Expression of wild type and variants of human apolipoprotein A-I in Pichia pastoris / Expression de type sauvage et des variantes de l’Apolipoprotéine A-I humaine chez Pichia pastoris

Les lipoprotéines de haute densité (High Density Lipoprotein, HDL) permet deréduction de risque de maladies cardio-vasculaires principalement en raison de leurcapacité à éliminer le cholestérol accumulé des artères (via transport inverse ducholestérol). Les effets protecteurs des HDL sont médiés par l'apolipoprotéine AI(ApoA1), qui est le La protéine la plus importante quantitativement du HDL. L’ApoA1favorise l'efflux de cholestérol vers le foie pour l'excrétion. Une augmentation desniveaux plasmatiques de l’ApoA1 est généralement acceptée d'êtrecardioprotecteur, ce qui en fait un potentiel thérapeutique. Deux variantes naturelle(mutants) de l’ApoA1, Milano et Paris, sont caractérisées par une mutationponctuelle unique a permis l'introduction d'un résidu cystéine. Populations avecApoA1-Milano ont été rapportés d'avoir un système cardiovasculaire, même avec defaibles niveaux de plasma de ApoA1 et HDL. Il est donc d'intérêt pour générerrecombinante de type sauvage et des variantes de ApoA1 humaine pour desapplications thérapeutiques potentielles. Dans cette étude, de type sauvagerhApoA1 a été produit chez P. pastoris et purifié par chromatographie en modemixte en une seule étape. Par la suite, un processus intégré a été le développementde la production et la récupération rapide de type sauvage rhApoA1 chez P. pastorispar chromatographie par lit expansée. En outre, les variantes de l'ApoA1, Milano &Paris, ont été générées par mutagenèse dirigée et ont été exprimés chez P. pastoris.Les motifs d’adsorption de rhApoA1-Milano et rhApoA1-Paris ont été comparés àcelle de type sauvage ApoA1 et les différences ont été discutées. / The high-density lipoprotein (HDL) complex helps reduce the risk of cardiovasculardisorders mainly due to its ability to remove accumulated cholesterol from arteriesvia reverse cholesterol transport. These protective effects of HDL are known to bemediated by Apolipoprotein A-I (ApoA1), which is the major protein component ofHDL. ApoA1 is a lipid binding protein and promotes cholesterol efflux fromperipheral tissues to the liver for excretion. An increase in the plasma levels ofApoA1 is generally accepted to be cardioprotective, making it a potentialtherapeutic. Two naturally occuring variants of ApoA1, namely the Milano & Parismutants, are characterised by a single point mutation resulting in the introduction ofa Cysteine residue. Populations with ApoA1-Milano have been reported to have ahealthier cardiovascular system even with low plasma levels of ApoA1/HDL. It ishence of interest to generate recombinant wild type and variants of human ApoA1for potential therapeutic applications. In this study, wild type rhApoA1 was producedin P. pastoris and purified by mixed-mode chromatgraphy in a single step.Subsequently, an integrated process has been development for the production andrapid recovery of wild type rhApoA1 in Pichia pastoris. This has paved way to theestablishment of a scalable integrated process that could be further developed toindustrial levels. In addition, the cysteine variants of ApoA1, Milano & Paris, havebeen generated by site directed mutagenesis and have been successfully expressedin P. pastoris. The binding patterns of rhApoA1-Milano and rhApoA1-Paris have beencompared with that of wild-type ApoA1 and the differences have been discussed.

The diploma thesis deals with the use of event marketing in the communications strategy of OBO Bettermann Praha which operates in business-to-business market in electrical engineering. The first part looks into event marketing from a theoretical point of view. The second part firstly introduces the company OBO Bettermann Praha and its marketing communications. Subsequently is the influence of event marketing on the company's business relations with its customers and business partners explored (currently the main pillar are theater evenings). The survey is conducted by online quantitative questioning of event's participants and qualitative in-depth interview with the director of the company. The survey results are evaluated in the final chapter of the thesis.

Beitrag zur Herstellung langfaserverstärkter Aluminium-Matrix-Verbundwerkstoffe durch Anwendung der Prepregtechnik

