This thesis deals with the sliding contact of electrical machines and its diagnostic methods. Namely, the dust-particles method for assessing the length decrease of brushes is deeply described. Then, usage of the method in industry is described. The comparison of accuracy between manual version and new automated version of the method using computer analysis is presented. The final section is devoted to the practical verification of the new automated version which was developed especially for large machines. Measurements were performed on a small machine.

This master’s thesis deals with the construction design of microscope episcopic LED illumination source and subsequent functional sample realization. Illumination source is compatible with industrial microscope Nikon Eclipse LV150 and designed as a replacement for halogen light source. Way of collecting light is chosen with respect to search analysis results and collector optical element and LED is selected with respect to the analysis of the episcopic illuminator. Two construction designs are designed with respect to the results of photometric analyzes. Optimal design is realized and compared with used lighting sources – halogen and xenon.

The aim of diploma thesis are modifications in mechanical design of the coherence controlled holographic microscope which will lead to reduction of some deficiencies. In this thesis there is a brief description of the history interference microscopy and differences between each types of interference microscopes. In this work there are stated some design deficiencies of actual condition of the microscope and also several solutions are suggested and shortly described. Then the best solution is chosen which is adjusted to a final form. It is described in detail and then realized. There is a summary included in the conclusion whether the new solution is suitable and if it fulfills the required condition.

This diploma thesis deals with phase and amplitude objects observation through scattering media by means of a coherence-controlled holographic microscope (CCHM). A brief history of development and construction of the microscope, its advantages compared to the classical light microscopy and hologram processing are described. Quantitative phase imaging through scattering media by means of ballistic as well as diffuse light is verificated in the experimental part. A comparison of an image obtained through a scattering layer by means of CCHM and a classical microscopy in the light field is demonstrated.

This thesis focuses on cell tracking in image sequences acquired using a multimodal holographic microscope (MHM). The principles of holographic microscopy are described together with the application in cells acquisition. The main part of the thesis describes a complete approach for segmentation and tracking of single cells in acquired in long-term sequences. The approach is designed based on parametric active contour models with specific modifications to achieve reasonable precision and robustness. The implemented method is described in detail, including the evaluation and demonstration of results.

Two-photon laser scanning microscopy is a modern method of in vivo neurophysiological research, capable of imaging up to hundreds of neurons at once. However, this method produces a large amount of data, difficult to process and analyze manually. This thesis presents Two-Photon Processor, a new toolkit for complex processing of data from two-photon microscope. During the work on this thesis, we designed the SeNeCA segmentation algorithm for detection of neurons in full-frame recording from a two-photon microscope. SeNeCA combines high speed and high quality of segmentation and, according to our evaluation, it currently is the best algorithm for segmentation of neurons in in vivo data. Two- Photon Processor is already routinely used in the Institute of Experimental Medicine of the ASCR, Department of Auditory Neuroscience, and it was published in the Journal of Neurophysiology.
