Effect of phytogenic feed additives on gonadal development in Mozambique tilapia

Thesis (PhD)--Stellenbosch University, 2013. / ENGLISH ABSTRACT: This study investigated the “Effect of Phytogenic Feed Additives on Gonadal Development in Mozambique tilapia (Oreochromis mossambicus). Aquaculture remains the fastest growing animal food-producing sector and it is set to overtake capture fisheries as a source of food fish, and also to outpace population growth. This rapid growth can be attributed to the increasing demand for aquaculture products, and an urgent need for a sustainable food resource and the safe production of food. Globally, fish provides over 3.5 billion people with up to 15 percent of their average per capita intake of animal protein. The introduction of aquaculture to sub-Saharan African (SSA) took place during the 1940s and 1950s, and the main objectives were to improve nutrition in rural areas, allow for the diversification of activities to reduce the risk of crop failures, to generate additional income, and to create employment opportunities. Aquaculture was seen as a viable option for rural development in SSA, and substantial resources were invested to support its development. Poor results were however, recorded in terms of production and sustainability. Tilapia is the most widely cultured of all fish species farmed, and the second most important group of freshwater and brackish water fish after carps. Tilapia has all the necessary traits that makes it an excellent species to culture, but costefficient production is hampered by the animals attaining sexual maturity at a an early age, which then result in precocious breeding in aquaculture systems. The production of single-sex populations (i.e. all male) is a potential means to address the problem of precocious breeding, but the technologies used to establish single-sex populations are not readily available to resource-poor communities that farm with tilapia for food purposes. Phytochemicals, also known as phytoestrogens, are plant-derived compounds that structurally or functionally mimic mammalian estrogens that affect the sexual differentiation of fish. Phytochemicals occur in plants like Pawpaw (Carica papaya) and Moringa (Moringa oleifera). The study thus had a threefold objective. Firstly, the study investigated the potential of Pawpaw seed meal (P) and Moringa seed meal (M), as part of a commercial tilapia diet, to be used as endocrine disrupting compounds (EDC’s) to control the reproduction of sexually mature Mozambique tilapia (20-45g). Secondly, assess the potential of P and M to inhibit the attainment of sexual maturity in immature tilapia (2-8g). Finally, P and M to determine its influence on sexual differentiation of tilapia fry (9-12 days posthatch) to produce all-male populations. The study indicated that both P and M seeds contain bio-active chemicals that are capable of disrupting the gonad function, differentiation and sexual maturation of Mozambique tilapia. Sperm production was affected, evident in the degeneration of the testicular tissue samples. Egg production, ovulation and spawning were all affected, as evident in the difference in colour of the degrading eggs, as well as the absence of spawning. Ovo-testes were observed in cases where diets containing 10.0g P and 10.0g M /kg basal diet were fed. Eggs were observed in the ovaries of sexually immature fish, but spawning did not occur. The study also presents the first report on the isolation of Oleanolic acid in Moringa seeds. The evident of antifertility properties of both Pawpaw and Moringa seeds can be exploited to control or prevent reproduction of Mozambique tilapia in SSA aquaculture systems. This could be of particular importance to aquaculture development in rural areas of Sub-Saharan African countries, given the abundant year round availability of these compounds. Further studies are required to optimise the preparation of the experimental compounds; as well as determining the optimal inclusion level of the phytogenic compounds, as well as how their efficacy to manipulate the reproductive potential and ability of Mozambique tilapia are influenced by environmental factors such as water temperature. / AFRIKAANSE OPSOMMING: Akwakultuur is die vinnigste groeiende dierlike voedsel-sektor. Daar word verwag dat dit visserye sal oortref as ʼn bron van voedsel en dat groei in dié bedryf selfs die bevolkingsgroei sal verbysteek. Die vinnige groei in die sektor kan toegeskryf word aan die toenemende vraag na akwakultuur produkte en 'n dringende behoefte vir 'n volhoubare voedsel hulpbron, wat ook die veilige produksie van voedsel sal verseker. Wêreldwyd voed vis meer as 3.5 miljard mense en dra tot 15% van die gemiddelde hoeveelheid dierlike proteïen per kapita ingeneem, by. Die bekendstelling van akwakultuur in sub-Sahara Afrika (SSA) het gedurende die 1940's en 1950's plaasgevind, met die belangrikste doelwitte om voeding in landelike gebiede te verbeter, geleenthede vir diversifisering te skep wat die risiko van misoeste verminder, om bykomende inkomste te genereer en werksgeleenthede te skep. Akwakultuur is gesien as 'n lewensvatbare opsie vir die ontwikkeling van die landelike gebiede in SSA en aansienlike hulpbronne is belê om die ontwikkeling daarvan te ondersteun. Die produksie van enkel-geslag bevolkings (d.i. slegs manlike vis) is ʼn potensiële oplossing vir dié probleme, maar die tegnologie wat gebruik word om enkel-geslag bevolkings te produseer is nie geredelik toeganklik vir hulpbron-arme gemeenskappe wat met Tilapia vir kosdoeleindes boer nie. Fitochemikalieë, anders ook bekend as fito-estrogene, is verbindings wat in plante voorkom en wat struktureel of funksioneel die werking van die natuurlike soogdier estrogene/androgene naboots, met die fitochemikalieë wat die seksuele differensiasie van vis beïnvloed. Fitochemikalieë kom in plante soos papaja (Carica papaya) en Moringa (Moringa oleifera) voor. Die studie het dus gepoog om die potensiaal van papaja saad meel (P) en Moringa saad meel (M), as deel van 'n kommersiële tilapia dieet, om as endokriene ontwrigters (EDC's) gebruik te word om die reproduksie van seksueel volwasse Mosambiek tilapia (20-45g) te beheer, om te voorkom dat onvolwasse tilapia (2-8g) geslagsrypheid te vroeg bereik en ook om die geslagsdifferensiasie van tilapia vingerlinge (9-12 dae na uitkom) te manipuleer om enkel-geslag (manlike) produksiegroepe te produseer. Die studie het aangedui dat beide papaja en Moringa sade bio-aktiewe chemikalieë wat die werking van die gonadotrofien hormone, geslagsdifferensiasie die stadium waarop Mosambiek tilapia geslagsrypheid bereik, kan beïnvloed. Spermproduksie is negatief beïnvloed, soos waargeneem in die degenerasie van die testisweefsel. Eierproduksie, ovulasie en die vrystelling van eiers is almal negatief beïnvloed, soos duidelik waargeneem in die kleurverskil (van normale eiers) van eiers wat ʼn mate van reabsorbsie aandui en die feit dat geen eiers vrygestel is nie. Ovo-testes is waargeneem in gevalle waar diëte met 10.0g papaja en / of Moringa / kg basale dieet gevoer is. Eiers is waargeneem in die eierstokke van seksueel onvolwasse vis, maar vrystelling het nie plaasgevind nie. Die studie is die eerste verslag oor die isolasie van Oleanoliese suur in Moringa sade. Die duidelike reproduksie-beperkende (anti-vrugbaarheid) eienskappe van beide papaja en Moringa sade kan benut kan word om reproduksie in Mosambiek tilapia te voorkom of te manipuleer in SSA akwakultuur stelsels. Dit is veral van besondere belang vir akwakultuur ontwikkeling in die landelike gebiede van SSA lande, gegewe dat beide sade regdeur die jaar geredelik beskikbaar is. A quantitative and qualitative histological assessment of selected organs of Oreochromis mossambicus after acute exposure to cadmium, chromium and nickel