In Kenntnis der beschriebenen verfahrenstechnischen Einflussfaktoren auf die Struktur und die Eigenschaften der faserverstärkten MMC geht es darum, ein prepregtechnologisches Verfahren für das Herstellen lang- bzw. endlosfaserverstärkter MMC mit Aluminiummatrix zu erarbeiten und die Machbarkeit im Hinblick auf die Reproduzierbarkeit und das Erreichen der prinzipiell möglichen mechanischen Eigenschaftskennwerte nachzuweisen. Es geht darum, den Einfluss der Prozessparameter auf die Struktur der Prepregs und der verdichteten MMC`s zu erfassen und zu bewerten. In Kenntnis dieser Zusammenhänge sind die qualitätsrelevanten Kennwerte der Werkstoffstruktur denen der mechanischen Eigenschaften gegenüberzustellen. In Weiterführung der o.g. Darstellungen zwischen Struktur- und Eigenschaften der Verbundwerkstoffe geht es darum, die experimentell bestimmten Kennwerte der Festigkeit und des E-Moduls auf Übereinstimmung mit den entsprechenden Korrelationsmodellen (Shear Lag Modell, Grenzwert- und Modellkonzept, EIAS-Methode) zu überprüfen. Ferner geht es darum, die Zusammenhänge zwischen den im Modell definierten idealen Gefügebedingungen mit den realen im Hinblick auf deren Einfluss auf die Eigenschaftskennwerte kritisch zu diskutieren. Aufbauend auf den definierten Zielstellungen kann nach der erfolgten Bewertung aller Untersuchungsergebnisse gezeigt werden, dass die entworfene Technologie zur Herstellung langfaserverstärkter Metallmatrix-Verbundwerkstoffe dazu geeignet ist, Fasern mit geringem Durchmesser und hoher Flexibilität (am Beispiel der hochfesten Kohlenstofffasern vom Typ HTA 5131) mit dem ausgewählten Matrixmetall (am Beispiel der AlSi5-Legierung) zunächst zu Prepregs und weiterhin in verdichtete Verbundstrukturen zusammenzuführen. Das vergleichende Gegenüberstellen von Simulation und Experiment dienen einerseits dem Verifizieren der Prozessmodelle. Die Prozessmodelle stellen andererseits die wertvolle versuchstechnische Grundlage zur Definition und auch der Einengung des Arbeitsfensters für die Prozessparameter und der Anzahl erforderlicher Verifikationsexperimente im Hinblick auf die Probebeschichtungen und deren Auswertung dar. Derartige Modelle leisten eine wichtige Hilfestellung zum weiteren Erhöhen der Prozessstabilität und damit auch dem Nachweis der Reproduzierbarkeit. / The aim of this work is a described prepreg-technological method to create aluminium based MMC reinforced with continuous fibres and the verification of reproducibility to achieve relevant mechanical properties. This aim is based on the knowledge of technological influences on structure and properties of fibre reinforced MMC. And so activities are focussed on the evaluation of the influence of process functions on structure and mechanical properties of prepregs and compacted MMC. The comparison between structure and properties is necessary to describe the correlation function of composite material. Furthermore the application of different correlation models (“Shear Lag”, “Grenzwert- Modellkonzept“, “EIAS”) to describe the influence of composite structures on strength and Young`s modulus is necessary to compare theoretical results with those of relevant experiments. The object is a critical quantification of the influence of real structure parameters compared with those of a model defined structure. In view of the described aim it is shown that the described technology to manufacture long fibre reinforced MMC is applicable for preparation of carbon fibres (HTA 5131) with low diameter and high flexibility and metal matrix (AlSi5) to prepregs and compact composites. The comparison of simulated and experimental results is the base for verification of different process models. So it is possible to describe and optimize the process function and moreover to minimize the number of technological experiments. After optimization specified models are a good base to achieve a high level of stability and reproducibility for all steps in prepreg technology.

Em se tratando das considerações finais observa-se que as jovens da primeira década do segundo milênio querem viver diferentes de seus pais e suas mães, muitas delas, não aceitam passivamente viver num mundo de privacidade, estando no foco de suas preocupações o acesso à terra, à renda e o estudo na perspectiva de construção de "uma vida melhor". / The MMC/SC Peasant Women's Movement in the state of Santa Catarina is a women's movement that, since 2004, has become national in character and has increased its presence in various Brazilian states. Allied to Via Campesina, the movement's principle struggles are for rights, equality and the Strengthening of family agricuhureThe focus of this thesis is directly linked to this construction process, especially related to the axes of gender and class, whose experience has made possible the emergence of a new social political subject: young rural women. To this end, I apply Eduard Thompson's theoretical references - bringing to the forefront the category "experience" - while using, as my baseline methodology, oral history and ethnography. In conclusion, this thesis has found that the young women of the second millennium's first decade want to live differently to their fathers and mothers; many of them don't passively accept their living conditions. Rather they struggle for access to land, to income and to education with the aim of constructing "a better life".

