Sledování tvorby plovoucího biofilmu Mycobacterium smegmatis - morfologická a proteomová analýza / Monitoring of Mycobacterium smegmatis floating biofilm development - morphological and proteome analysis

Microorganisms grow in planktonic form, but more often they adhere to a number of surfaces and create three-dimensional structures called biofilms. Floating biofilms, which are formed at the water-air interface, are one of the life strategies, which the bacteria can take. Non-pathogenic Mycobacterium smegmatis was used as a laboratory model for the study of this kind of biofilm. The understanding of mechanisms of their formation of this species may be applicable to the pathogenic species of the genus Mycobacterium, study of which in the laboratory brings a number of disadvantages. This diploma thesis focuses on the morphological and proteome analysis of the M. smegmatis floating biofilm. Using a stereo microscope and scanning electron microscopy was observed that bacteria clump and create the "nucleation centres" at the beginning of the biofilm development. This centers grow to the surroundings and connect afterwards. In the later stages of the development the centers fuse in compact layer, which then grows into the compact and multilayer biofilm. The key method in this study was two-dimensional electrophoresis of proteins. The proteome analysis of floating biofilm was performed with this method. The aim of this thesis is to analyze the methods used by translators when translating spoken language in contemporary Danish fiction into Czech. In the first chapter, the Czech translation tradition is defined. This tradition influences Czech usage and tendencies. The second chapter introduces the features of stylized language reflected in literature written in Czech. As will be shown, these are mainly lexicological and morphological features and related stylistical means. The following chapters are devoted to the analysis of individual features of stylized language and their translation into Czech. Discussed are loanwords, diminutives, expletives, morphological changes, realia and politeness. The translated units are compared with Czech translation tradition and literature written in Czech.

Jazykový rozbor vybraných památek tzv. Pinvičkova sborníku / Language analysis of the selected texts of the Pinvicka collection

This thesis deals with the language analysis of the Tandarias poem published as a part of Pinvicka collection which was written in the 2nd half of 15th century by Jan Pinvicka z Domazlic. The attention is paid to the description of ortography and all language dimensions. The main aim of this thesis is to describe the language statement of the relic and evaluate the influence of the previous versions of theTandarias poem on the examined version published in Pinvicka collection. We found out that it is not possible to give an evidence that the examined poem is significantly influenced by the language usus of the 14th century when this subject was translated to Czech. But we claim that some of the language phenomena refering to older language usus can be explained with the personal language manner of author. Nevertheless the influence of the original German writing is obvious in the lexical dimension. The addressing of the writing to the bourgeoisie and other lower social classes is supported with the tendency of the language of the text to approach to the spoken form of language.

Vliv představitelnosti slov na osvojování slovních tvarů v češtině / Effects of Word Imageability on the Acquisition of Grammatical Morphology in Czech Children

(in English): In many studies there was demonstrated that word's imageability - the ability of a word to produce a mental image of it's referent - affects the processing of its inflection (e.g. Prado - Ullman, 2009), its acquisition (e.g. McDonough et al., 2011) and the acquisition of grammatical morphemes (Smolík, in press). This thesis builds on the Smolík's paper, and its goal is to test whether the imageability of a word's stem influences the acquisition of its inflections in Czech children. Word imageability ratings and ratings of other variables that were assumed to affect the process of the acquisition of word inflections, were collected, then questionnaires were distributed to parents to examine how the forms of chosen words are acquired by children. The focus was on the acquisition of nominative plural of nouns, present tense in second person and past tense of verbs. The results show that the imageability significantly predicts the age of acquisition of the nominative in singular and all observed forms of verbs. The absence of the imegability effect on the acquisition of nominative in plural may be due to the overall high imageability and thus reduced variability of ratings in nouns in our dataset. A b s t r a c t This thesis is based on two projects which follow one another and which deal with study of development of language and literacy skills in children at risk of dyslexia. The thesis describes the development of morphological awareness in children with specific language impairment. The level of morphological awareness was repeatedly tested during three years at children in preschool and early school age with the Test of morphological awareness. This method was prepared on the basis of Žlab's Test of language awareness (1992) and maps development of several subareas of morphological skills - gender forms, plural nouns, past tense and third person singular verb, adjective derivation and gramatical agreement. Several issues were concerned. First, whether the development of children in this age group is still ongoing and whether the Test of morphological awareness can measure this ongoing. Second, whether there are differences in the performance of children from normal speech development group (control group) and specific language impairment children group, where according to the disorder the morphological deficits can be expected. This final thesis works with the topic of standard Czech language at the 1st level of primary schools. The theoretical part deals with the units of the Czech national language, standard Czech language and its development, current phenomenons in contemporary Czech, common Czech language, Czech dialects and sociolects. Furthermore, it briefly describes the development of teaching of standard Czech language at schools and other possibilities of teachers. It also analyzes particular morphological aspects of individual morphological changes subsequently examined in the practical part. The empirical part is a quantitative analysis and comparison of the ability of pupils of the first grade of selected schools in the Central Bohemian Region to identify the non-standard expressions and replace them with the correct standard forms. The work partly verifies the selected hypotheses and compares the results primarily with the results of similar research carried out at the 2nd level of primary schools.

