Venkovské brownfields: Šance pro podnikání / Rural Brownfields: An Opportunity for Doing Business

The thesis deals with brownfields in the Podkletí micro municipalities cluster. In the theoretical part, the work focuses on the basic terminology associated with the topic of the thesis, describes the most significant obstacles to the brownfields revitalization and different approaches in addressing this issue in the Czech Republic and in the selected foreign countries. Additionally, an overview of financial instruments to support the revitalization of brownfields, or reconversion, and methods of work are included. The analytical part contains characteristics of all identified brownfield sites in the Podkletí micro municipalities cluster. Based on the brownfield analyses and the given microenvironment, the particular sites are assessed according to their chances of an easy and fast recovery and readiness for the revitalization carried out by the small businesses. All brownfields are also evaluated in terms of their potential for different types of small and medium enterprises.

The aim of this thesis was to study how to increase effectiveness of cancer immunotherapy based on synergy of compounds stimulating phagocytosis and TLR agonist. Tumor microenvironment was modified by enzymes, which catalised conversion of lactate to pyruvate or acetate. It was monitored effect of enzymes on tumor size, survival of experimental mice and cytotoxicity on tumor cells.

To reach this aim, I made own evaluation of the macro environment using STEP analysis, using Porter{\crq}s model I analysed the micro environment, and for the company assessment financial analysis and marketing mix were used. I have made a summary of all my findings and created SWOT analysis, in which I identified strenghts and weaknesses of the company, as well as its opportunities and threats.

Multiple myeloma and its aggressive variant, plasma cell leukemia, are still considered to be incurable diseases despite the progressive treatment approaches comprising novel drugs. This can be attributed to the presence of the bone marrow microenvironment which plays an important role in drug resistance of myeloma cells. Hematopoietic cell lines derived from hematologic malignancies are suitable models for the study of etiopathogenesis of these malignant diseases and for testing new potential drugs. Establishment of these cell lines is still considered to be coincidental and rare event. The first part of the thesis is focused on establishment and characterization of the cell line UHKT-944 derived from a patient with primary plasma cell leukemia, and on completion of characterization of the cell line UHKT-893 derived from a patient with multiple myeloma. Additional analysis of UHKT-893 cell line were performed including sequence analysis of IgVH gene rearrangements and cytogenetic analysis which contributed to more detailed characterization of this cell line. During cultivation of UHKT-944 cells, we monitored the cell growth and confirmed dependence on interleukin-6 (IL-6). Metastasis is the main cause of death from solid cancer. The dissemination of cancer cells from a primary tumour is a very complex process that involves many steps and cells must overcome many obstacles to colonize distant organs. The tumour microenvironment influences the mode and the dynamics of invasion of cancer cells. Cancer cells have the ability to adapt to distinct environmental conditions in order to stay motile. Invasive cancer cells form membrane protrusions called invadopodia that are able to degrade extracellular matrix. The formation of invadopodia by cancer cells is interconnected to the production of matrix metalloproteases (MMPs). Metastasizing tumour cells use MMPs to break through extracellular matrix barriers and migrate in dense matrix. Both invadopodia formation and MMPs secretion is crucial for the degradation of the extracellular matrix. The most important is the membrane bound MMP-14 (MT1-MMP) and soluble MMP-2 and MMP-9. The invasive structures of tumour cells and the proteolytic enzymes in 2D environment is well described. However, a suitable model of localization and transport of MMPs and connection with invadopodia of tumour cells in 3D environment is still lacking. Solid tumors are complex structures comprising besides the cancer cells vasculature, extracellular matrix (ECM), soluble molecules and a plethora of various other cell types. These components form a so-called tumour microenvironment. From the numerous cell types that are part of tumor microenvironment, tumor infiltrating lymphocytes (TILs) play a major role in patient prognosis. Their presence is also of major importance with regard to new biological therapies based on immune checkpoint inhibitors. Crucial role of TILs is also reflected by the new approaches in cancer diagnostics namely by Immunoscore method (currently used in clinical settings). Immunoscore is based on localization and quantification of CD3+ and CD8+ TILs in thin histological sections of tumor tissue. The question remains to which extent the information obtained from 2D slices reflects the situation in tumor microenvironment considering its spatial heterogeneity. The development of new methodological approaches allowing evaluation of histological information in 3D is the key to answer this question. Detekce a charakterizace makrofágů v nádorech virové a nevirové etiologie / Detection and characterization of macrophages in the tumors of viral and non-viral etiology

Head and neck cancers are etiologically associated with smoking and alcohol consumption. Part of these tumors is induced by HPV and their incidence is increasing in the last decade. Patients with virally induced tumors have better prognosis even though they are usually diagnosed with tumors in advanced stage. One of the possible explanations may be better stimulation of the immune system by viral antigens. Macrophages are cells of the innate immune system which belong to professional phagocytes. They are called TAM upon infiltration to the tumor where they represent heterogeneous group of cells. Two main phenotypes are antitumor M1 and protumor M2 macrophages. TAMs are a major component of tumor microenvironment of many types of tumors, one of them are also head and neck cancers. In my thesis I focused on the immunohistochemical detection of M1 and M2 macrophages in the head and neck tumors of viral and non-viral etiology and at the same time RT-qPCR analyses of gene expression of macrophage-associated and/or immunosuppressive genes IDO1, ARG1, CD163, NOS2 a PTGS2 was performed. My data showed that HPV- negative tumors had higher number of M2 macrophages with typical markers CD163, ARG1 and PTGS2. Fibroblasts are stromal cells ubiquitously present in the human body. They often appear in a quiescent state and can become activated in response to tissue remodeling signals. Activated fibroblasts acquire biosynthetic, pro-inflammatory and contractile properties, key functions for wound healing. In addition, the presence of permanently activated fibroblasts is one of the hallmarks of cancer. The purpose of this work is to investigate the differences between newborn and adult fibroblasts and keratinocytes in their implication in scarless wound healing, the origin of cancer associated fibroblasts (CAF)s and the influence of fibroblasts in melanoma invasion. Evidence suggests that wounds heal almost without scar in newborns. To understand the mechanisms that contribute to scarless wound healing we focused on the differences between newborn and adult fibroblasts and keratinocytes, which are cells present in human skin and participating in wound healing process. A comparison of the expression profile between newborn and adult fibroblasts showed differentially regulated genes related to the acute phase of the inflammatory response and ECM organization, traits involved in wound healing.