M.Sc. / South Africa is renowned for its exploitable mineral resources and continues to be a major player in the world’s mineral markets. The country is well known for containing the world’s largest gold and platinum repositories and electroplating industries, which is the major cause for delivering by-products such as cadmium (Cd), chromium (Cr) and nickel (Ni). Environmental pollution caused by active mining and seepage from closed mines, continuously threatens South African water resources. Such pollution can cause a shift in water chemistry and increase the availability of certain metals to the living organisms of such a system. Even at low concentrations metals are amongst the most toxic environmental pollutants. As a result of their persistence and capacity to accumulate in the environment, metals have a lasting detrimental effect on the ecosystem. Although there is progress in the treatment of metallic wastes, the discharge thereof by industries is still a serious water pollution problem. In the past, chemical analysis of water has proven to be of great use for the detection of pollutants within the environment. The value of chemical analysis alone has become limiting, as chemical analysis supplies information on the levels of chemicals at a certain time. Furthermore, the monitoring of water quality variables often does not reflect long-term events that may play a critical role in determining the ecosystem health. It is now generally understood that measurements of only the physical and chemical attributes of water cannot be used as surrogates for assessing the health of an aquatic ecosystem. The new trend is to incorporate biological monitoring into Abstract existing monitoring strategies. Fish are entirely dependent on the aquatic environment for their survival, rendering them a good monitor of water pollution. Macroscopic changes in organs are preceded by changes at the tissue, cellular or molecular level. These changes are the net result of adverse biochemical and physiological changes within an organism. Histological analysis is a therefore very sensitive parameter and a valuable technique in determining cellular changes in target organs as a result of exposure to stressors. Fish histology can thus be used as an indicator of exposure to contaminants and assess the degree of pollution. Because of the subjective nature of morphological studies correlations with other quantitative studies are difficult. However, incorporation of quantitative methods is essential to the continued development of histopathology as a biomarker of pollution exposure, and to the interpretation of histological responses. The aim of this study is to qualitatively and quantitatively describe the toxic induced histological changes in the selected organs of Oreochromis mossambicus after acute exposure to Cd, Cr and Ni. Fish were exposed to 10% (n=20) and 20% (n=20) of the LC50 concentration of Cd, Cr and Ni respectively under controlled conditions (23 ± 1°C) for 96 hours in an environmental room with a control group (n=5) for each exposure.

