Vliv textury briket z biomasy na jejich mechanické vlastnosti / Influence of texture of briquettes from biomass to their mechanical properties

With the rising amount of AD technology utilizers, the issue how to storage, such liquid material, which digestate is. In recent time, there are some other ways to utilize it, except fertilizing with liquor, liquid part of digestate. Digestate can be dried and pressed into briquette or pelleted form. In briquette form is important to understand relations between properties of individual components because one property can influence another one, which constituting texture of the material and its reprocessing into the final form of the product. Thus this study was conducted, to find and/or quantify the relation between the texture of biomass briquettes and their durability and strength of shape during manipulation, storage and use. Basic hypothesis of this work is based on open presumption that briquette texture is dependent on many parameters. Some of those parameters were tested. Namely, composition of matter, physical properties of digestate briquettes, durability and hardness, size analysis and other. Digestate has approved as good material for compression with high content of nutrients. The rate of abrasion varied 7-12% at the beginning decreasing to 3-5%. Shore testing showed results predicating relation between particle size and hardness. Sieve and image analysis then showed range of particles ranging size 0.01 mm from 99.5% and 10 mm from 99.7%, most of them with needle-like shapes. This study suggested the method that allows analysing particle size distribution in sample and describes other briquette properties, which can be useful for next research and commercial purposes.

Návrh a realizace přístroje na sledování a vyhodnocování pohybové aktivity hmyzu / Design and implementation of instruments for monitoring and evaluating physical activity of insects

The aim of the diploma thesis is the development and subsequent construction of the device whose function is measuring and evaluating physical activity of insects. This instrument is unique in the Czech Republic and will be able to record physical activity of insects by using a video camera. Later that it will be able in this video shot back to reconstruct kinematic and statistical data about the movement of insects with software which is developed with Visual Basic .NET. On the base sufficient od obtained data it can be used to verify the premise of biologists about nervous systém, brain function and behavioral of Drosophila Melangostar which is more than a century used as model for genetics experiments. Finally, the conclusion compares the result of previously known and available software solutions in the world of biology, which are still used as a source for the results of performed experiments.

Digitální steganografie a stegoanalýza / Digital Steganography and Steganalysis

This thesis deals with digital steganography and steganalysis. It explains the significance of both disciplines and gives a brief overview of the history in the given fields. The paper separates existing steganographic and steganalytic methods and describes the attributes of various branches of steganography. With respect to the chosen field of steganography (the image steganography), the paper narrows down the set of suitable steganographic and steganalytic methods, whose features are then described in detail. The results of the thesis inlcude experiments that verify the features of chosen steganographic methods and evaluation of steganalytic methods and their success in detection of steganography.

Analýza strukturálních změn nanovlákenných scaffoldů vzhledem k jejich relativnímu prodloužení / Analysis of structural changes of nanofiber scaffolds in relation with their relative extension

The goal of this thesis was to find a suitable method for evaluating structural changes of scaffolds as influenced by external mechanical pressure and to verify the validity of hypothesis which assumes a change of directionality of fibers and also thinning of fibers according to the stretching of a scaffold. Assumptions formulated in these three hypothesis were tested on a scaffold with a plasma surface treatment and without any plasma surface treatment. To examine structural changes an electrone microscope was used to observe the surface of scaffolds. Incurred photos were then processed with the help of automatic software picture analysis and observed data were statisticly evaluated. The result of this experiment is a description of used method which can be used in future for larger studies. It was found that by the effect of external mechanical pressure fibers of examinated scaffolds turn in the direction of the pull. It has also been found that the average thickness of fibers didn't change. Results of this work give insight into the evaluation of structural changes of scaffolds when pressured by an external mechanical power and open possibilities for deeper and more exact research in this field. The Master Thesis The depiction of events in Ukraine on photographs - ethical aspects of depicting conflict presents a case study focused on depiction of Ukrainian crisis in photographs during a defined period of time of 2014 in five Czech and five foreign newspapers. The first part of the thesis introduces the theoretical concepts that are the basis for the analysis. The analytical part presents the research of depiction of the Ukrainian events in news photographs in the daily newspapers using the technique of content picture analysis. The results let us compare the differences between the approach of the individual Czech and foreign newspapers to the issue of publishing graphic images of violence and its victims. Assessment of semen quality (sperm viability) is an essential for the efficient honeybee semen storage. The motility of spermatozoa is not the ultimate parameter of sperm viability. Instead, the viability of spermatozoa is assessed by testing their structural or functional plasma membrane integrity. These assays are based primarily on the technique of microscopy evaluation of either fluorescent DNA dyes or hypo-osmotic "tail-coiling" reaction of spermatozoa. However, the technique of microscopy evaluation is time-consuming, subjective and often is not accurate. On the other hand, the technique of flow cytometry is rapid, objective and precise. Nowadays, this technique is affordable for many institutions, as the price of "benchtop" flow cytometer is relatively low. To best of our knowledge, there is currently no flow cytometry assay available for evaluation of functional integrity of honeybee spermatozoa. Here we report the establishing of novel flow cytometry assay for simultaneous evaluation of structural and functional plasma membrane integrity. DRAQ7, the new fluorescent DNA dye was used to test the structural integrity of plasma membranes. The hypo-osmotic "tail-coiling" reaction was used for flow cytometric assessment of functional integrity of spermatozoa plasma membranes. The combination of both is a valuable tool for rapid and precise evaluation of sperm viability in honey bees.

