Využití rychle reagujícího SBS polymeru při výrobě asfaltových směsí / Usage of fast-reacting SBS polymer in the production of asphalt mixtures

The subject of this diploma thesis is a research of bituminous binder modification by fast react-ing polymer SBS. The theoretical part covers an introduction to bituminous binders, their pro-duction, chemical and physical-mechanical properties. The identification of bituminous binders and their use in bituminous mixtures is briefly described. To conclude, current modification methods are described in detail and important modifiers are introduced. In the practical part, three applied tests (softening point, elastic recovery and penetration) are described in detail. The bituminous binders tested (20/30, 30/45, 70/100, PMB 25/55-55 and PMB 45/80-50) and the fast-reacting polymer SBS are introduced. Samples with two weight percentages of SBS, 7,5% and 10%, are produced. Two mixing times, 60 seconds and 600 seconds were used to mix the bituminous binder and SBS. In conclusion, the results are evaluated and commented. Further research suggestions regarding this field are recommended.

The diploma thesis deals with the use of recycled polymers in asphalt mixtures. In the theoretical part of the thesis there is discussed the global issue of plastic waste including the experience of foreign countries, where this technology has already been introducted and used. The practical part is devoted to the proposal of using recycled plastic in asphalt mixture, description of used laboratory tests and the evaluating the effect of low-density polyethylen (LDPE) on the asphalt mixture type AC. There were performed tensile strenght tests, water resistant tests and stiffness modulus tests in the laboratory (E). The output of this work is an evaluation of measured values, processing of results and their interpretation depending on the possible use of this technology in Czech republic.

Topic: Analysis of game in girls minibasketball from the perspective of interruptions by referees. Aim: The aim in this thesis is to make a analysis in one season in girls minibasketbal games in all categories to find out how often and why referees are interrupting the game. Than to make a statistic of all games and compare categories between themselves. Primary aim is to make the analysis which will show characteristic of interrupting the game and how often they are happening in game. Methods: Taking into consideration quantitative survey, my dissertation work was based on methods of secondary information collecting that implies secondary statistical analysis and observation with taking records down. Results: In average there are 98,14 interrupting in game of minibasketbal from referee. The travelling and ball outside of playcourt are the most often violations. There are differences between categories. Stanovení butylhydroxyanisolu na elektrodách modifikovaných uhlíkovými nanotrubičkami / Determination of butylhydroxyanisole using electrodes modified by carbon nanotubes

This work was focused on study of a behaviour of the food additivum butylated hydroxyanisole on modified carbon electrodes by the voltammetric techniques - cyclic and differential pulse voltammetry. Glassy carbon and carbon paste electrode were used. Multiwalled carbon nanotubes (MWCNT) in combination with three different binders (acetonitrile, nafione or chitosane) were employed for the electrode modification. Carbon paste electrode was unable to modificate with film containing carbon nanotubes and acetonitrile, its active surface was treated only with nafione and chitosane film. All three mentioned modifications were applied in case of glassy carbon electrode. Butylated hydroxyanisole provided a significantly higher signal using electrodes modified with carbon nanotubes with all three binders in contrast to electrodes without any surface modification. The glassy carbon electrode with carbon nanotube / acetonitrile film on its surface appeared to be the most effective for analytical purposes. Voltammetric determination of butylated hydroxyanisole using this electrode provided a better defined and higher analytical signal and lower relative standard deviations in comparison with other ways of modification. Wnt signalling represents an important mechanism participating in control of cellular and developmental processes, including establishment of cell polarity, cell fate specification, stem cell self-renewal, tissue patterning and organogenesis, homeostasis maintenance and regeneration. Misregulation of the Wnt signalling during embryogenesis leads to developmental defects while aberrant activation later in development is associated with degenerative diseases and a number of cancers. The presented PhD thesis is based on four original publications that deal with the post-translational modifications of Wnt ligands and molecular mechanisms contributing to the regulation of a transcriptional profile of the so-called canonical Wnt pathway. Wnt signalling pathway is used repetitively both in time and different cellular contexts throughout development of multicellular organisms. Inevitably, in each single situation -catenin/TCF complexes, the downstream effectors, induce only subsets of all potential target genes. How this differential tissue- and stage-specific control over various subsets of target genes is achieved with such a limited number of nuclear effectors is not fully understood. Signální dráha Wnt v obnově a tumorigenezi střevního epitelu / Wnt signaling in intestinal homeostasis and tumorigenesis

The canonical Wnt signaling pathway is one of the most important pathways involved in cell proliferation and differentiation. It is highly conserved in evolution and participates not only in embryonic development but also in adult tissue homeostasis. In the intestine, Wnt signaling is closely connected to maintenance of intestinal stem cells and renewal of the epithelia. Conversely, aberrant activation of the Wnt signaling pathway underlies different types of human diseases. Its constitutive activation results in neoplasia and specifically in development of colorectal cancer, which is the third most common malignancy in western world. The aim of this thesis was to uncover various aspects of the regulatory mechanisms of the Wnt/β-catenin signaling cascade. Furthermore, I headed to find novel Wnt pathway modulators and confirm their function in vivo. The results are presented in four publications. The first study examines murine Wnt proteins processing and the sequential order of Wnt post-translational modifications which are required for the secretion and signaling activity of the ligands. Next publication focuses on the gene Troy, which we identified as negative regulator of Wnt signaling. Magnetické biokompozitní materiály pro odstranění významných xenobiotik z vodních systémů / Magnetic biocomposite materials for removal of significant xenobiotics from water systems

The theoretical part of this doctoral thesis provides a comprehensive overview on the topic of preparation and subsequent utilization of magnetic derivatives of biological materials for xenobiotic separation from water. Main attention is paid to magnetic modification of waste materials and by-products originating from agricultural and food industry, which represent widely available and low-cost materials, and also to magnetic modification of microbial cells. In addition to the description of magnetic particle preparation and individual developed techniques of magnetic modification, a brief characterization of selected pollutants and a detailed table overview on utilization of magnetically responsive biomaterials for biosorption of organic dyes, heavy metals, pharmaceutical and personal care products together with ubiquitous industrial endocrine disruptors and also of crude oil derivatives is presented. Experimental part of this thesis is focused on the preparation and optimization of new types of magnetic materials. Emphasis is placed on the employment of simple, fast and simultaneously low-cost magnetic modification techniques (e.g., postmagnetization using microwave-synthesized magnetic iron oxides or one-step modification by magnetic fluids). Selected plant materials (barley and rye straw) were chemically modified to significantly (up to five-times) increase the maximum adsorption capacities for tested dyes. All prepared biomaterials exhibited a great magnetic response and simultaneously relatively high adsorption capacity for selected xenobiotics under experimental conditions used. Factors substantially affecting adsorption process, such as pH, initial concentration, incubation time or temperature were also studied. Adsorption equilibrium data were assessed using Langmuir, Freundlich or Sips isotherm models. Experimental data from time dependence study were analyzed by chosen kinetic models, namely the pseudo-first-order and pseudo-second-order ones and by intraparticle diffusion model. Thermodynamic parameters (Gibbs free energy, enthalpy and entropy) describing the nature of adsorption were also included in study.

There are also topics have focused on specific learning disabilities, their social aspects, and artistic expression of children with this diagnosis and the graphic skills of children in general. Followed by part dealing with the colors, their use in psychodiagnostic and their symbolism. This section was prepared analyzing literature and its aim is characteristic Wartegg drawing test and color modifications.In the empirical part of the selected characters analyzed in terms of processing individual fields Wartegg drawing test, which are then processed statistically.