The advantages of this topology are related to its modularity and scalability. This work presents the study and implementation of this converter, which includes the presentation of the main methods of modulation and voltage balancing of the foating capacitors and startup. The used modulation in modeled using switching functions, its allow one minimize the current ripple at system inductor due the correct selection of carriers shift angles. Moreover a current control and voltages equalization methodology are proposed. It is performed dynamic modeling and quantitative analysis of the converter and it is derived a design methodology. This methodology is used to design and build a 3 kVA prototype with bus voltage of 800 V. The results include transient analyses, efficiency, voltage charging and steady state. / O conversor modular multinível emergiu como uma nova topologia de conversores mutiníveis, sendo introduzido a partir de 2002. As vantagens desta topologia estão relacionadas a sua modularidade e escalabilidade. Este trabalho apresenta o estudo e implementação deste conversor, o que inclui a apresentação das principais metodologias de modulação e equilíbrio da tensão e pre-carga dos capacitores flutuantes. Apresenta-se um estudo da modulação por meio de funções de chaveamento que permite a minimização da ondulação de corrente nos indutores por meio da escolha adequada dos ângulos de defasagem das portadoras empregadas. Para que o projeto da estrutura seja possível, e realizada a modelagem dinâmica e a analise quantitativa do conversor em diferentes condições de operação, sendo derivada uma metodologia de projeto. Esta metodologia de posta a prova com a construção de um protótipo de 3 kVA com tensão de barramento de 800 V. Os resultados obtidos do protótipo incluem avaliações transitórias, verificação do rendimento, pre-carga e operação em regime.

Mise en forme à chaud de tôles fines en alliage AA 5383 : Approches expérimentales et numériques / Forming of deep-parts in AA5383 alloy : experimental and numerical approach

Les alliages d'aluminium ont été largement utilisés dans l'industrie automobile et maritimes en raison des avantages d'une faible densité, d'une bonne résistance à la corrosion. Les travaux présentés dans ce mémoire de thèse s’intéressent à la mise en forme à chaud de tôles minces en alliage d’aluminium AA5383. L'objectif principal est de réduire le temps de formage sans sacrifier l'intégrité de la pièce. Tout d'abord, le comportement à la déformation à chaud de l'alliage AA5383 est caractérisé expérimentalement. Une campagne expérimentale comprenant d’essais de traction uniaxiale, de traction entaillées, de cisaillement et de gonflement libre est réalisée pour couvrir une plage importante de températures (623~723 K) et de vitesses de déformation (10-4~10-1 s-1). Ensuite, les modèles de matériau, tels qu'une règle de flux composite avec le critère de plasticité BBC2003 et le critère de dommage Mohr Coulomb Modifié, sont développés et mis en œuvre dans ABAQUS à l'aide du sous-programme utilisateur. Enfin, les simulations numériques des processus de formation de gaz sont effectuées et comparées aux résultats expérimentaux correspondants. / Aluminum alloys have been extensively used in the automotive and marine industry due to the advantages of low density, high strength to weight ratio and good corrosion resistance. Major challenge of their application lies in the ability to form deep-drawing shapes. Superplastic Forming is widely used to produce this type of parts. However, high forming cycle time due to the low forming strain rate limits their wide application. The present dissertation focuses on hot forming strategies to produce deep drawing parts from AA5383 aluminum thin sheets. The main objective is to reduce the forming time without sacrificing the part integrity. Firstly, the hot deformation behavior of the AA5383 alloy is experimentally characterized. An experimental campaign, including uniaxial tension, notched tension, shear and free bulging tests, is performed to cover an important range of temperatures (623~723 K) and strain rates (10-4~10-1 s-1). Then, the material models, such as a composite flow rule with the BBC2003 anisotropic yield criterion and the modified Mohr-Coulomb damage criterion, are developed and implemented in ABAQUS by using user subroutine. Finally, the numerical simulations of the gas forming processes are performed and compared with the corresponding experimental results.

The main topic of the thesis is to suggest and realize the measuring card with analog inputs and outputs. In the first part of this thesis we are concerned with key-elements of measuring cards, respectively ADC and DAC, and here are explained all their activities. Then we attend of choosing parts for suggested measuring card as is voltage reference, power supply and operational amplifier. Among others we are concerned with a question of protecting inputs against overvoltage and reversing of polarity and beside against disturbance. Then follows analysis of used microcontroller ADUC831 with description of its used outskirts. Here is described the stored for measured samples of MMC memory, how to work with it and the programme service. The last chapter describes suggested firmware and servicing programme for PC. In fine we value the quality of measuring card and its mistakes.