The fisheries potential of Marcusenius pongolensis, Oreochromis mossambicus and Schilbe intermedius in Mnjoli dam, Swaziland

This thesis investigates aspects of the diversity, abundance and biology of the fish species inhabiting Mnjoli Dam, an irrigation dam that is located in the rural lowveld region of Swaziland. Specific objectives for this thesis were: to determine the fish species present and select three principal species based on their abundance; to investigate the relative abundance and distribution of the three selected species; and to describe key population parameters, such as growth, maturity, reproductive seasonality and mortality. The three dominant species were selected on their fisheries potential. These were Marcusenius pongolensis, Oreochromis mossambicus and Schilbe intermedius. Marcusenius pongolensis was the most abundant species in terms of both catch per unit effort and mass. Catch rates were shown to vary according to habitat type. Age and growth parameters were obtained from sectioned otoliths. Marginal zone analysis showed that annulus formation occurred in winter for all three species. The maximum-recorded ages were 8, 6 and 8 for M. pongolensis, O. mossambicus and S. intermedius, respectively. Growth for the three species was best described by the three parameter von Bertalanffy growth model as Lt = 1.88.67(1- exp⁻¹·⁴⁸⁽t⁺°·⁶⁷⁾) mm FL for M. pongolensis; Lt = 226.83(1- exp⁻°·⁴⁽t⁺²·°²⁾) mm TL for O. mossambicus and Lt = 214.59(1- exp⁻°·⁶°⁽t⁺¹·²°⁾) mm FL for S. intermedius. Sexual maturity was estimated for male and female M. pongolensis at 134 mm FL and 119 mm FL, respectively. Oreochromis mossambicus matured at 239 mm TL and S. intermedius at 205 mm FL. Two spawning peaks for M. pongolensis and S. intermedius were observed, one at the onset of summer (November) and the second at the beginning of autumn (February-March). Oreochromis mossambicus also exhibited two spawning peaks, one in spring (September) and the second in autumn (February –March). Mean mortality rate (Z), estimated using catch curve analysis was 0.77 year⁻¹ for M. pongolensis, 0.49 year⁻¹ for O. mossambicus and 0.79 year⁻¹ for S. intermedius. Natural mortality was assumed to be equal to Z since there is no fishery activity at the dam. Size specific selectivity curves were developed for each species. Of all the mesh sizes used, the 44 mm mesh size net had the highest catch rates in terms of numbers for M. pongolensis and S. intermedius. The 75mm mesh caught the highest catch rates for O. mossambicus. . The 44mm and 75 mm meshes seem to be the most suitable mesh sizes for harvesting the candidate species. It is concluded that no commercial fishery should be established on the dam, yet a small subsistence fishery could be a possible option. Management options such as input controls, open and closed seasons and areas and minimum sizes are discussed.