Stanovení velikosti genomu jeseterů 2-D a 3-D obrazovou cytometrií. / The genome size determination in sturgeons using 2-D a 3-D image cytometry.

The genome size of evolutionary polyploid, neopolyploid and hybrid sturgeons is well known for its high variability. Aim of this study was 1) to specify the genome size of polyploid and neopolyploid sturgeons using an analysis of 2-D and 3-D images of specifically stained cells nuclei, 2) to evaluate the samples of populations for cytogenetic analysis needs and thereafter, 3) to compare both methods and record the data either for next research or for negative selection from the broodstock. This test has been done at the laboratory of molecular, cellular and quantitative genetics, Faculty of Fisheries and Protection of Waters USB in Vodňany using all sturgeons spawners and using samples obtained from some foreign fish farms which cooperated with the faculty. The samples included A. ruthenus, A. baerii, A. stellatus, A. gueldenstaedtii and Huso huso, intentionally bred hybrids of A. gueldenstaedtii (8n) x A. baerii (12n), A. baerii (8n) x A. ruthenus (4n), A. gueldenstaedtii (8n) x A. baerii (10n) a A. gueldenstaedtii (8n) x A. ruthenus (4n). As methods have been chosen image cytometry and confocal microscopy which use image digitalization and subsequently its computer analysis. The genome size was measured from the size of specifically stained nuclei of erythrocytes in specimens sampled. Result of this study was measuring the genome size in sturgeons under study using different methods, recording the obtained data, description of spatial conformation changes of cell nucleus with increasing ploidy level and deduction of impact to their physiology and comparing the methods between each other. The conclusion is necessity of another sturgeons genome size determination , choice of the best methods for more effective search and research of non-standard individuals and subsequently an examination of their physiological differences.

Využití generativních modelů neuronových sítí v obrazové rekonstrukci / Generative neural networks in image reconstruction

Recent research in generative models came up with a promising approach to modelling the prior proba- bility of natural images. The architecture of these prior models is based on deep neural networks. Although these priors were primarily designed for generating new natural-like images, its potential use is much broader. One of the possible applications is to use these models for solving the inverse problems in low-level vision (i.e., image reconstruction). This usage is mainly possible because the architecture of these models allows computing the derivative of the prior probability with respect to the input image. The main objective of this thesis is to evaluate the usage of these prior models in image reconstruction. This thesis proposes a novel model-based optimization method to two image reconstruction problems - image denoising and single-image super-resolution (SISR). The proposed method uses optimization algorithms for finding the maximum-a- posteriori probability, which is defined using the above mentioned prior models. The experimental results demonstrate that the proposed approach achieves reconstruction performance competitive with the current state-of-the-art methods, especially regarding SISR.

Automatická detekce přechodů pro chodce z leteckých snímků / Automatic detection of zebra crossings from aerial images

There are a number of studies that deal with detection of road network from image Remote Sensing data. However, little work has been done on algorithms for horizontal road signs detection from Remote Sensing data. On the other hand there are many papers that deal with horizontal road signs detection in the field of Computer vision. Theoretical part of master thesis sumarizes the methods that are used to detect objects from both, Remote Sensing and Computer vision data. In the practical part an algorithm of automatic Zebra-crossing detection based on Aerial Images and vector road layer is designed. Zebra-crossings are detected using matching of image with set of predefined patterns. Obtained set of potential objects is then filtred using geometric and relational criteria. At the end of this thesis, the proposed algorithm is validated and the results are discussed with literature. Key words: Zebra-crossing, Horizontal road signs, Image matching, Aerial images
