Využití spalin jako zdroje CO2 pro kultivaci řas v bioreaktorech / Usage of flue gas as a source of CO2 for algae cultivation in bioreactors

This master`s thesis is focused on possibility of use of flue gas as source of CO2 for algae cultivation in bioreactors. The first part of the master`s thesis is devoted to a qualitative and quantitative description of flue gas sources as CO2 source for algae cultivation. In the thesis industry sectors with the largest share of CO2 emissions on a global scale and in the Czech republic are summarized. On the basis of these data it was possible to determine cheap sources of CO2 for algae cultivation producing valuable biomass and other bio-products. The composition of flue gas from these sources was discussed in this section also with given expected outlook within the following years. The next part of the thesis is devoted to the influence of flue gas properties on algae cultivation. The impact of flue gas individual parameters on cell growth rate, quality and composition of biomass is described there. The description and design of the bioreactor is processed in the next part of the thesis. The experimental part of the thesis was performed on a bioreactor with similar properties. The next part of the thesis describes the effect of light on algae cultivation and the design of a regulation scheme for bioreactor light source. The experimental part of the thesis describes methods used to determine the concentrations of flue gas substances that could pass from flue gas into cultivation medium for algae cultivation to measure the changing composition of the flue gas in the closed experimental bioreactor system. The processed data from experimental measurements are presented and are discussed. The change in the composition of the culture medium for algae (without the presence of algae) due to flue gas aeration was monitored in the experimental part. It includes also the measurement of the change in the composition of the flue gas in time. All obtained and collected information are then summarized and discussed in the final part of the master`s thesis.

Osteoporosis is a disease of the bone metabolism which is characterised with a decrease of bone substance. The cause of this disease is the imbalance between the creation of a new bone substance by osteoblasts and the resorption of a bone tissue by osteoclasts, in favour of the bone resorption. The risk group of the development of this disease are women after menopause, who naturally register a decline of the estrogen hormone. Estrogen operates as an inhibitor of proosteoclastic factors such as receptor activator of NFκB ligand (RANKL), interleukin (IL)-1, IL-6 or TNF-α. The imbalance of the bone metabolism can also be caused by a disbalance in the production of Prostaglandin E2 (PGE2) and 1α,25-dihydroxyvitamin D3. They are strong mediators which can both stimulate and inhibit an osteoclastogenesis in vitro in concordance with the conditions of the culture/co-culture. This thesis focuses on the examination of an influence of those mediators (PGE2 in the concentration of 10-6 M and 10-8 M; 1α,25-dihydroxyvitamin D3 in the concentration of 10-8 M and 10-9 M) on the osteoclastogenesis from the rat PBMC at the presence of osteoblasts, with or without the combination of proosteoclastic factors macrophage colony-stimulating factor (M-CSF) and RANKL. The goal of my thesis was to evaluate origin of mastisis diseases and specify the best way to cure this disease with antibiotics. Secondary goal was to examin origin of things that cause these diseases, efficiency of the cure and how it is influenced by the stage of the illness. This research was made in cowshed, fron april to december 2018 with 136 specimen. During this period I took samples of milk, that had more than 800 000 somatic cells in 1ml of milk. Based of possitve NK test, samples of mammary gland were taken and cultivated on one used only Petri dish. Depending of the origin of the infection, the proper atibitics were chosen for the cure. After 7 to 14 days after antibiotics treatment, milk sample was taken from the cured part od mammary gland and It was tested to see, if the treatment was successful. 77% of the specimen were sick because of microorganisms and most of the specimen were afflicted the most during 1st lactation (36.8% to 47.1%). Following microorganisms caused mastisis - Staphylococcus spp. (KNS), Streptococcus uberis, Klebsiella oxytoca, Escherichia coli, Candida rugosa a Enterococcus faecalis. 36% of mastisis was caused by Staphylococcus spp. (KNS), followed by coliform bacteria (22%). 17% of mastisis was caused by Streptococcus uberis. 10.5% of specimen was infected by Enterococcus faecalis. Candida rugosa was found in 7.4% of the samples and in 9.5% of samples the diseasehad no specific microorganism responsible. From 105 infected cows, 68% of them fully recovered. At the end, based on the stage of the mastisis, efectivness of the treatmend was statistically evaluated. The evaluation showed, that stage of infection massively influence the result of the treatment.
