This thesis is focused on a design of a freestanding family house made of solid wooden panels. It includes two different designes for the construction of the house. One construction uses commonly materials and the other features a maximum use of wood and wooden products. For both designs, the price calculation is based on the URS price indicators and both designs have been processed truth the Passive House Planning Package (PHPP). The designs both consists out of 2 parts, namely text and drawings.

Thesis is focused on thermal-insulation properties of wood fibreboards and building structure model including non-ventilated air layer. In the first part, the influence of temperature and moisture content on heat conductivity coefficient and heat transfer coefficient of wood fibreboards was investigated. In the second part, the building structure model was prepared, consisting of CLT, air layer and wood fibreboard. Subsequently, the two above mentioned characteristics were determined in dependence on measuring temperature, air layer thickness and inner surface emissivity.

It is possible to analyze cell structures, detect accumulation of certain metabolites, ions, enzymes and other substances thanks to the isolation. The goal of this diploma thesis was to provide an overview of isolation methods used for the isolation of cell wall, protoplasts, chloroplasts and vacuoles of plant cells. Isolation processes used for individual types of cell structures, the pros and cons of the various isolation methods, components of used media and their functions, as well as the structure and function of individual plant structures were described.

Analýza obsahových látek rodu Psilocybe I. / Analysis of Psilocybe constituents I.

The goal of this work was to determine the contents of two main alkaloid types of Psilocybe species occurring in the Czech Republic. Correct identification of species was checked by mycology specialist. Qualitative and quantitative determination of substances contained in three collected species of the genus Psilocybe was carried out by LC-MSn (LIT). Samples were mainly collected in the north and northeast Bohemia. Three species of Psilocybe were found. The most found species were P. serbica var. bohemica then P. semilanceata and only one deposit of P. serbica var. arcana. Total of 35 deposits within eighteen locations. From qualitative point of view, the two main alkaloids were confirmed in all samples. Quantitative results for content of psilocin and psilocybin (in this order) are: P. bohemica 0,005 - 0,152 %, 0,000 - 0,048 %; P. semilanceata 0,087 - 0,337 %, 0,000 - 0,005 %; P. arcana 0,018 - 0,031 %, 0,003 - 0,022 %. Results in all locations are similar and rather dependent on the fungi type. Our results are based on dry mass. This work brought fundamental knowledge for future optimisation of analytical methods and assumptions for the project extension of other locations and determination of content of minority substances alongside the content of majority substances.

This thesis focuses on the design house using wood for house construction and straw as its thermal insulation. In the text section describes the properties of straw bales and the principles for proper installation of the structure. The following describes the variations of external walls using straw. Each option contains the track walls with the materials used and is assessed in terms of the heat transfer coefficient and condensation. Alternative options of exterior walls are then compared on the basis of selected parameters. House is designed to my liking . Drawing section contains drawings, the basic design details. In accordance with natural stavitelstvím is based on the above ground ventilated gap. The roof structure is designed from the trusses to be used straw bales for insulation and roof deck. Trusses are statically evaluated by the EC 5.

Study on the chromosome number in the alveolate alga \kur{Chromera velia} by TSA-FISH

This thesis is focused on the chromosome-number studies in alveolate alga C. velia. This Apicomplexa-related photosynthetic organism proved to be a unique instrument for the study of Apicomplexa and their unique organelle apicoplast. To better understand the similarities and differences within these two phyla, a further chromosome analysis was needed. The first part of this thesis sums up the present knowledge about ploidy, life cycle and genome organization within C. velia and the close-relative phyla of Apicomplexa and Dinoflagellata. The second part describes our attempt to examine the ploidy and total number of chromosomes in C. velia using fluorescence in situ hybridization. The ploidy of C. velia was successfully determined and we also made a significant progress in the determination of the total chromosome number. (max. 4000 znaků)

This thesis deals with description and verification of different methods of DNA isolation from insect tissue of order Lepidoptera -- butterflies with an emphasis on family Sphingidae -- hawk moths and subsequent amplification of COI gene by PCR method. All tested methods were compared and evaluated in terms of efficiency, yield and purity of the DNA isolation, duration and financial costs. We selected to establish a methodology which is sufficient for obtaining COI gene sequences using genetic analyzer, its application in the system BoldSystems and subsequent determination of the sample. This is important in effort to map the species diversity.

Diploma thesis is dealing with the proposal of family house with basement at a part of the house at specified land in Žižkovo Pole. In the theoretical part will be the list of used literature for the thesis. Eventually, the variants of disposition solutions will be created and technical documentation will be done for chosen variant. A part of work will be a suggestion of constructions compositions with clarifying coefficient of thermal transmittance, static evaluation of chosen wooden particles of roof structure and valuation of building facility according to JKSO indicators. The work will be supplemented with visualization of the object. In the proposal, the requirements for habitation will be respected, according to valid legislation of Czech Republic.

Key words: Liriodendron tulipifera, alkaloids, biological activity. The alkaloids from the wood of the species Liriodendron tulipifera L. were isolated and their inhibitory activity against acetylcholinesterase, butyrylcholinesterase and propyl oligopeptidase, which are enzymes involved in the pathophysiology of Alzheimer's disease (AD), was investigated. The search and testing of new substances, which are used in AD treatment, is very relevant, as this disease cannot be casually treated yet. An alkaloid extract of Liriodendron tulipifera L. wood was tested in a preliminary testing for inhibitory activity against human cholinesterase. Because of the promising results, it was chosen for an isolation and identification of possible effective alkaloids. The extract was carried out by column chromatography with a step gradient elution. A preparative TLC was used to isolate alkaloids. The identification of alkaloids was done by structural analyses (NMR and MS). Optically active substances were measured for their optical rotation.