The thesis aims to describe and theoretically explain the range of complex nominal word structures and phrases that evolved in all Slavic languages with staggering productivity during the last decades. In particular, it focuses on the so called analytical adjectives, i.e. formally invariant lexical units mostly of foreign origin that manifest their attributive syntactic role (and the word class they belong to) not through a morpheme but through fixed word order position. The thesis rejects the view that analytical adjectives are first parts of compound words and explores several transitional cases. In more general terms, the thesis explores phenomena that test the boundary between words and phrases, morphology and syntax. It concludes that in Bulgarian there already exists a well established word subclass of analytical adjectives. They have resulted from an originally lexical innovation that has been grammatcalized due to intensive borrowings of words and phrase models mainly from English and their replication. The comparative study of present-day Bulgarian and Czech provides evidence of the strategies that the two genetically related languages use to achieve their nomination goals. The linguistic analysis of The Apostle of Skopje Štefan Pilát The Apostle of Skopje (Skop) is a Macedonian Church Slavonic manuscript, which contains a short Praxapostle. The scribal inscription on the last page places its compilation to the city of Skopje in the year 1313 thus allowing for its inclusion among the north-Macedonian texts. The goal of the present thesis was to evaluate its paleographic, orthographic and phonological characteristics and its morphological and syntactic peculiarities. The established facts were then compared with other north-Macedonian manuscripts and writing traditions of the Ochrid, Preslav, Tarnovo and Raška schools. Another aim was to evaluate how much could the reception of individual scribal norms be impacted by the north-Macedonian dialect and identify the dialect elements of Skop. On this account, I conclude that Skop, while belonging to the circle of north-Macedonian manuscripts and reflecting rather evident influence of the local dialect, remains conservative, especially from the perspective of paleography, orthography and phonology, a feature typical of the older tradition of the Ochrid School. This dissertation aims to identify and analyse the frequency distribution of mor- phological forms of nouns in contemporary Czech. The work is based on an error analysis of texts of non-native speakers revealing and describing frequent types of errors in noun inflection. Against the background of recent research in the field of second language acquisition on the one hand and the principles of communicative approaches to language teaching on the other hand nine textbooks of Czech as a foreign language are analysed for approaches to the introduction and presentation of grammatical cases, their functions and typical constructions. It is concluded that it is necessary to exemplify case functions by prototypical constructions and sufficient sample nouns for which these functions are typical. With regard to the often observed emphasis on the noun as a part of the inflectional paradigm and thus on the nominative singular as its basic form, it is eligible for textbooks and language classes to shift the focus from the dominant paradigmatic to the syntag- matic aspect of nouns and their word forms. Master thesis deals with changes of elements, that constitute a fairy tale of the Cinderella in different times, cultural and media contexts. Therefore, thesis analyzes and compares different literal and film renderings of the story based upon morfological composition of the fairy tale and motives and gender stereotypes present in it. Individual features of tales are then processed and divided on archetypal and changeable. Main character, the Cinderella , is also considered, and her common characteristic is documented within gender analysis for every version of the story. Thesis also deals with fairy tale in general, with its definition, characteristic and theory of origin. Fairy tale is then interpreted as a part of folklore creation and as a literary work.
