Graphene is a material suitable for electronic applications including sensors and biosensors operating in atmospheric conditions and at varying relative humidities. This work concerns the charge diffusion in Hall bar shaped graphene based field effect transistor that is simultaneously investigated by Kelvin probe force microscopy and macroscopic transport measurement. Furthemore it studies the possibilities of graphene Hall bar modification by local anodic oxidation, local cathodic hydrogenation and mechanical lithography.

Caracterização imunoistoquímica da infiltração de células imunes na histiocitose de células de Langerhans em pacientes pediátricos e adultos / Immunohistochemical characterization of immune cell infiltration in pediatric and adult Langerhans cell histiocytosis

A histiocitose de células de Langerhans (HCL) é uma neoplasia mieloide inflamatória comumente afetando pacientes pediátricos e apresenta frequentemente mutações ativadoras somáticas em genes da via MAPK, incluindo BRAF e MAP2K1. Vários estudos sugerem que as células lesionais da HCL podem recrutar e modular células inflamatórias e cujas citocinas parecem fornecer sinais recíprocos de sobrevivência celular. Para o presente estudo foram selecionados 15 casos de HCL (10 crianças, 5 adultos), sendo as amostras de tecido avaliadas através de imunoistoquímica utilizando marcadores para macrófagos (CD68 e CD163), células dendríticas maduras (CDm) (CD83 e CD208), linfócitos T regulatórios (LTregs) (CD4, CD25 e FOXP3) e linfócitos citotóxicos (LCs) (CD56, CD57, perforina e granzima B). Além disso, marcadores de células B (CD20), células T (CD3, CD8) e confirmatórios de HCL foram analisados. Todos os casos de HCL foram positivos para S100, CD1a, CD207 e CD4; enquanto que Bcl-2 e Ciclina D1 foram positivos em 13/15 (86,7%) casos. No microambiente imune intralesional, macrófagos M2 (CD68+/CD163+), seguidos por LTregs, foram as populações celulares mais predominantes. Em quantidade significativamente menor, foram observadas CDm, seguidas por escassos LCs. Considerando a população linfoide, linfócitos T CD3+ foram mais numerosos do que linfócitos B CD20+. Dentro dos linfócitos T, linfócitos T CD4+ foram mais numerosos do que linfócitos T CD8+ (p<0,05). Nossos resultados sugerem que a infiltração de células imunes na HCL, provavelmente através de mecanismos pró-tumorais, inflamatórios e/ou imunossupressores mediados por citocinas, pode promover o desenvolvimento e sobrevivência das células lesionais da HCL, fornecendo uma justificativa para a combinação de imunoterapia e terapia gênica (BRAF) na HCL / Langerhans cell histiocytosis (LCH) is an inflammatory myeloid neoplasia often affecting children with constitutively somatic activating mutations in MAPK pathway genes including BRAF and MAP2K1. Several studies suggest that LCH cells can recruit and modulate inflammatory cells and whose cytokines appear provide reciprocal survival signals. For the present study, 15 cases of LCH (10 children, 5 adults) were selected, and the tissue samples were evaluated through immunohistochemistry using markers for macrophages (CD68 and CD163), mature dendritic cells (mDC) (CD83 and CD208), regulatory T-cells (Tregs) (CD4, CD25 and FOXP3) and cytotoxic lymphocytes (CLs) (CD56, CD57, perforin and granzime B). Moreover, B-cell (CD20), T-cell (CD3, CD8) and LCH markers were analyzed. All LCH cases were positive for S100, CD1a, CD207 and CD4, while Bcl-2 and Cyclin D1 were positive in 13/15 cases (86.7%). In the immune microenvironment, M2-polarized macrophages (CD68+/CD163+), followed by LTregs, were the predominant cell populations. In a significantly lower amount, mDC were observed, followed by scarce CLs. Moreover, CD3+ Tcells than CD20+ B-cells were more numerous (p>0.05), the former presenting a higher number of CD4+ than CD8+ T-cells (p<0.05). Our results suggest that immune cell infiltration in LCH, probably through cytokine-mediated pro-tumoral, inflammatory and/or immunosupressive mechanisms, can promote LCH cell development and survival, providing a rationale for combining immunotherapy and BRAF-targeted therapy in LCH

Regression models to assess the thermal performance of Brazilian low-cost houses: consideration of natural ventilation / Modelos de regressão para avaliação do desempenho térmico de habitações de interesse social brasileiras: consideração da ventilação natural

