Sekvenční varianty genu HNF1B u autozomálně recesivní polycystické choroby ledvin / Sequence variety of HNF1B gene in autosomal recessive polycystic kidney disease

Autosomal recessive polycystic kidney disease (ARPKD) is a rare severe inherited disease manifested by cystic renal disease, congenital hepatic fibrosis and dilatatation of bile ducts. The spectrum of clinical manifestations is very wide and variable, depends on the age at which the disease was manifested. In severe forms of the disease, it is possible to detect the first symptoms prenatally around the 20th week of pregnancy due to increased echogenic kidneys and the presence of oligohydramnios. The causal gene of this disease is thePKHD1 gene with protein product fibrocystin that is most likely contributing on maintaining the intracellular concentration of Ca2+ cations. The exact phatophysiology mechanism of ARPKD remains unknown. Phenotypic manifestations of this disease may overlap with mutations associated with other genes. One of the genes mimicking the ARPKD phenotype is the HNF1B gene. Mutations associated with HNF1B gene are the most common monogenic cause of developmental kidney abnormalities. HNF1B is a tissue-specific transcription factor that regulates the expression of PKHD1. (in English): The subject of my dissertation is the decision of the subjects to participate in the early phase of clinical trial. The aim of this disseration is to detect and understand the ethical aspects of decision making of subjects with oncological diseases to participate in the early phase of clinical trial. In the theoretical part of my dissertation, I define the field of clinical research, describe the phases of clinical research and ethical codes important for clinical research. The current ethical dilemmas related to the participation of subjects in the early phase of clinical trial, which are being discussed by the professional public, are presented. The research part contains the results of interviews with seventeen respondents whose content correlates with the aim of the research part and it is to understand the decision- making of subjects to participate in the early phase of clinical trial in terms of subjectively constructed realities, which are different for individual respondents. The results of the research show that fourteen respondents identified one of the most important aspects of the decision-making to participate in the early phase of clinical trial of the recommendation of principal investigator and trust in a physician. St Vitus' Cathedral in Prague: Liturgy, Symbolism and Architectural Imitation in Medieval Bohemia In this thesis, the focus is put on the Wenceslas Chapel in St Vitus' Cathedral, the unusual design of which seems to have two levels of meanings: On the one hand, it was probably designed as a recreation of an earlier rotunda that had been founded by St Wenceslas. On the other hand, it seems that the intention was made the chapel and the whole cathedral would resemble the Church of the Holy Sepulchre in Jerusalem, where the Calvary Chapel occupied the same position as the Wenceslas Chapel. This was most likely inspired by presence of the Imperial insignia containing the valuable Passion relics that Charles had acquired in 1350 and placed in the basilica, probably near the Wenceslas Chapel. Perhaps from that reason Charles had the new Gothic chapel decorated with semi-precious stones, because its red spots were associated with the blood of Christ and the Passion. The Wenceslas Chapel together with two Passion chapels at Karlstein Castle, containing Charles IV's private passion treasure, gave rise to the "Passion" architecture of the Luxembourg period. This diploma thesis deals with selection, design and creation of new education materials for high school teacher supporting webpage www.studiumchemie.cz. The selection of the proper theme was done on the basis of research and evaluation of recent materials on the mentioned webpage and materials created at the Department of chemical education of Faculty of Science of Charles University in Prague. Preliminarily, the themes of synthetic macromolecular materials and new materials were selected. The convenience of the selected themes was confirmed by research and evaluation concerned to the preliminarily selected theme of recent high school textbooks and webpages. Consequently, educational materials in the form of PowerPoint presentations, websites, worksheets, tests, games and experiments focused on the themes of synthetic macromolecular substances and new materials were created. Materials were processed in accordance with the Framework Education Programme for Secondary General Education (Grammar Schools) as well as in accordance with the requirements of the Catalogue of graduation exam requirements from chemistry. The diploma thesis solves the new utilization and renovation of an industrial building near the Baťa canal in the village of Huštěnovice which was used as former pumping station for agricultural purposes. The aim was to find such uses that will support the development of the adjacent area and at the same time will also refer to the original usage of the building. New community centre „PŘÍLIV“ with aquaponics connects both of them and what is more, it offers modern connection of people with nature. The construction plan is solved as a complex project form architectural study, through the project for the building permit to the design of the aquaponics technology itself. Project of the community centre and aquaponics is designed as off-grid building with minimal environmental impact.

The designed area of residential buildings responds to the surrounding buildings, the development of existing block and defines the frontage streets of Gorkého and Údolní street. The ends of the lines extend to the building of a hospital and between them is the private garden for the residents. Recessed floor apartment buildings react to remote vista through Čápkova street to the Orthodox durch of sv. Václav from the 30s of the 20th century. The proposed passage in the extension of Čápkova street, bordered on one side with three-story apartment buildings, allowing access to Špilberk, which is an alternative to the uncomfortable route along the busy Úvoz stree. The new use of the historic building should be private fertility clinic and center for assisted reproduction, which follows the 130 years of unbroken tradition, and the object corresponds to the developing trend of contemporary luxury parenthood.

Územní studie „Zbrojovka“ / Town planing study "Zbrojovka"

The aim of this thesis is a revitalization of the grounds once belonging to Zbrojovka Brno. These grounds are in the zoning map defined as “brownfield”. Within Brno’s limits, the area has an extremely convenient location. Since the grounds are situated in the central part of Brno, the complex is in terms of traffic and transport in the vicinity of both, the current and the newly planned ring-road. Important factors influencing the project are the river Svitava which borders the west side of the area, and a railroad corridor skirting the east side of the location. Assigned area was, for the purposes of this thesis, expanded by the grounds once belonging to Zetor and the adjoining area towards Markéty Kuncové Street. Several buildings from the former grounds, which fit in into the urban concept and were in good technical conditions, have been preserved. From a functional point of view, the newly designed grounds will meet all the requirements for a fully independent municipal district. The multifunctional development contains primarily housing complexes, administrative buildings, public facilities, recreational areas, as well as other functional structures. The urban concept of this project is a result of a study of surrounding blocks. Based on an analysis of the size and height of these buildings, an idealized block was designed and then recreated into a symmetrical raster of blocks with the street width of 20m. This raster was subsequently applied to newly designed points of entry into the area. Total urban and mass proposal of the development areahas been further influenced by principal conceptual points. Among these conceptual points belong the perspective axes which connect the conceptual points with an orientation point, in this case the former factory’s smokestack. Creating a new arm for the Svitava River is another point. Taking into account that the whole vicinity of the factory grounds is without quality public greenery elements.
