Kontejnerový nosič / Container trailer

This thesis deals with the design of a container carrier with interchangeable superstructures up to 3000 kg of technical weight. The introduction renders a critical review of similar devices and consecutive review of the device in terms of legislative matters. A dynamic simulation was used to calculate the loaded states. The outcome of the simulation was used to set the border conditions, which were then used for strength analysis calculations using finite element method (FEM). Blueprints of the final design are a part of the thesis.

This Diploma Thesis is dedicated to a proposal of a carrier swap bodies hooked up to a agrotruck 6x6 with a maximum technical weight 38000 kg and has been worked out in corporation with ZDT Nové Veselí company. The first section of the Thesis is occupied with legislation and then especially with a description of available agrotrucks and similar constuction solutions not just of the carrier swap bodies. In the following part of the Thesis the own carrier swap bodies construction proposal has been described, based on which the stress analysis has been done in the final part of the Thesis. Also the technical drawing forms an integral part of the Thesis.

The aim of the thesis was development of an innovative treatment of bone defects. Human multipotent mesenchymal stromal cells (MSC) play a crucial role in bone healing. Clinical applications of MSC require large amount of cells, which could be obtained by autologous expansion of MSC harvested from bone marrow. As a first step, the standard protocol of MSC expansion based on αMEM medium and fetal bovine serum (FBS) was used. Experiments replacing FBS by pooled human serum (HS) in the culture medium concluded in patenting of a new MSC cultivation protocol (EU 1999250, CR 301141). This one-step cultivation protocol and xenogeneic protein-free cultivation medium is based on CellGro® for Hematopoietic Cells' Medium, HS, human recombinant growth factors, dexamethasone, insulin and ascorbic acid. The preclinical in vitro and in vivo experiments with MSC from both expansion protocols were carried out. Fibrillar polylactic scaffolds were seeded with MSC, cultured, differentiated and implanted in immunodeficient mice (NOD/LtSz-Rag1-). Bone-like mineralized tissue containing vessels was observed. The MSC cultured according to patented method were classified as Advanced-therapy Medicinal Product and has to fulfil the European Medicines Agency regulations to enter the clinical trials. This results were used like a input data to solve the construction with finished element method. After evaluation of results were made design changes of hook loader. For a calculation of load states is used a dynamic simulation and its results are used as a boundary conditions for the strength analysis using finite element method (FEM). The results of the analysis is evaluated and on theirs basis are designed a construction improvements. The part of the thesis is drawings documentation of the construction improvements.

The next chapter deals thoroughly with the legal definition of fine art, including both its traditional and modern forms. The following part summarizes the means of legal protection of fine art, by both private and public actions. This diploma thesis deals with the optimization of ultra-high vacuum scanning electron microscope - UHV SEM, which is developed within the Amispec project in cooperation with BUT, Institute of Scientific Instruments of the Czech Academy of Science and Tescan Brno, s.r.o. The theoretical part deals with the description of the actual state of the developed equipment and the research of competing systems. The next part describes the optimization of the sample holder and the pallet receptor for studying nanostructures over a wide range of temperatures. Part of the optimization is the sapphire thermal diode development and experimental verification of the functionality of the designed components. This is followed by the verification of the functionality of the whole system for the transport of samples to the UHV area, deposition with effusion cell and in-situ observations.

Létající atmosférický nosič pro vypouštění raket / Flying atmospheric carrier for rocket launches

The main objective of this thesis is to introduce the reader to the problematics of air-launch and to a custom design solution applying this concept. The specifics of this method of bringing a payload into orbit are described and explained. Overview of projects which use aircraft to launch spacecraft is included. Determination of primary parameters of a launch vehicle designed to carry a payload of a specified mass is conducted. The required flight performance has been estimated, a computational model has been developed in software MATLAB, and a multidisciplinary optimization of the design parameters has been performed using a genetic algorithm optimization method. Parameters of the designed air-launched rocket are compared with those of a ground-launched rocket. According to the specific criteria, the Airbus A310-300 aircraft was selected as the most suitable transport aircraft to be used for launching the designed launch vehicle. The last part of the thesis is devoted to the proposal of necessary modifications and estimation of the flight performance.
