Dagmar Říhová, Ph.D. ANNOTATION This diploma thesis is focused on leeches (Annelida: Hirudinea) in the Czech Republic. It provides a description of all species occurring in the Czech Republic, as well as the key to their classification into families. A detailed guideline about the organization of field trip and implementation of extracurricular teaching is part of this thesis. The field trip is focused on sampling, observation and determination of leeches in an excursion to a site with a traditional occurrence of the European medical leech Hirudo medicinalis - national nature reservation Hrabanovská černava. In addition, the work defines the advantages, disadvantages and other possibilities of application of the proposed program, and the inclusion of extracurricular education. Preparation for Nerve Membrane Potential Readings of a Leech, Laboratory Setup and Dissection Process

A well documented laboratory setup, leech preparation process, and bio-potential data recording process are needed. Repeatability and quality data recordings are essential and thus dictate the requirements of the laboratory setup and processes listed above. Advances in technology have both helped and hindered this development. While very precise equipment is required to record the low voltage bio-potentials, noisy electronic equipment and wires surrounding the work area provide high levels of interference. Proper laboratory setup and data recording processes, however, limit the unwanted interference. Quality data can only be recorded from a properly handled and prepared leech subject. Proper setup and procedures result in quality recordings which lend a clean signal for furthering the understanding of nerve functionality. The electrophysiology lab at California Polytechnic State University in San Luis Obispo is an example of a proven lab setup for high quality signal capture.

Análise proteômica do complexo salivar da sanguessuga Haementeria depressa / Proteomic analysis of the salivary complex from the leech Haementeria depressa

O complexo salivar da sanguessuga H. depressa é composto pelas glândulas salivares e probóscide. Análises bidimensionais (2D) mostraram que a maioria das proteínas apresentam pI entre 3.5 à 9.5 e MM entre 10 à 105 kDa. O mapa 2D do complexo salivar apresentou 352 spots totais, sendo 249 exclusivos da saliva (corado por nitrato de prata) e 219 spots totais (corado por Coomassie Blue). As proteínas foram identificadas após sequenciamento tandem MS (proteoma) pela complementariedade às sequências traduzidas do cDNA (transcriptoma). As proteínas mais abundantes foram: antiagregante plaquetário; miohemeritrina e anidrase carbônica. Estas proteínas devem exercer um papel na digestão ou anticoagulação do sangue durante a alimentação. A zimografia também identificou uma protease gelatinolítica (45 kDa). Porém, lefaxin (inibidor de FXa) e hementerina (fibrinogenolítico e inibidor de agregação plaquetária) não foram identificados por estas técnicas biotecnológicas, o que mostra a necessidade de técnicas adicionais para a completa elucidação da constituição desta amostra. / The salivary complex from H. depressa leech is composed of salivary gland and proboscis. Two-dimensional (2D) analysis showed that majority of proteins have pI from 3.5 to 9.5 and MW from 10 to 105 kDa. The 2D gel of salivary complex showed 352 total spots, 249 of them were from saliva (silver stained) and 219 total spots (Coomassie Blue stained). Proteins were identified after tandem MS sequencing (proteome) and complementar analysis of translated sequences from cDNA (transcriptome). The most abundant proteins were: antiplatelet protein; myohemerytrin; carbonic anhydrase. These proteins may have a role in digestion or antihemostatic system during blood feeding. The zymographic assay identified a gelatinolytic protein (45 kDa). Duty Cycle Maintenance in an Artificial Neuron

Neuroprosthetics is at the intersection of neuroscience, biomedical engineering, and physics. A biocompatible neuroprosthesis contains artificial neurons exhibiting biophysically plausible dynamics. Hybrid systems analysis could be used to prototype such artificial neurons. Biohybrid systems are composed of artificial and living neurons coupled via real-time computing and dynamic clamp. Model neurons must be thoroughly tested before coupled with a living cell. We use bifurcation theory to identify hazardous regimes of activity that may compromise biocompatibility and to identify control strategies for regimes of activity desirable for functional behavior. We construct real-time artificial neurons for the analysis of hybrid systems and demonstrate a mechanism through which an artificial neuron could maintain duty cycle independent of variations in period.

