In our environment there are many compounds which can negatively influence humans and wildlife. Every day, a vast number of environmental pollutants are released into our environment and there is no way to avoid their exposure. Some of these compounds can even mimic endogenous hormones and interfere with our endocrine system (so called endocrine disruptors), which is the key regulatory system controlling almost all physiological processes in human and animal bodies. Also the reproductive system is largely regulated by various hormones, and their proper function is crucial for gamete formation, fertilization and embryo development. Environmental pollutants are therefore considered as one of the possible causes of increased infertility in human population. This prompted us to study the effect of two endocrine disruptors (tetrabromobisphenol A - TBBPA, and zearalenone - ZEA) on the male mouse reproductive system in vivo. According to our results, TBBPA is able to induce apoptosis as well as changes in the expression of selected testicular genes and sperm protamination. Our results also suggest that permanent exposure to TBBPA slightly enhances its effect in the next generation, depending on whether the parents have been affected or not. Toxoplasma gondii is one of the most widespread parasitic protozoa in humans in developed countries. It has a wide range of hosts including various warm-blooded animals and humans. Recent studies suggest that it could influence the fertility of humans. The aim of this work was to find out, what is the prevalence of toxoplasmosis in groups of women and men with fertility problems and whether Toxoplasma gondii has an effect on specific fertility parameters. In women, Significant positive correlation between toxoplasmosis and the percentage of oocytes in the follicles was found among women. Group of men with semen pathology diagnosis revealed a statistically significant negative association between toxoplasmosis and the percentage of morphologically normal sperm. It seems that latent toxoplasmosis in men can also worsen the negative effect of tobacco smoking on fertility. The impairment of fertility in men could be the result of the manipulation activity of Toxoplasma gondii, which leads to more frequent sexual intercourse due to decreased ability to conceive. This could increase the likelihood of sexual transmission, which is currently suggested as one of the possible ways of transmission of toxoplasmosis.

The subject of my thesis is to analyze the evolution of the reproductive behavior of the Czech population to assess changes over time, especially after 1989 and to analyze the success of assisted reproduction in the Czech Republic in the period 2007 to 2013. It is divided into several parts. The first theoretical part describes the change in reproductive behavior after 1989, risk factors and causes of female and male infertility, possible tratments of diagnosed infertility and various methods of assisted reproduction. Another analytical part is dedicated to the evolution of fertility and the phenomenon of childlessness of young women. Some indicators are compared with selected European countries for an overall assessment. Subsequently are analyzed in detail the most widely used methods of assisted reproduction IVF, ICSI and kryoembryotransfer and of using indicators Pregnancy rate and Take-home baby rate is assessed their effectiveness. The thesis is supplemented by own research reproductive and family behavior on a selected sample of the Czech population.

The main purpose of the work is to evaluate the commercial surrogacy system efficiency. Within the solution of this question, the work focuses first on the analysis of the arguments against a market in human organs and consequently theise objections applies to the surrogate motherhood with emphasis on the problem of adverse selection, which from an economic point of view appears to be the most serious. In the applied part the paper tests the existence of the signs of the adverse selection in the countries where payment for the surrogacy is legal. The paper leads to the conclusions, despite some specifics of the market, this solution is efficient and the market itself creates sufficient mechanisms to prevent adverse selection

It reveals how womanhood and motherhood are constructed in different discourses in context of uterine transplantation through critical reading and thinking. By using of critical discourse analysis I focus on texts produced on this subject in the popular and professional discourse and reveal what kind of gender stereotypes and constructs they are using to support their arguments. These areas of our everyday lives have great power in society and influence the way we perceive and construct reality and thus participate in its creation. This master thesis deals with infertility and its treatment. Both in the theoretical and empirical part, it focuses on the female aspects of the issue. The theoretical part summarizes the knowledge which is important to understand the presented problems. We define the terms; we describe the psychological and physiological causes of infertility, the process of treatment of infertility and aspects thereof. We focus on the role of stress and the strategies of coping with stress related to infertility and its treatment; furthermore, we deal with personal causality orientation related to the experienced level of stress as one of the determinants of coping with stress. Our aim in the empirical part is to describe the use of strategies of coping with stress within the sample of women acquired by us, the distribution of personal causality orientation of these women and to capture the possible connections. Infertility is defined as an inability to conceive a child within one year of regular sexual intercourse. It affects up to 15 % of couples worldwide (WHO, 2010). The male factor contributes to the total infertility with more than 50 %. Fertility of a man is influenced by several factors such as genetic background, environment and various diseases such as diabetes mellitus (DM). Diabetes mellitus is a serious health problem that affects 451 million people worldwide (18-99 years) and the number of people with this disease still increases (Cho a spol., 2018). In addition, parenthood is postponed to middle age when the fertility decreases and metabolic diseases such as type 2 diabetes mellitus (DM2) appear. The aim of this thesis was to investigate the effect of type 2 diabetes on reproductive parameters of mouse inbred line C57BL/6J compared to the control group and the possible effect of paternal diabetes on the first filial generation. In the evaluation of the effect of DM2 on reproductive parameters, we used innovative methods to study internal state of sperm and testes. Currently infertility affects 10 to 15 percent of couples. In nearly 50% of all cases male factor contributes to infertility of the couple. Majority of causes of male infertility remains unexplained. For this reason, finding simple and clinically useful tools for improving male infertility diagnostics can be important at present. Based on the analysis of sperm transcriptome, in this diploma thesis we aimed to find genes that show differential expression between normal and pathological sperm, this could provide information about molecular basis of male infertility, moreover, expression profile of such genes in sperm could be employed for noninvasive diagnosis of male infertility. This study was conducted by using 67 sperm samples, including 16 control sperm samples from fertile men. The samples of infertile patients were divided into 3 groups according to morphology and motility using results of standard sperm evaluation according to WHO.