Building performance simulations [BPS] tools are important in all the design stages, mainly in the early ones. However, some barriers such as time, resources and expertise do not contribute to their implementation in architecture offices. This research aimed to develop regression models (meta-models) to assess the thermal discomfort in a Brazilian low-cost house [LCH] during early design. They predicted the degree-hours of discomfort by heat and/or by cold as function of the design parameters changes for three Brazilian cities: Curitiba/PR, São Paulo/SP, and Manaus/AM. This work focused on using the meta-models to evaluate the impact of the parameters related to natural ventilation strategies on thermal performance in LCH. The analyzed Brazilian LCH consisted in a naturally ventilated representative unit developed based on the collected data. The most influential parameters in thermal performance, namely as key design parameters, were building orientation, shading devices positions and sizes, thermal material properties of the walls and roof constructive systems as well as window-to-wall ratios (WWR) and effective window ventilation areas (EWVA). The methodology was divided into: (a) collecting projects of Brazilian LCH, and based on that a base model that was able to represent them was proposed, (b) defining the key design parameters and their ranges, in order to compose the design space to be considered, (c) simulating thermal performance using EnergyPlus coupled with a Monte Carlo framework to randomly sample the design space considered, (d) using the greater part of the simulation results to develop the meta-models, (e)using the remaining portion to validate them, and (f) applying the meta-models in a simple design configuration in order to test their potential as a support design tool. Overall, the meta-models showed R2 values higher than 0.95 for all climates. Except for the regression models to predict discomfort by heat for Curitiba (R2 =0.61) and São Paulo (R2 =0.74). In their application, the models showed consistent predictions for WWR variations, but unexpected patterns for EWVA. / Simulações do desempenho de edificações são ferramentas importantes em todo processo de desenvolvimento do projeto, especialmente nas etapas iniciais. No entanto, barreiras como tempo, custo e conhecimento especializado impedem a implementação de tais ferramentas nos escritórios de arquitetura. A presente pesquisa se propôs a desenvolver modelos de regressão (meta-modelos) para avaliar o desconforto térmico em uma habitação de interesse social [HIS] brasileira. Estes meta - modelos predizem os graus-hora de desconforto por calor ou por frio em função de alterações nos parâmetros de projeto para três cidades brasileiras: Curitiba/PR, São Paulo/SP e Manaus/AM. O foco deste trabalho é o uso dos meta-modelos para avaliar o impacto de parâmetros relacionados com estratégias de ventilação natural no conforto térmico em HIS. De modo geral, os meta-modelos apresentaram R2 superiores a 0,95 para todos os climas, exceto os meta-modelos para predizer desconforto por calor para Curitiba (R2 =0,61) e São Paulo (R2 =0,74). Na fase de aplicação, os modelos mostraram predições consistentes para variações na área de janela na fachada, mas incoerências para variações nas áreas efetiva de abertura.

In this research study three subject didactical measuring instruments were compared with regard to certain psychometric characteristics of a group of grade nine learners in the Tzaneen and Phalaborwa region, in the Northern Province. The primary goal of this study was to optimize learning competencies with regard to subject context. The SOM-, LEMOSS(II)- and LCH-questionnaires can be applied at the start or at the end of an academic year to an individual or to a group in order to identify learners who need specific help, counseling, remediation and support in the subjects mathematics, natural and domestic sciences in order to facilitate learning. The following aspects have been handled in this study, such as: In chapter 1 an attempt is made to clarify and define the title and certain relevant concepts. The research statement and questions are formulated. The research design is briefly discussed and the primary goal and objectives are set. Other aspects that enjoy attention include the types of research that were used, the method of sampling, data gathering and analysis, as well as the progress of the study. Literature on subject didactics is discussed in chapter 2 for the conceptualising of basic related aspects by means of a macro-model for the optimising of learning competencies in the context of a specific subject. This model emphasises subject content, form and competencies that should be taken into consideration when planning a learning opportunity. Other aspects concerning learning, namely relevant theories, approaches, models, styles, strategies and motives were discussed in order to facilitate learning in mathematics, natural and domestic sciences. In chapter 3 the SOM-, LEMOSS(II)- and LCH-questionnaires as diagnostic instruments are described. The diagnostic value, background, categories and psychometric aspects of the measuring instruments are briefly discussed. In chapter 4 the method of investigation used for the comparison of the SOM- LEMOSS(II)- and LCH-instruments regarding their reliability and validity is described and it includes the goal, hypothesis and method of sampling as an extension of the research design. The way in which descriptive and inferential statistics are conveyed is theoretically described In chapter 5 the results and qualitative conclusions of the empiric-analytical investigation are included. The SOM-, LEMOSS(II)- and LCH-questionnaires are compared with regard to reliability and validity of a group grade nine learners in the Tzaneen and Phalaborwa region. Reliability is discussed by means of test-retest reliability and pared observations. Pared observations include staggering, locality and pared t- and sign-rank testing. Validity refers to criteria referenced validity with regards to simultaneous and predictable validity. Simultaneous validity is described by means of the relation between the subject mark and the field, and by stepwise and multiple regression analysis. In chapter 6 the study is concluded with the brief answering of the research questions and by describing the research findings by means of the research statement. Algoritmy přepočtů gamutů ve správě barev / Gamut mapping algorithms in color management

The thesis deals with colors - their representation in digital devices and how to provide the best color preservation accross different devices. In the first part of the work, the knowledge of colors and human vision is briefly summarized. Then color models and color spaces are elaborated, mainly those device independent. Spectrum of colors viewable or printable on a device - the gamut - is different for every device and there's a need of precise reproduction or record of color. That's why the system of color management is described further and especially the gamut mapping approaches and algorithms are mentioned. In the second part of the work, the implementation of how two algorithms of color gamut mapping (HPMINDE, SCLIP) can be implemented in MATLAB is described. In the third and last part of the work, the results of implemented algorithms are presented and discussed. These results are compared to results of commonly used color gamut mapping technique (Adobe Photoshop).
