This diploma thesis deals with the issue of mental anorexia (AN), clinical and laboratory manifestations of the disease. The theoretical part is focused on summarizing the existing knowledge about the diagnosis, complications and treatment of AN. Attention is paid to the effects of the disease on individual organ systems and clinical and laboratory consequences. The practical part deals with the evaluation of laboratory parameters and clinical symptoms of patients monitored at the Department of Pediatrics and Inherited Metabolic Disorders, 1st Faculty of Medicine, Charles University and General Hospital in Prague. The performed clinical examinations include anthropometric measurements, bioimpedance, measurement of resting energy consumption by indirect calorimetry, measurement of skin algae with a caliper and comparison of the results of methods for determining the proportion of adipose tissue using a caliper and bioimpedances. All patients underwent EKG and cardiological examination, blood pressure measurement, heart rate. The laboratory examination was focused on the determination of serum protein levels, lipidogram, electrolyte levels, triiodothyronine and vitamin D. The issue of legislative and ethical requirements on research of innovative medicines became more intense in connection to the expansion of the pharmaceutical industry in last few decades. This master thesis aims to complexly describe and analyse theoretical bases of current legislative regulating the process of clinical trials of medicinal products for human use in the Czech Republic, as well as to take a closer look to the issue of ethical review administered by specialized ethical review committees. The author also deals with some specific responsibilities of the committees from the perspective of administrative law, especially focusing on aspects of legal nature of the positive opinions, which are one of the conditions for commencement of a clinical trial. The thesis is divided into nine chapters and the introductory chapter is focused on the general outline of the issue of clinical trials of medicines for human use in the Czech Republic. The aim of the second chapter is to define the basic concepts stated in the Czech medicinal products act, which are specific for this area of pharmaceutical law. My diploma thesis deals with the Clinical trial in the Czech Republic in the period from 1989 to present and on its impact on management in organizations dealing with clinical trials. The aim of this thesis is to identify key changes that occurred in the clinical trial and to identify how these changes infuenced the management of clinital trial organizations. This will be done based on the qualitative research. The theoretical part provides information about the history of the clinical trial, about internation and the Czech legislation, including ethical principles, the submission process including description of institution that are reviewing the clinical research. The theory also explains the issue of management, its organizational developments and its expected development in the future. In the research part I described the chosen research method, which extend from the semi- structured interview after the pilot part to the use of the focus group method - in the combination with semi-structured interview. Based on these interviews and focus group have been evaluated 7 parts to describe the evolution of the clinical trial and its impact on the management of clinical trials, including one topic that addresses the expected development of this field in the future. Kolonizace lidských plic nepatogenními streptomycetami / Colonization of human lung by non-pathogenic streptomycetes

Streptomycetes, primary soil saprophytic microorganisms are at the center of interest in many research groups, mainly because of their ability to produce a wide range of biologically active substances useful in medicine, biotechnology and agriculture. The marginal, and little explored areas are the interactions that streptomycetes create with humans. Recent metagenomic studies have shown that streptomycetes colonize the skin, the respiratory and possibly the urogenital tract of humans. In addition to apparent pathogens such as S. somaliensis and S. sudanensis, the clinical impact of these streptomycetes on human health is unknown. For this reason, a unique collection of non-pathogenic streptomycetes isolated from human clinical specimens was developed. The isolates were collected by the National Reference Laboratory for Pathogenic Actinomycetes in Trutnov, Czech Republic. On the basis of pilot studies, an isolate labeled TR42 was selected from the Trutnov collection, showing a very broad spectrum of biological activities. This strain was isolated from sputum from a patient with unknown respiratory diagnosis. Tomáš Kavka Abstract: Nonspecific back pain (nonspecific LBP) is the most common reason for visiting a doctor and it is the leading cause of activity limitation and inability to work by patients. The theoretical part of this thesis describes the issue of LBP and related clinical guidelines. In addition, it summarizes current research results of the influence of health professionals' fear-avoidance beliefs (FA beliefs) on their clinical decision making for LBP patients. The practical part relates to the research of FA beliefs and non-adherence to clinical guidelines regarding care of individuals experiencing LBP among health professionals in the Czech Republic. The work deals with evaluation of the incidence of individual types of urinary stones in dogs, and the effect of specialised clinical diets on the treatment of urolithiasis. The evaluted material consists of an aggregate of analytical results from extracted urinary stones from the Veterinary clinic Vltava, in the period from January 30, 2008 to September 12, 2011. The effect of administration of specialised clinical diets for urolithiasis was evaluated on the basis of case histories of sample patients with proven struvite urolithiasis and an occurence of calcium oxalates and urates. The analysed aggregate of laboratory results of tested urinary stones has shown that besides a high incidence of struvite, the frequency of occurence of calcium oxalates increases as well. In this analysis even up to an identical percentage with struvite occurence at 44,44 % (that is, 24) analysed concrements. A link has been established with overal greater proneness of males to the formation of urinary stones. In the patient group with analysed majority percentage of whewellite, only 12,5 % of the analysed aggregate were females. Considering the widely varied pedigrees of analysed patients, incidence of urolithiasis is expected with ever greater frequency even in dogs not predisposed by breed and crossbreeds. Specialised clinical diets are employed in the treatment and prevention of certain types of urinary stons with high success rate, which adjust the urine environment and thus reduce the risk of occurence of urinary stones. In my thesis I deal with canine hip dysplasia. The paper consist of two parts. The first part summarizes all the information available about this disease. These include factors influencing the emergence of the disease, clinical signs, evaluation systems, diagnosis, conservative ang surgical treatment, prognosis and prevalence. The second part is methodological. There are summarized data about particular breeds of dogs that were obtained Mastiff, Auvergne Pointing Dog, Beauceron, Fila Brasileiro, Brittany, Cavalier King Charles Spaniel, Czechoslovakian Wolfdog, French Pointing Dog, Italian Pointing Dog, Italian Wirehaired Pointing Dog, Small Blue Gascony Hound, Blue Picardy Spaniel, Newfoundland, Portuguese Pointing Dog, St. Germain Pointing Dog, Tibetan Terrier and Golden Retriever. In the end I gave breeding recommendations regarding to reducing the incidence of high grade of canine hip dysplasia.

The diploma thesis deals with the monitoring and evaluation of data in clinical research. Usual methods to identify incorrect data are one-dimensional statistical methods per each variable in the register. Proposed method enters directly into database and finds out outliers in data using machine learning combined with multidimensional statistical methods that transform all column variables of clinical register to one, representing one record of patient in the register. Algorithm of proposed method is written in Matlab.

This thesis deals with the theory of infrared thermography in medicine and its subsequent practical application in the creation of the program. As part of this diploma thesis, researches were carried out on the topic of thermovision imaging techniques, its use mainly in clinical medicine and also research in the field of image data processing. Theoretical knowledge is written into the first three chapters of this thesis. The next chapter deals with the design of a web application for the analysis of thermographic images of patients with vascular disease lower limbs and the PHP program environment with its particulars. In the second half of the thesis there is a detailed description of the practical creation of the mentioned application in PHP environment with the use of some PHP libraries. One chapter is also devoted to the discussion of the method of obtaining thermographic images for testing the created application and to the statistical evaluation of acquired data. This thesis is divided into 8 different chapters and also into many sub-chapters.
