This work deals with the corporate culture, its description, significance and corporate culture analysis in the hotel chain Hong Ngoc Hotel Group, especially the Hong Ngoc Dynastie hotel. The work is divided into three parts. Content of the theoretical part are basic knowledge about corporate culture, the author analyses the term of corporate culture, its basic features and types of corporate culture. The practical part is focused on the hotel chain Hong Ngoc Hotel Group, where the Hong Ngoc Dynastie hotel is introduced. The author concerned with the analysis and description of the corporate culture of two hotels -- Hong Ngoc Dynastie and XY located in Prague. The last part of the study is dedicated to comparing this two corporate culture and improvement proposals for Hong Ngoc Dynastie hotel to approach its goals to become one of respected hotels at international level.

1 ABSTRACT Kateřina Muráriková Influencing of production of plant tissue culture of Trifolium pratense L. I The aim of this thesis was to check the influence of cinnamic acid, as a precursor of the phenylpropanoids biosynthesis, on the flavonoids production of Trifolium pratense L. suspension culture, to compare two varieties of this plant, the DO-8 one and the Tempus one, in terms of the effect of this precursor, and to help achieve the successful increase of the production of secondary metabolites by the Trifolium pratense L. explant cultures. Concerning the DO-8 variety, the changes of the isoflavonoids production as a result of the cinnamic acid influence have been also tested. The suspension cultures were being cultivated on the Gamborg's nutrient medium with the addition of 2 mg.l-1 2,4-dichlorphenoxyacetic acid and 2 mg.l-1 6-benzylaminopurine. The cultivation was being realised on a slow moving roller at temperature of 25 řC and in the period of 16 hours of light and 8 hours of dark. The cinnamic acid was added in the four concentrations and its influence was observed after 6, 24, 48 and 168 hours. Milk thistle, Silybum marianum L. Gaertn., is a source of flavonoid taxifolin and flavonolignans - silymarin complex (silybin, silydianin, silycristin and isosilybin). Milk thistle is usually obtained by field cultivation. Alternative way for getting the active components, is the use of in vitro cultures. But the production of secondary metabolites by the in vitro cultures is low in comparison with plant. One of the possibilites how to increase this produciton is the method of elicitation. In this study, ethephon as the elicitor, in the concentrations of 500 µmol/l, 400 µmol/l, 200 µmol/l, 100 µmol/l and 50 µmol/l was used with the aim to increase secondary metabolite production in suspension and callus cultures. The effect of ethephon was compared to its inhibitor (AgNO3, 120 µmol/l). The levels of flavonolignans and taxifolin were measured by the method of HPLC. The samples were taken 24, 48, 72, 96 and 168 hours after the ethephon application and inhibitor treatment. The nutrient medium of suspension culture was also tested for the possibity of secondary metabolites releasing into medium. The highest content of flavonoid taxifolin was found in the suspension culture medium after 48 h treatment with ethephon in conc. of 400 µmol/l. Genista tinctoria, family Fabaceae, is a potent source of isoflavonoids (genistin, genistein, daidzein, formononetin, biochanin A) with a wide spectrum of potential medical impact. Genista also contains quinolizidin alkaloids (cytisin, anagyrin, lupanin, spartein, etc.), which are toxic. The reason why in vitro cultures are used is an absence of toxic alkaloids production and higher yield of isoflavonoids in comparison with intact plant. For an increase of isoflavonoid production method of elicitation is beeing used. Isoflavonoids are studied for their phytoestrogenic effects, for which they could be used in treatment of postmenopausal symptoms and even in treatment of hormon-dependent tumours. The elicitor ethephon in concentration of 7000 µM, 700 µM and 70 µM was used in this work. The effect of ethephon inhibitor (AgNO3) in concentration of 120 µM was investigated too. Samples were examined after 24, 48, 72, 96 and 168 hours and then analysed by HPLC method. An effect of ethephon and its combination with AgNO3 was observed in callus and suspension cultures. Release of isoflavonoides into culture media was studied too. The work is concerned with the archaeological finds from Lovosice, Ustecky kraj, CR obtained in 2002. The major part consists of description and drawing documentation of 15 well-equipped from the Early Iron Age. Movable artifacts are analyzed typologically and chronologically. The funeral rite is evaluated in the frame of spatially-functional relations between human relicts and burnt offerings. Immovable finds are analyzed with respect to the parameters of holes, construction and things. In the same way the graves are categorized into the finding groups. The funeral site is evaluated with regard to the form, structure, total range and chronological trend. The main goal consists in a definition of evolutionary horizons of the funeral