This master thesis entitled "Development of coloraturas arias " is divided into several parts, which branch out into individual sections. The first part deals with an explanation of basic terms of coloratura and aria, as they are described in encyclopedic dictionaries and encyclopedias. This part is followed by brief formal development of aria throughout ages. I continue with the historical development of coloratura from the chapter Middle Age through the following era of renaissance and polyphony to the baroque, where I am describing the origins of opera as a dramatic genre. This part is divided into two parts , in which I describe the formation of coloratura aria and recitative and also the birth of other genres in which are coloratura arias widely used. The next chapter is devoted to the phenomenon of the eunuchs, who contributed to the expansion of coloratura arias around the world. The chapters Alessandro Scarlatti, Georg Friedrich Handel and Johann Sebastian Bach, mention their compositional techniques and also examples of work with coloratura and their contribution to the future generations. The following Classicism brings compositional techniques of Wolfgang Amadeus Mozart and his work. The next chapter, the romantic period, is dedicated to "Belcanto composers" Rossini, Bellini and Donizetti, who had contributed to the trend of coloratura techniques. Giuseppe Verdi was the last representative of belcanto era. In the penultimate chapter I write about selected roles that might be suitable for coloratura soprano and in the end I describe my relationship with the soprano voice in coloratura field.

This master thesis is focused on analyzing the development of cello concerts written by Russian composer, respected cellist and teacher Karl Julievich Davydov. In the first place, the analysis is about the upward development of the genre – from virtuosic type of concerts to virtuosic-symphonic type of cello concerts. This thesis is divided into three main chapters. The first chapter in the three subsections is focused on typing and characterization of string concerts. The second chapter in the six subsections devoted to the context of Western European traditions and peculiarities of the development of cello art in Russia. The third chapter in two separate sections and four smaller subsections focus on the personality of Karl Julievich Davydov, periodization of his work and analysis of his cello concerts.

The main idea of this thesis is to describe the development of german romantic opera focusing on the significant evolution of its drama, in the musical, textual and finally the interpretative aspects. A selection from their artworks (with regard to the main topic of thesis) was difficult and highly subjective. They have created a lot of outstanding feminine roles, so I chose five heroines that have served me as individual milestones along the way for almost a century. They have provided me with a suitable structure to map artistic maturation and the development of new, original and completely unique operatic form.

The main topic was the analysis of media investments to TV and internet, what are the trends of investment preferences to various media types and what factors influence these preferences. At the end I made a prognosis how will the TV and internet investments progress in next ten years.

This diploma thesis deals with the behavior of consumers in the food market during the period of economic growth. The goals of the work is to describe changes in consumer behavior and to identify factors influencing consumers when purchasing food. Thesis describes developmental trends of macroeconomic indicators related to the Czech Republic, household incomes, expenditures based on purpose and food consumption of Czech households. The source of information for this work is also a questionnaire sur-vey which provides background material for identifying factors affecting consumers when buying meat products, bakery products, milk and dairy products. Another source is indepth interviews that extend the results of the questionnaire and provide a more detailed picture of the issue. The results can ultimately be used by food business or pro-ducers.

Cells are the basic units of life. Studying complex tissues and whole organs requires an understanding of cell heterogeneity and responses to stimuli at the single-cell level. Even the cells, which belong to the same cell type, behave differently at a specific moment and contain different amount of mRNA. Quantitative polymerase chain reaction (qPCR) is one the most sensitive methods for the detection of mRNA, however, gene expression profiling in single cells leads to a large amount of missing data due to the fact that the transcript is missing, or is below the level of detection. Therefore, it is necessary to establish a new statistical approach for analysis of single cells. In this thesis the potential of single-cell gene expression profiling using the high throughput instrument Biomark, focusing on data analysis and biological interpretation, is discussed. Data normalization and handling of missing data are two important steps in data analysis that are performed differently at the single-cell level. Single cells are not normalized by reference genes but the number of cells as a normalizer is applied. Missing data are replaced by value, which is equaled one quarter of transcript amount in the cell. This dissertation is focused on the impacts of new legal regulations concerning non-profit making organizations on citizen associations. The introductory part gives the old and the new legal regulations of non-profit making organizations. The next chapter describes the transformation method of citizen associations(now societies) to other legal forms. This is followed by the suggestion of criteria when the organizations decide on the change of legal form a part of which is a case study showing the differences in taxation of individual legal forms. The final part describes results of research of transparency of non-profit making sector. Discussion contains recommendations how to solve the problems connected with the new legal regulation.

The theme of this thesis is groundbreaking 70s and 80s playing the tuba. The work divided into four chapters and subchapters. In the first chapter I write about major tuba players 70s and 80s. The first chapter and its subchapters consider crucial part of this thesis. The second chapter presents the most important authors and their songs that were to develop playing the tuba very important. The third chapter is devoted to the International Association of tuba, which was created at that time and for progress in the style of playing the tuba, it was very important. The fourth chapter is devoted to an interview with Jim Self and Pavel Trnka.

The overall structure of photosynthetic pigment-protein complexes and thylakoid membranes of various photosynthetic organisms was studied using electron microscopy.

There are deseribed the masseur touches, their technique and indications. The suitability and unsuitability of massage, effects, effects on the human body. It lists and uses of funds and equipment for sports massage. Based on the literature and captured images are developed methodology sheets for your convenience and are accompanied by photographs. The leaves have been tested in practice, a masseur in the fitness center.