Efeitos da soldagem por laser de fibra de Yb na microestrutura do compósito A356/SiCp / Effects of laser welding by Yb fiber in the microstructure of A356/SiCp composite

Materiais compósitos são projetados e fabricados para várias aplicações de alto desempenho, incluindo componentes para os segmentos automobilístico, aeroespacial, aeronáutico, naval, de defesa, de óleo e gás, energia eólica e até equipamentos esportivos. Porém, a união por soldagem de Compósitos de Matriz Metálica de Alumínio (Al-CMM) ainda é um grande obstáculo para a maior disseminação desta classe de materiais estruturais. As mudanças microestruturais decorrentes do ciclo de soldagem e/ou do tratamento térmico afetam sensivelmente as propriedades mecânicas e físico-químicas finais da junta e do metal base nas proximidades de mesma, daí a importância de se estudar a evolução microestrutural que prospera nestas etapas. O presente trabalho caracterizou a microestrutura do compósito liga-A356/SiCp soldado por laser de fibra de Itérbio, empregando-se nessa tarefa técnicas de microscopia óptica, radiografia e microscopia eletrônica de varredura, assim como difração de raios-X e de elétrons retroespalhados, ensaio instrumentado de dureza e microtomografia computadorizada. O foco das análises realizadas restringiu-se à geometria dos cordões de solda, à expulsão de SiC particulado da zona soldada, à volatilização de elementos químicos da poça de soldagem, à formação de precipitados fragilizantes de Al4SiC4 em formato de agulhas no cordão de solda e à determinação das regiões com concentração de poros, todos estes fenômenos tendo efeitos nocivos, em maior ou menor extensão, no desempenho global da junta do Al-CMM soldada a laser, notadamente em suas propriedades mecânicas e eletroquímicas. / Microstructural materials composites are designed and manufactured for various high performance applications, including components for different industries like automobile, aerospace, aeronautical, naval, defense, oil and gas, wind energy and even sports equipment. However, the junction by welding of Aluminum Metal Matrix Composite (Al-MMC) is still a major obstacle to the further spread of this class of structural materials. The microstructural changes resulting from the welding cycle and / or post-weld heat treatment significantly affect the final mechanical and physicochemical properties of the joint and the base metal near it, hence the importance of studying the microstructural evolution that thrives in these steps. This study aims to characterize the microstructure of the composite alloy-A356 / SiCp welding by Ytterbium fiber laser, using optical microscopy, X-ray, scanning electron microscopy, X-ray diffraction, electron backscatter diffraction, instrumented test hardness and computed microtomography. The focus of the analyzes was restricted to the geometry of the weld bead, the particulate SiC expulsion of the welded zone, volatilization of chemical elements from the welding zone, formation of precipitates embrittlement of Al4SiC4 in needle shape in the weld bead and determining the concentration of regions with pores, all these phenomena have harmful effects to a greater or lesser extent, the overall performance of the joining Al-CMM laser welded, mainly in their mechanical and electrochemical properties.

Caracterização de material composto de matriz metálica a partir de um liga de alumínio aeronáutico / Characterization of metal matrix composite material from an aeronautical aluminum recycled alloy

Materiais compostos de matriz metálica (CMM) vêm sendo estudados para diversas aplicações. Entretanto, pouco estudo é apresentado na sua confecção a partir de materiais reciclados. Este trabalho teve como objetivo a caracterização de um material composto de matriz metálica (CMM), utilizando material reciclado como matéria prima. Foram usados aparas de chapas de liga de alumínio 2024 descartados para a matriz e carbeto de silício (SiC) como reforço, sendo a produção realizada através do processo de fabricação com base na técnica de conformação por spray para possível aplicação em componentes automotivos. Foi realizado o tratamento térmico do material e a caracterização, determinando as propriedades de dureza, resistência mecânica, resistência ao desgaste, MEV e EDS. Foram ensaiadas amostras do material composto fundido e extrudado, bem como de ferro fundido de uma peça automotiva e da liga de alumínio 2024. Observou-se que o CMM ainda precisa de melhorias no processo de produção para obter propriedades de dureza e resistência que permitam que ele possa ser usado como um substituto para o ferro fundido, porém o mesmo apresentou melhores propriedades quando comparado com o material da matriz. / Metal matrix composites (MMC) have been studied for several applications. However, little study is presented in its manufacture from recycled materials. This study aimed to characterize a metal matrix composite (MMC), using recycled material as raw material. Were used aluminum alloy 2024 plates clippings discarded for the matrix and silicon carbide (SiC) as reinforcement, being the production accomplished through the manufacturing process based on the technique of spray forming for possible application in automotive components. The thermal treatment and the characterization of the materia was accomplished, determining the properties of hardness, mechanical strength, wear resistance, SEM and EDS. Were tested samples of the cast and extruded composite material, as well as cast iron of an automotive part and aluminum alloy 2024. It was observed that the MMC still needs improvements in the production process to obtain properties of hardness and strength that allows it to be used as a substitute forcast iron, but it presented better properties when compared with the matrix material.
