An Analysis Of Deformation Behavior Of Muratli Asphalt Faced Rockfill Dam

Unsever, Yesim Sema 01 July 2007 (has links) (PDF)
In this study, settlement and seepage behavior of Muratli Dam, which is the first asphalt faced rockfill dam in Turkey, is investigated for the &ldquo / end of construction&rdquo / and &ldquo / reservoir impoundment&rdquo / loading conditions. Two dimensional plane strain finite element analyses are carried out in order to determine the total stresses, displacements and pore water pressures. Hardening soil model is used in order to represent the non-linear, inelastic and stress dependent behavior of rockfill material. Material model parameters are selected mainly referring to the previous studies on the dams consisting of similar materials and then back analyses are done to find the best fit. Calculated stresses, displacements and pore water pressures are compared with the observed values for both end of construction and reservoir filling conditions.

High Hydrostatic Pressure Induced Inactivation Kinetics Of E. Coli O157:h7 And S. Aureus In Carrot Juice And Analysis Of Cell Volume Change

Pilavtepe, Mutlu 01 December 2007 (has links) (PDF)
The main objective of this study was to determine the pressure induced inactivation mechanism of pressure-resistant Escherichia coli O157:H7 933 and Staphylococcus aureus 485 in a low acid food. Firstly, inactivation curves of pathogens were obtained at 200 to 400 MPa at 40&ordm / C in peptone water and carrot juice. First-order and Weibull models were fitted and Weibull model described the inactivation curves of both pathogens more accurately than first-order model, revealing that food systems could exhibit either protective or sensitizing effect on microorganisms. Carrot juice had a protective effect on E. coli O157:H7 whereas it had a sensitizing effect on S. aureus, due to the naturally occurring constituents or phytoalexins in carrot roots that could have a toxic effect. Secondly, scanning electron microscopy (SEM) and fluorescent microscopy images of studied pathogens were taken. Developed software was used to analyze SEM images to calculate the change in the view area and volume of cells. Membrane integrity of pressurized cells was also examined using fluorescent microscopy images. The increase in average values of the view area and volume of both pathogens was significant for the highest pressure levels studied. The increase in volume and the view area could be explained by the modification of membrane properties, i.e., disruption or increase in permeability, lack of membrane integrity, denaturation of membrane-bound proteins and pressure-induced phase transition of membrane lipid bilayer. The change in volume and the view area of microorganisms added another dimension to the understanding of inactivation mechanisms of microbial cells by HHP.

Dimensions and Operationalisations of IT Governance: A Literature Review and Meta-Case Study

Novotny, Alexander, Bernroider, Edward, Koch, Stefan January 2012 (has links) (PDF)
This paper seeks to tackle the current confusion about the constituent dimensions of IT Governance (ITG) and inconsistent operationalisation approaches inhibiting advances in research and organisational ITG practice. Through a structured literature review of ranked high-quality publications augmented by a meta-case study with five underlying projects, we find nine distinct dimensions of ITG. The input-oriented dimensions Compliance Management, IT Investment Management and ITG Improvement have received little attention in earlier conceptualisations, while the more output-oriented dimensions Business/IT Alignment and Business Value Delivery have featured more often in related studies. Scope and application of ITG may depend on the organisational context and the intentional use, such as regulatory or strategic. Depending on the context, more research seems to be warranted to develop context-dependent measurement constructs of ITG that can be compared over studies. (author's abstract)

Protein dynamics: a study of the model-free analysis of NMR relaxation data

d'Auvergne, Edward J. Unknown Date (has links) (PDF)
The model-free analysis of NMR relaxation data, which is widely used for the study of protein dynamics, consists of the separation of the Brownian rotational diffusion from internal motions relative to the diffusion frame and the description of these internal motions by amplitude and timescale. Through parametric restriction and the addition of the Rex parameter a number of model-free models can be constructed. The model-free problem is often solved by initially estimating the diffusion tensor. The model-free models are then optimised and the best model is selected. Finally, the global model of all diffusion and model-free parameters is optimised. These steps are repeated until convergence. This thesis will investigate all aspects of the model-free data analysis chain. (For complete abstract open document)

Eseje o přírodním Zdroji Bohatství, Hospodářském Růstu a Institucionální Kvalitě / Essays on Natural Resource Richness, Economic Growth and Institutional Quality

Zeynalov, Ayaz January 2017 (has links)
Charles University in Prague Faculty of Social Sciences Institute of Economic Studies DISSERTATION THESIS ABSTRACT Essays on Natural Resource Richness, Economic Growth and Institutional Quality Author: Ayaz Zeynalov Supervisor: Doc. Roman Horvath, Ph.D Academic Year: 2016/2017 Abstract This dissertation consists of three empirical papers on natural resource, economic growth and institutional quality. The first paper analyzes possible publication bias and the reason for contradictory findings in the natural resource literature, the second paper examines the effect of natural resource exports on manufacturing performance in the 15 former Soviet Union countries, and the last addresses whether similarities in country income size and at the institutional level encourage increased amounts of bilateral trade between countries. An introductory chapter puts these three papers into perspective. In the first paper, I analyze 43 studies providing 605 different regression specifica- tions and found that approximately 40% report a negative and statistically significant effect, another 40% report no effect, and the remaining 20% report a positive and statistically significant effect of natural resources on economic growth. The findings show that including interaction between natural resources and institutional quality, controlling...

Quantitative tool for in vivo analysis of DNA-binding proteins using High Resolution Sequencing Data

Filatenkova, Milana S. January 2016 (has links)
DNA-binding proteins (DBPs) such as repair proteins, DNA polymerases, re- combinases, transcription factors, etc. manifest diverse stochastic behaviours dependent on physiological conditions inside the cell. Now that multiple independent in vitro studies have extensively characterised different aspects of the biochemistry of DBPs, computational and mathematical tools that would be able to integrate this information into a coherent framework are in huge demand, especially when attempting a transition to in vivo characterisation of these systems. ChIP-Seq is the method commonly used to study DBPs in vivo. This method generates high resolution sequencing data { population scale readout of the activity of DBPs on the DNA. The mathematical tools available for the analysis of this type of data are at the moment very restrictive in their ability to extract mechanistic and quantitative details on the activity of DBPs. The main trouble that researchers experience when analysing such population scale sequencing data is effectively disentangling complexity in these data, since the observed output often combines diverse outcomes of multiple unsynchronised processes reflecting biomolecular variability. Although being a static snapshot ChIP-Seq can be effectively utilised as a readout for the dynamics of DBPs in vivo. This thesis features a new approach to ChIP-Seq analysis { namely accessing the concealed details of the dynamic behaviour of DBPs on DNA using probabilistic modelling, statistical inference and numerical optimisation. In order to achieve this I propose to integrate previously acquired assumptions about the behaviour of DBPs into a Markov- Chain model which would allow to take into account their intrinsic stochasticity. By incorporating this model into a statistical model of data acquisition, the experimentally observed output can be simulated and then compared to in vivo data to reverse engineer the stochastic activity of DBPs on the DNA. Conventional tools normally employ simple empirical models where the parameters have no link with the mechanistic reality of the process under scrutiny. This thesis marks the transition from qualitative analysis to mechanistic modelling in an attempt to make the most of the high resolution sequencing data. It is also worth noting that from a computer science point of view DBPs are of great interest since they are able to perform stochastic computation on DNA by responding in a probabilistic manner to the patterns encoded in the DNA. The theoretical framework proposed here allows to quantitatively characterise complex responses of these molecular machines to the sequence features.
