This work is focused on the molecular conditionality of flax resistance to heavy metals. Attention was therefore devoted to the identification of proteins whose role in stress response to heavy metals have not been discovered yet in flax. Two contrasting flax varieties displayed differences in steady state levels of short thiol peptides (GSH and PC) and heavy-metal binding proteins (ferritin and lipocalin) which can detoxify cadmium via chelation. These results are the first published analysis of the proteomic response to cadmium exposure in commercially important annual crop with potential use in phytoremediation. Another of the objectives was to verify the possibilities of influencing the flax resistance to heavy metal using transgenosis. There were obtained a multiplied flax plants, in which the presence of a transgene was verified, and these lines were tested for tolerance and accumulation of cadmium in both in vitro and in the field conditions.

During the 21st century, pollution of environment was steadily increasing. Continuous monitoring of pollutants levels represents important part of effective environment protection. Demands for fast analysis of a large numbers of samples caused introduction of automation to analytical chemistry. Techniques based on flow analysis attracted big attention. They are able to decrease need for manual treatment of samples and decrease analysis duration. The advantage of flow-based analytical devices is also compatibility with several detection systems. Among others, electrochemical detectors are very attractive due to their high sensitivity, good selectivity and possibility of miniaturization. The presented thesis focuses on methods for detection of heavy metal and bacterial contamination. In order to it, electrochemical automatized methods and methods which utilized nanotechnology were developed.

Summary: This thesis is focused on mapping of contaminants released into the environment probably due to uranium mining. These chemical contaminants are used to reconstruct the sedimentary processes. In the thesis we evaluate geochemical situation in the catchment area in the locality Borecek (part of the municipality Ralsko) using the overbank sediments as sedimentary archives of pollution during the last decades by elements U, Ba, Zn, Ni and 226 Ra. A map of aerial survey of gamma activity was used to select the area of interest, for more accurate localisation we used a surface gamma spectrometry. On Borecek we retrieved ten depth profiles of alluvial sediments (up to 230 cm) and one in Mimon, all samples were subjected to X-ray fluorescence spectral analysis (XRF), across the floodplain we performed gamma wire logs. For interpretations of floodplains architecture electrical resistivity tomography (ERT) was used and selected profiles were analysed for the activity of 226 Ra and 210 Pb. Interrelation of 226 Ra and Ba proved that radiobarite is the main source of the gamma activity. 210 Pb / 226 Ra ratio was used as sediment age indicator. With depth profiles, gamma wire log, XRF and enrichment factors (LEF), we have tried to perform chemostratigraphic correlation of the sediments. As target elements U, Ba, Zn and Ni were chosen, whose concentrations are related to the grain size of the sediment. Furthermore, the theory of secondary pollution is developed.

Chemismus vody a sedimentů fluviálních jezer Labe / Water and sediment chemistry of fluvial lakes of the Elbe River

The presented paper aims at the problems of fluvial lakes in middle course of the Elbe River which has been influenced by the human activity since the Middle Ages. The oxbow lakes which are extremely precious ecosystems contributing to the stability of the river ecosystem show not only the changes of the course of the Elbe River. With regard to sedimenting a great amount of pollutants, these lakes also tell about the industrial pollution to which the river was exposed mainly in the second half of the 20th century due to insufficient precautions on industrial sources causing contamination. 5 cut lakes between Hradec Králové and Mělník have been studied within this research. These lakes differ in the intensity of communication with the river, in the distance from significant sources of industrial pollution and in the use of nearby land. The research included morphometric and bathymetric measures, observation of hydrological regime, seasonal measures in the water column and regular analysis of surface water samples. Profiles of sediments in the length that the used technology allowed were sampled, so that older anthropogenic load could be found out. Analýza trhu drahých kovů a diamantů / Analysis of precious metals and diamonds

The main intention of this thesis is the analysis of the factors that influence the precious metals market according to the selected methodology. The secondary intention is the consideration of the posibility to include the commodities traded on the precious metals market in the portfolio of investments. The introductory chapter deals with the listing of the commodities, the most important commodity exchanges and derivatives. The second chapter individually describes the precious metals. The first subsection of the second chapter analyzes the supply and demand for the individual metals. The following subsections focus on the factors that influence the price of the precious metals in history and in present time. The analytical part, which concludes this chapter includes the calculations of correlation coefficients, average growth coefficient and standard deviation of the factors that influence the precious metals. The last chapter of this thesis is dedicated to diamonds and is divided to two major subsections. The first subsection describes the history of the most significant diamonds, the characteristics of diamonds, the types of the deposits and the major producers of diamonds. The second subsection analyzes the prices of natural and synthetic diamonds and the development of profit of the selected company in terms of time. The last chapter deals with investments in diamonds.
