This thesis deals with possibilities of isolation DNA yeasts genus Brettanomyces from wine with following identification using PCR. Fourteen wine samples with assumption of occurrence Brettanomyces were analysed. The literary section describes characteristic of yeasts Brettanomyces and their impact on wine. Further is described principle of PCR and use of this method in detection of Brettanomyces. Four different methods of isolation DNA directly from wine were tested. Wine samples were analysed using of five different primers by PCR. In this work were tested designed probe and primers for identification of Brettanomyces by real-time PCR.

This diploma thesis deals with problematics of noisiness and its importance for human health. The theoretical part examines the basic types of materials used in the production of door panels and their sound isolation properties. Another theoretical part is dedicated to the determination of weighted sound insulation of commonly used door panels by means of calculations. The practical part consists in the design of self-made panels usable in the production of entrance doors. The aim of this thesis is to increase the sound isolation of the door fillings while the thermal isolation properties and the aesthetic value of the door are maintained. The production and subsequent detection of acoustic properties of the designed door filling will be the result of the practical part. The thesis also includes economic evaluation and comparison with fillings used in door production.

The diploma thesis is divided into two parts. The first part, which is rather theoretical, is engaged in the construction of the house based on wood meets the requirements of low-energy building. Furthermore, this section gives the most widely used thermal insulation and its comparison. In the second part, which is rather practical, is the proposal a two-generation family wooden-house, which is situated in the village Dolany.

The thesis deals with the evaluation of thermal properties of buildings and their possible improvement. The work includes a summary of the different types of materials used in construction and insulation to minimize heat losses through transmission. Work involves the calculation of heat loss of the house, alternative solutions to improve its thermal properties and compared concept solutions with modern construction. At the end of the work.is peformed comparison of the existing building with the proposed alternatives for improvement.

This thesis deals with the problems of insulating older buildings. The goal of this work is to evaluate the conditions of interior environement of stables for breeding hens and to propose an improvement of its thermal properties. The first chapter describes findings from literature about individual methods of insulating buildings. The second chapter describes the basic methodology and the principle of used measurements of the technical properties of the stable for breeding hens. In the third chapter there are evaluations of the measurements and necessary calculations for the improvement of thermal properties of the building. The last chapter lists the economic evaluation of the proposed insulation method of the building in respect to the egg production and feed consumption.

This thesis describes the design implementation loft above the selected object. The work consists of text and drawings. The text of the first development of construction and technical survey of the current structure and truss ceiling. Furthermore, in the attic stairs and drafted alternative proposals tracks curtain wall, partition wall, ceiling and floor structures. For All proposed structures is then given a list of works with bill of quantities. In the second part of the thesis is prepared drawings of existing and newly designed truss structure under which it is possible to realize the attic.
