildings are single-storey construction with a height of 4000 mm and wheelchair accessible. Stage and auditorium is solved afloat on circular piers mobile on rails. These piers conclude two buildings into the arc from the shore; their mass is hidden behind the structure of steel beams covered by wooden logs (driftwood) acting as waterfowl nest in the view from the shores. Pedestrian and bicycle promenade continues along the shore and is connected by the bridge to the yacht club.

Cultural and Public Center is located on the dam in Brno dock, addressing the promenade between the marina and Kozí Horka, starting points and bicycle path , and include marine terminal and a natural amphitheater with a restaurant. Promenade is the multilevel net of wheelchair interconnected trails and green spaces with objects of public amenities covered by reinforced concrete shell structure with walkable green roof and with color features as landmarks. The building of the cultural center is fitted in the field wave that rises and opens towards the audience. The back part of the building is composed by field wall with employees' inputs, which forms a barrier between the amphitheater and the bike path behind the building. The object reacts to different heights of promenade's trails by Raumplan arrangement. It is divided into performers' background with store of properties, public toilets and restaurant facilities. The supporting structure is a monolithic reinforced concrete wall system in module 4 m, which carry exposed reinforced concrete shell structure with walkable green roof. The main walls are rotated by 5 °. Overall height ranges from 2700 mm to 4750 mm and headroom ranges from 2300 mm to 3800 mm according to the curved shell structure. Nationwide building foundation by the base plate with circumferential bands into the frost resistant depth is affected by bottom water pressure.