site and their comparison between Czech and Central European chronological systems. These horizons, in the future, can make one of the main pillars for evaluating grave and settlement finds of Hallstatt age in NW and C Bohemia. The area of Lovosice necropolis is interpreted as a site of central relevancy and the dead as elite with significant influence over the cultural and economic development of the region. II The aim of this thesis was to observe the effect of the abiotic elicitor vanadyl sulfate on the production of flavonoids in the suspension culture of Trifolium pratense L., variety DO-8 and variety Sprint. The effects on the production of isoflavonoids caused by the elicitor have been also studied. The cultures were cultivated at the temperature of 25 žC, 16 hour light/8 hour dark period, in the Gamborg's nutriet medium with the addition of 2 mg.l-1 of 2,4- dichlorophenoxyacetic acid and 2 mg.l-1 of 6-benzylaminopurine. Vanadyl sulfate was added in four concentrations and its influence was observed after 6, 24, 48 and 168 hours. The quantity of flavonoids was determined spectrophotometrically according to the Pharmacopoeia Bohemica 2009. The quantity of isoflavonoids was determined using HPLC. The maximal content of flavonoids, 0,459 %, was measured in the suspension culture of Trifolium pratense L., variety DO-8, taken after 48 hours of action of elicitor vanadyl sulfate, which concentration was 1 µmol.l-1 . The main purpose of this thesis is the analysis of fictional characters, their role in Japanese visual culture and society in general. For a better understanding of the argumentation that follows in chapters 2 to 4, I first introduce the reader to the world of postwar Japanese visual culture, its most iconic works and characters. In the following chapter I observe fictional characters from a psychological and sociological standpoint, and highlight their significance for contemporary society. Chapters 3 and 4 are dedicated to some of the most significant subcultures in postwar Japan - the shojo and otaku. In each of these chapters I first offer a synopsis of their respective histories and proceed to analyze some of the main trends associated with these subcultures; trends that have subsequently led to a major shift in consumer culture and a significant popularization of characters, the character business and its products. I argue that the most significant phenomenon associated with shojo culture is kawaii, the aesthetic of cuteness. Kawaii is arguably one of the defining aesthetic categories of contemporary Japan and is closely linked to postwar visual culture and the character goods industry. The study is aimed to enhance in vitro production of secondary metabolites in Genista tinctoria L. via elicitor treatment. The different levels of elicitor concentration - selenium dioxide were utilized to affect the quantity of isoflavonoids occurred in cultures. Experiment was perfomed in callus and suspension cultures on MS nutrient media supplemented with 10 g l-1 of NAA (α-naphtylacetic acid) as growth regulator. The elicitor was added in the form of solution in concentrations of 9,012.10-3 mol l-1 ; 9,012.10-4 mol l-1 and 9,012.10-5 mol l-1 . It was exposed for 6, 12, 24, 48, 72 and 168 hours. The content of isoflavonoids was determined by HPLC in dry weight (DW) and medium. The most effective production of genistin (6,20 mg. g-1 DW, 8,30 mg. g-1 DW) in callus culture was measured. It was reached in concentrations of 9,012.10-4 mol l-1 and 9,012.10-5 mol l-1 after 168 h elicitor treatment. The second most satisfactory genistin level 5,20 mg g-1 DW was detected after elicitor application in concentration of 9,012.10-4 mol l-1 after 6 h. The content of genistein, daidzein and formononetin in callus culture was low and in the most cases equal zero compared to control samples. In vitro cultures of medicinal plants - IXX The subject of this thesis is the evaluation of secondary metabolites production in in vitro cultures of Silybum marianum L. after elicitor treatment. In this study selenium dioxide as elicitor in concentrations of 9,012.10-3 mol/l; 9,012.10-4 mol/l; 9,012.10-5 mol/l was used. The samples were taken after 6, 12, 24, 48, 72 and 168 hours of elicitor treatment. The effect of elicitor was compared with control samples, which were cultured without elicitation. The content of taxifolin and flavonolignans was determined by the method of HPLC. The results showed, that almost all observed metabolites were released into a nutrient medium. Cells of callus and suspension cultures produced only small amounts of taxifolin (0.01 mg/g DW). Taxifolin and silymarin complex releasing into nutrient media was observed as in control and also in the elicitated samples of callus and suspension cultures. Selenium dioxide elicitation caused statistically significant increases in releasing taxifolin and silymarin complex into the nutrient medium. The statistically significant releasing of flavonolignans (2.2 mg/100 ml) to the medium of suspension culture was reached after 72 hours of treatment with selenium dioxide in concentration of c1 (9,012.10-3 mol/l).