Bioaccumulation of metals in selected fish species and the effect of ph on aluminium toxicity in a cichlid oreochromis mossambicus

M.Sc. / The upper catchment of the Olifants River, from its origin near Bethal, to its confluence with the Wilge River, north of Witbank, as well as it tributaries, are being subjected to increasing afforestation, mining, power generation, irrigation, domestic and industrial activities. These activities have a profound effect on the water quality and the major point sources of pollution in this area include mines, industries and very importantly, combined sewage purification works, located alongside the river, which, in addition to oxidizable material contains detergents, nutrients, and metals. It was therefore necessary to determine the extent to which these activities affect the water quality of the system. The impact of these activities was therefore addressed by a Water Research Commision Project namely "Lethal and sublethal effects of metals on the physiology of fish" of which the present study investigated effects at two localities, namely in the Olifants River (locality OR1) before its confluence with the Klein Olifants River and a locality in the Klein Olifants River (locality KOR1). Apart from the field study, toxicity tests were also performed in a laboratory, in order to determine the effects of low pH and elevated aluminium concentrations on the haematology, osmoregulation and carbohydrate metabolism of the Mozambique Tilapia, Oreochromis mossambicus as the acidification of soil systems may cause the transfer of aluminium into aqueous solutions, where it may be present in different forms. During the field study, the chemical and physical characteristics of the river water were evaluated, with special attention to the concentrations of certain metals (manganese, copper, chromium, lead, nickel, zinc, iron and aluminium) in the water and sediment, as well as in fish, which are known to accumulate the elements supra and are therefore valuable as indicators of these pollutants. The two fish species used for the investigations were the African sharptooth catfish, Clarias gariepinus and the moggel, Labeo umbratus. Four tissue types were dissected, namely the muscle, liver, skin and gill tissues. The metal concentrations in these organs/tissues, as well as in the water and sediment, were determined in a laboratory with an atomic absorption spectrophotometer. Statistical analyses were performed on the results obtained from this study and the order and extent of bioaccumulation of these metals in the water and sediment were determined, as well as in the fish organs/tissues. Its dependence on the size, sex and species of the fish and the localities and seasons were investigated.

Effects of manganese on the haematology of Oreochromis mossambicus and the bioaccumulation of metals in Labeo umbratus

M.Sc. / Metal ions have become an increasing source of pollution in the natural waters of South Africa. This phenomenon is related to the rapid industrial, mining and agricultural activity growth along rivers, especially in the Upper catchment of the Olifants River, Mpumalanga. In general, metal ions have - negative effects on the physiology of fish and other aquatic biota in metal polluted waters. In this study. the sublethal effects of, manganese were determined by exposing the freshwater fish, Oreochromis mossambicus, to this metal in an experimental flow-through system. The exposure times were divided in acute (96 hours) and chronic (26 days) exposures, both at 23 ± 1°C. The results obtained showed changes in the general, haematological, metabolic, osmoregulatory variables, as well as the differential white blood cell counts. These changes indicated several effects in the physiology of a mossainblais after sublethal manganese exposure. Significant differences (P < 0.05) were found between the haematological variables of the control and exposed fish. During chronic exposure, an oxygen deficiency developed due to , the epithelial lifting of gill lamellae. As a result of hypoxia the red blood cell numbers, haemoglobin-, haematocrit and lactate concentrations increased to uplift the depleted oxygen situation. The significant differences (P < 0.05) found in the sodium, potassium,. calcium and chloride concentrations are mainly as a result of gill damage. Manganese showed a disruptive influence on the structural organization of the gill. The differential white blood cell counts performed , showed significant fluctuations. This leucocytosis and leucopenia are normal reactions of the fishes’ body against infections of foreign substances, such as metal ions. It was concluded that the general haematology, metabolism, osmoregulation and differential white blood cell counts can, be used as indicators in detecting the effects of sublethal manganese exposure on fish.

The work includes: an overview of world wide trends in power plant thermal effluent aquaculture, the identification of suitable power stations for aquaculture, long term monitoring of the physical and chemical characteristics of the cooling water at selected power stations, including the effects of the cooling water on the growth response, food conversion, condition factor, survival rate and suitability for human consumption of the aforementioned fish species reared in the cooling water at Matla and Kriel Power Stations.
