A adição de diversas concentrações de PD153035 em suspensões mitocondriais hepáticas diminuiu o controle respiratório de maneira dose-dependente, promoveu a inibição do fenômeno de transição de permeabilidade mitocondrial (TPM) sensível a ciclosporina A, aumentaram também a capacidade em reter o Ca2+ e diminuíram a produção de espécies reativas de oxigênio. Nas mesmas condições experimentais, avaliando suspensões mitocondriais cardíacas de animais controles e tratados com PD153035 (32 mg/Kg, v.o.), nenhum efeito significativo foi observado. Comparando mitocôndrias isoladas de animais controles e tratados com PD153035, submetidos à isquemia/reperfusão, observamos que o controle respiratório do animal tratado manteve-se semelhante ao de organelas controles. Observou-se também que mitocôndrias isoladas de corações de ratos tratados aumentaram mais rapidamente de volume em relação ao controle, quando incubadas em solução hiposmótica contendo íons potássio, o que não ocorreu quando o K+ do meio de reação foi substituído por Na+ ou Li+. Efeito semelhante foi observado quando mitocôndrias isoladas de coração de rato foram incubadas in vitro com PD153035 (10 nM). O aumento de volume mitocondrial induzido por PD153035 in vivo e in vitro foi inibido por ATP e 5-hidroxidecanoato (5HD) (antagonistas de canal mitoKATP). Em mitocôndrias isoladas de ratos tratados com PD153035 observou-se um menor potencial de membrana mitocondrial interna formado pela hidrólise de ATP pela FoF1 ATPase. Em adição, fibras musculares cardíacas obtidas de ratos tratados com PD153035 in vivo apresentaram um leve aumento na respiração basal, o que está de acordo com uma maior permeabilidade mitocondrial à K+. Em células cardíacas (miócitos HL-1) submetidas a um tratamento transitório com cianeto de potássio na ausência de glicose e na presença de PD153035 mantiveram a viabilidade celular; o efeito foi abolido pelo 5HD. Estes resultados indicam que a cardioproteção conferida por este composto pode estar relacionada à ativação de canais de potássio mitocondriais (mitoKATP), pois o 5HD reverteu o efeito causado pelo PD153035 no aumento na permeabilidade mitocondrial cardíaca a K+ e na viabilidade observada nas células HL-1 / Abstract: The action of the aminoquinazoline PD153035 on the mitochondrial functions was investigated. The addition of various PD153035 concentrations (5, 10, 20 and 40 µM) to liver mitochondria decreased the mitochondrial respiratory control, induced mitochondrial permeability transition cyclosporin A-sensitive, increased the mitochondrial calcium uptake capacity and reduced mitochondrial reactive oxygen species production. Under this same experimental conditions, no effect was observed in rat heart mitochondria when the rat was treated or not with PD153035 (32 mg/kg v.o.) No effect was also observed in the mitochondrial respiratory control of controls and PD153035-treated animals submitted to ischemia/reperfusion. A rapid rat heart mitochondrial swelling was observed when the mitochondrial suspensions were previously incubated in a hyposmotic solution containing potassium ions while no effect was observed when all K+ of the medium were replaced by sodium or lithium ions. Similar results were observed in isolated rat heart mitochondria incubated in vitro with PD153035 (10 nM). This mitochondrial swelling induced by PD153035 in vivo and in vitro was inhibited by ATP or 5-hydroxydecanoate, both mitoKATP channel antagonists. A smaller rat liver mitochondrial inner membrane potential was observed after PD153035 treatment and this effect was due to ATP hydrolysis by FoF1 ATPase. In addition, cardiac muscle fibers from PD153035-treated rats presented a slight increase in basal respiration supporting the higher mitochondrial K+ permeability. Cardiac cells (HL-1) treated with PD153035 and submitted to a transitory treatment with potassium cyanide in the absence of glucose maintained the cellular viability that was abolished by 5HD. These results suggest that the cardioprotection conferred by the PD153035 probably was related with the mitoKATP activation, since the mitoKATP channel antagonist 5HD inhibited the PD153035 effect on both cardiac mitochondrial K+ permeability and HL-1 cell viability / Doutorado / Ciencias Biomedicas / Doutor em Ciências Médicas
