Vývoj HPLC-FLD metody pro stanovení žlučových kyselin ve žluči myší / The Development of HPLC-FLD Method for the Determination of the Bile Acids in the Bile of Mice

Aim of the master thesis was the development and the optimization of the HPLC method with fluorescence detection for the determination of the bile acids in the bile of mice. On the basis of literature, the HPLC-FLD method has been proposed and optimized for the quantification of eight bile acids (cholic acid, glycocholic acid, deoxycholic acid, glycodeoxycholic acid, chenodeoxycholic acid, glycochenodeoxycholic acid, α-muricholic acid, and λ-muricholic acid) in the biological material. 1,2-benzo-3,4-dihydrocarbazole-9-ethyl-p-toluenesulfonate is suitable derivatization reagent for labeling of the bile acids. The calibration dependences, limits of detection and quantification (in the range of 0.88-1.78 μmol dm -3 ) for all bile acids are measured. In the biological sample five bile acids have been successfully identified and then determined (cholic acid, glycocholic acid, glycodeoxycholic acid, glycochenodeoxycholic acid, and α-muricholic acid). Vliv přídavku tuzemských vín při výrobě vinných klobás / The effect of the addition of local wines in the production of wine sausages

The aim of this diploma thesis was to determine the effect of the addition of several types of local wines in the production of wine sausages. Selected wine varieties were characterized using basic and modern analytical methods (liquid chromatography with a diode array detector) and the obtained results were compared with the results of sensory analysis.The sensory analysis was divided into two parts, the first evaluated six non-traditional wine varieties used in the production of wine sausages and the second part evaluated samples with the addition of the traditionally used wine variety Veltlínské zelené, but in three different volumes. Using the Statistica program, the data obtained from the sensory analysis were processed and the mutual correlation was evaluated. Using the gas chromatography method with a flame ionization detector the fatty acid content of a standard sample of wine sausage has been determined. Based on all the determined results, the best wine variety for the production of wine sausages was chosen, it was the blue grape variety André.

Enkapsulace aktivních látek a jejich aplikace v potravinářském průmyslu / Encapsulation of active components and their applications in food industry

The Diploma thesis is focused on encapsulation of bioactive compounds which are contained in selected superfoods for the purpose of application to childrens nutrition supplements. In view of the nature of selected samples, which are seaweed, walnuts, hemp seed and flax seed, the emphasis is put on the content of omega-3 and omega-6 acids. The theoretical part introduces topic of children food, further it describes the nature of selected superfoods and last but not least it contains screening of available childrens food supplements containing omega-3 fatty acids on the market. In the experimental part, an optimization of the appropriate type of extract was made. A 24 hour aqueous macerate and 20% ethanol macerate were selected for next analyzes. Samples were characterized by content of polyphenols, flavonoids and the antioxidant activity and they were used to forming of extracts were encapsulated into liposomes for which their encapsulation efficiency, stability and long-term stability have been tested. Hexan extracts were also prepared for the same reasons. It was found that liposomes of selected superfoods should not be stored for 12 weeks. The samples were used analyzed for the carbohydrate content by the duboise method, a protein content by the Kjeldahl method and the determination of the fatty acid profile by gas chromatography. Furthermore, the content of carotenoids and chlorophylls in algae samples was determined by spectrophotometry and this analysis was verified by high-performance liquid chromatography. The technique of high-performance liquid chromatography was also used to verified a content of polyphenols in the algae samples. Finally, a sensory analysis of prepared alginate supplements containing algae and cold-pressed oils was performer in combinations to achieve a higher omega-3 and omega-6 fatty acid content. The F01-labeled sample containing wakame algae was evaluated as the best one.

Komplexy miRNA s cílovou RNA sekvencí / miRNA complexes with the target RNA sequence

miRNAs are short non-coding RNA molecules which play role in post-transcriptional regulation of gene expression. Thus, miRNAs are related to a variety of physiological processes and diseases, e.g. some tumours or viral infections. In this diploma thesis we studied temperature-induced dissociation of complexes that simulated pairing of miRNA with target mRNA. For these purposes we used UV absorption spectroscopy and Raman spectroscopy. Measured spectra were then treated by means of a factor analysis. The aim of the thesis was to determine thermodynamic parameters of the complexes dissociation and to clarify how inner loops and bulges affect the stability of oligonucleotide duplexes. Also, we have tested the suitability of the used approach for study of oligonucleotide complexes. We have shown that factor analysis of temperature dependent UV absorption spectra is a proper base to determine thermodynamic parameters of the duplex dissociation. Raman spectra were affected by large errors, nevertheless they enabled to observe some structural changes related to the transition. It has been shown that the bulge loop present in the studied complex reduces both its flexibility and the energetic preference of its formation.