High temperature forearc metamorphism and consequences for sulfide stability in the Pacific Rim Terrane, British Columbia

The Pacific Rim Terrane in British Columbia is a group of fault-bound forearc metasedimentary and metaigneous rocks subcreted to Wrangellia, comprising three lithological units: the Leech River Complex (LRC), the Pandora Peak Unit (PPU), and the Pacific Rim Complex. Of these three, the LRC and PPU were subject to an elevated thermal metamorphic event which locally overprinted typical low temperature, medium pressure forearc assemblages with low greenschist through amphibolite facies assemblages. The field study shows that biotite, garnet and staurolite isograds occur concentrically in the LRC, centered on the Leech River fault, which separates the Pacific Rim Terrane from the underlying Metchosin Igneous Complex of the Crescent terrane. Local thermal overprint in the PPU is sub-biotitic and is characterized by local replacement of prehnite-pumpellyite and lawsonite-bearing assemblages with muscovite ± chlorite. Multi-method geothermobarometry shows peak metamorphic temperatures from ~230 °C in the northern PPU to ~600 °C near the Leech River fault at ~4 kbar, and isotherms are continuous across the LRC-PPU boundary. The interfoliated Tripp Creek metabasites and Eocene Walker Creek intrusions do not control the distribution of isotherms, and syn-metamorphic felsic sills rarely have contact aureoles. Intercalated metabasites show two distinct rare earth element (REE) patterns, including NMORB-like light REE depletion among most Tripp Creek metabasites, and light REE enrichment in PPU metabasites. The lack of thermal aureoles associated with metabasites, and interlayered garnetite bands with negative Ce-anomalies attributed to seafloor hydrothermal processes, suggest the Tripp Creek metabasites are not syn-metamorphic sills and formed prior to accretion. The subcretion of then recently formed oceanic crust belonging to the Crescent terrane is identified as the probable cause of anomalously high temperature forearc conditions, as well as possible proximity to an Eocene mid ocean ridge. The high temperature metamorphic rocks in the Pacific Rim Terrane document the conversion of inherited primary pyrite to pyrrhotite in carbonaceous metasediments. S-inclusive pseudosections for LRC protoliths predict a low temperature (<420 °C) narrow pyrite desulfidation window that produces pyrrhotite and releases negligible S to the fluid phase. Conversely, sulfide petrography in the LRC shows pyrite can persist up to ~550 °C as inclusions in andalusite and staurolite porphyroblasts, as well as possibly in the rock matrix. S contents in carbonaceous pelites show a marked reduction at medium grade, associated with a dearth of visible sulfide in LRC phyllites. Sluggish pyrite desulfidation, pyrrhotite desulfidation, and terrane-scale S mobility are interpreted as the driver for mobility of intra-terrane sourced Au, leading to the formation of a hypozonal orogenic Au deposit in the central LRC. / Graduate / 2022-06-11

Mapping South African internet user's opinions about the use of peer-to-peer file sharing technology to infringe on copyrighted films and/or television series content

The aim of this study was to investigate the various reasons consumers continue to infringe on copyrighted content, specifically in the South African context, even if the law forbids it. This investigation is two-fold since it also recognises that there are many individuals who do not infringe on copyrighted content even though they have access to peer-to-peer file sharing technology. This information could prove valuable since it can then be used to find comprehensive market-led solutions to the problem that targets the end-user. This study adopted a mixed method approach in order to cross validate findings and to reveal aspects of empirical reality. The target population for this study consisted of 100 adult South Africans who have access to the internet. Data was collected through an online, self-administered questionnaire. Quantitative data was analysed through descriptive statistics, while qualitative data was analysed through thematic analysis. The results show that there are variety of factors that influence respondents’ attitudes towards copyright infringement of films and/or television series through peer-to-peer file sharing technology, each of which is discussed in detail. The study concluded by identifying 24 factors that favourably influence people’s attitudes towards copyright infringement, including high prices of legitimate goods, historical inequality in South Africa, and perceived low risk of being caught and punished. Based on the conclusion above, the study recommends that policy makers such as government officials, boards of directors, managers, committees, and executives use the results of the study when making decisions and determining policies, especially in the South African context. / Communication Science / M.A. (Communication Science)