Long-chain fatty acids (LCFA) are the primary energy source in the myocardium and an imbalance in the LCFA and glucose utilization could cause cardiovascular diseases. More than 50% of LCFA uptake by the heart is mediated by the fatty acid translocase CD36 and disruption of its function has been shown to impair cardiovascular functions. The spontaneously hypertensive rat (SHR) harbors a deletion variant of the Cd36 gene that results in reduced LCFA transport into myocytes. Therefore, the main aim of this thesis was to investigate the importance of a functional CD36 to sustain normal physiological functions of the heart. We used SHR and two genetic modified SHR strains, the congenic SHR-4 and the transgenic SHR-Cd36, with fully functional CD36. They differ in the CD36 expression and in the manner how they were derived from the SHR. CD36 has been proven to play a role in the pathogenesis of insulin resistance. Therefore we analyzed the effect of a functional CD36 on insulin resistance and protein kinase C (PKC) expression, which is known to be involved in the mechanism of insulin resistance, in the heart of SHR-4 and SHR. We showed that the SHR-4 had lower serum free fatty acids (FFA) and triacylglycerols (TAG) concentrations, indicating improved insulin sensitivity. Furthermore, SHR-4 had increased...

Vliv polymorfních variant kandidátního lokusu na spektrum mastných kyselin kravského mléka

Global studies show the effect of polymorphism of selected genes on dairy production and fatty acid spectrum. The aim of the diploma thesis was genotyping of candidate FASN locus with a focus on milk yield and fatty acid spectrum depending on genotype. The thesis describes the characteristics of cow's milk, milk fat and fatty acids in milk fat. Furthermore, the thesis deals with the genome of cattle and the potential influence of polymorphism of candidate genes affecting fatty acids of milk fat. Genotypes for FASN were determined by the PCR-RFLP method, the milk yield of selected dairy cattle was statistically evaluated according to the milk production indices for 1st lactation in individual breeds, and the determination of fatty acids was performed by spectrophotometry followed by statistical evaluation. The resulting genotypes in selected breeds were only two, the GG genotype with a higher relative frequency than the genotype AG. There is no statistically significant difference between FASN genotypes depending on the milk yield and the fatty acids spectrum.

Význam biosyntetické a katabolické dráhy cholesterolu u nádorových a zánětlivých onemocnění / The importance of biosynthetic and catabolic pathway of cholesterol in inflammatory and tumor diseases

This thesis focuses on the importance of intermediate products of biosynthetic and catabolic pathway of cholesterol. The aim of the first part of the thesis is mainly to investigate, whether statins (HMG- CoA reductase inhibitors) possess antitumor properties and to compare the differences in antitumor potential of individual statins. The other part of the thesis aims at the utilization of 7α-hydroxycholest-4-en-3-one (C4), a promising marker of cholesterol 7α-monooxygenase (CYP7A1) activity and bile acid malabsorption. We demonstrated antitumor effect of statins on an experimental model of pancreatic cancer. Individual statins, however, differed significantly in their efficacy, depending on their physico-chemical properties. Our data suggests, that the most likely (but not the only) mechanism of antitumor effect of statins is decreased prenylation of signaling proteins, especially Ras protooncogene. We set up a reliable method for measurement of C4, which facilitated our research in CYP7A1 regulation. We demonstrated, that promoter polymorphism -203A>C might affect CYP7A1 activity, that diurnal variability of CYP7A1 activity might be triggered by insulin, and that insulin resistance in patients with non-alcoholic fatty liver disease impedes the feedback regulation of CYP7A1, which may lead to disease...

Chemické modifikace polypropylénových povrchů / Chemical modifications of polypropylene surfaces

Diploma thesis is focused on modification and characterization of polypropylene (PP) products surfaces by the chemical way. In theoretical part of thesis are summarized the current knowledge of this topic including characterization methods. Polypropylene surfaces were modified by acrylic monomers (acrylic acid, acrylic acid with 20 mol% content of sodium acrylate and acryl amide). Modified surfaces were characterized by infrared spectroscopy with ATR technique. The comparative parameter was carbonyl index determined from absorption peaks of functional methyl groups (2950 cm-1 ) and functional carbonyl groups (1700-1750 cm-1) of FTIR-ATR spectra. Determination of effect of initiator concentration on the modification process was done for acetophenone and benzophenone. There were determined the time dependence of carbonyl index for time from 0 to 600 s of mentioned monomers. In the same condition were modified test specimens designed for preparation of adhesive joints. Joints were bonded by cyanoacrylate adhesive that provide reaction with surface of polyacrylic acid. There were founded conditions of modification that provided strength of adhesive joints higher than strength of original PP. There were identified changes of surface roughness by using of confocal microscopy.
