of many pathogens. Rhipicephalus sanguineus parasites mainly dogs. It is known
that Beagle dogs are less susceptible to R. sanguineus than English Cocker
Spaniel, and still have resistance patterns linked to immunity. The lower
susceptibility of Beagle dogs is due to chemical compounds they produce, which
act in reducing the parasitic load of R. sanguineus on these animals. Given that
this research aimed to verify the chemical repellency against R. sanguineus of
compounds that have been identified in the odor of resistant dogs, and synthetic
repellent compounds to other tick species. For testing chemical repellency two
tests were used, the Petri dish and the Y-olfactometer. Five compounds identified
in Beagle odors were selected and tested: 2 - hexanone, nonane, decane,
undecane and benzaldehyde, the compound of botanical origin β-citronellol and
the standard repellent DEET (N,N-diethyl-3-methylbenzamide). These compounds
were tested alone and also two mixtures were evaluated: benzaldehyde and 2-hexanone (1:1), and benzaldehyde, 2-hexanone and undecane (1:1:1). All these
compounds were prepared in a stock solution of 7.2 % ( ≈ 0.200 mg/cm²) and two
lower concentrations were also tested: 0.100 and 0.050 mg/cm². DEET was
repellent in the two highest concentrations and 68.33 to 86.67% of the ticks were
found in untreated areas in different time points. At the concentration of 0.050
mg/cm² ticks were distributed similarly (without significant difference) in both
areas. β-citronellol was the only compound tested that was repellent at all times
and concentrations tested. The percentage of ticks in untreated areas ranged from
73.33 to 93.33 %. When compared β-citronellol with DEET the first compound was
more effective at repelling the adults of R. sanguineus. For benzaldehyde, 30
minutes at a concentration of 0.200 mg/cm² a percentage of 63.33 % of ticks was
observed in the untreated area. Percentage of 76.67 was observed after 5 min. at
a concentration of 0.050 mg/cm². 2-hexanone was repellent at 30 min. at a
concentration of 0.200 mg/cm² at 10 min. at a concentration of 0.100 mg/cm² and
30 minutes at a concentration of 0.050 mg/cm². For undecane a rate of 65.00% of
ticks was observed in the untreated area in one moment of testing, the highest
concentration during the first 5 min. of the evaluation. Nonane and decane not
were not repellent at any of the concentrations and time intervals. The blend
benzaldehyde + 2-hexanone was repellent in the three highest concentrations in
almost all evaluated times, including in the olfactometer testing. During the whole
test this mixture showed statistical similarity when compared with DEET. The
addition of undecane to this blend did not potentiate its repellent power. The main
result was the finding of natural repellents in resistant dogs to the tick R.
sanguineus. / Carrapatos são artrópodes de grande importância dentro da saúde pública, devido
à transmissão de inúmeros patógenos. Rhipicephalus sanguineus parasita
principalmente cães. Sabe-se que cães da raça Beagle são menos susceptíveis
ao R. sanguineus que cães Cocker Inglês, e ainda apresentam padrões de
resistência ligados a imunidade. A menor susceptibilidade de cães Beagle está
ligada a compostos químicos que estes produzem, os quais atuam na diminuição
da carga parasitária de R. sanguineus sobre estes animais. Sabendo-se disso
com esta pesquisa objetivou-se verificar a repelência química de compostos que
foram identificados no odor de cães resistentes, além de compostos sintéticos
repelentes para outros carrapatos, contra o R. sanguineus. Para os testes de
repelência química foram utilizados dois ensaios de repelência, o da placa de
Petri e do olfatômetro em Y. Foram testados cinco compostos com provável poder
repelente e identificados nos odores de Beagle (2-hexanona, nonano, decano,
undecano e benzaldeído), o composto de origem botânica β-citronelol e o
repelente padrão DEET (N,N-dietil-3-metilbenzamida). Além destes compostos
testados isoladamente, duas misturas foram avaliadas: benzaldeído e 2 -hexanona
(1:1), e benzaldeído, 2-hexanona e undecano (1:1:1) Todos estes compostos
foram formulados a partir de uma solução mãe de 7,2% (≈ 0,200 mg/cm²) da qual
foi obtida posteriormente outras duas concentrações (0,100 e 0,050 mg/cm²). O
DEET foi repelente nas duas maiores concentrações, sendo que 68,33 a 86,67%
dos carrapatos foram encontrados nas áreas não tratadas, nos diferentes tempos
de observação. Na concentração de 0,050 mg/cm² os carrapatos se distribuíram
similarmente (sem diferença significativa) em ambas as áreas. β-citronelol foi o
único composto testado que foi repelente em todos os tempos e concentrações
testados. Os percentuais de carrapatos nas áreas não tratadas variaram de 73,33
a 93,33%. Quando comparado com o DEET o β-citronelol foi mais eficiente em
repelir os adultos de R. sanguineus. Para o benzaldeido, aos 30 minutos, na
concentração de 0,200 mg/cm² foi observado um percentual de 63,33% de
carrapatos na área não tratada. Percentual de 76,67 foi observado aos cinco
minutos, na concentração de 0,050 mg/cm². A 2-hexanona foi repelente aos 30
minutos, na concentração de 0,200 mg/cm², aos 10 minutos, na concentração de
0,100 mg/cm² e aos 30 minutos, na concentração de 0,050 mg/cm². Para o
undecano foi observado um percentual de 65,00 de carrapatos na área não
tratada em apenas um momento dos testes, na maior concentração, durante os
primeiros cinco minutos de avaliação. Nonano e decano não causaram qualquer
repelência para os carrapatos testados, em qualquer uma das concentrações ou
intervalos de tempo avaliados. A mistura benzaldeido + 2-hexanona foi repelente
nas três maiores concentrações, em quase todos os tempos avaliados, inclusive
no teste de olfatometria. Durante todo o teste esta mistura mostrou semelhança
estatística quando comparada com o DEET. A adição de undecano a esta mistura
não potencializou o seu poder repelente. O principal resultado obtido foi a
constatação da presença de repelentes naturais em cães resistentes ao carrapato
R. sanguineus.

Morfometria geométrica, dimorfismo sexual e avaliação da atratividade de Cratosomus flavofasciatus (Coleoptera: Curculionidae) ao feromônio sintéticoCampos, Philippe Correia Souza 24 February 2017 (has links)

Identifying the sex of individuals is an essential step in conducting behavioral studies. An integral part of animal behavior is communication, which can be intermediated by pheromones. Within different groups, insects are the animals use pheromones the most in order to carry out their fundamental activities. Cratosomus flavofasciatus, a Coleoptera of the Curculionidae family, popularly called the Orange-tree-borer, is considered one of the main citrus pests, occurring in Sergipan orchards. The objective of this work was to evaluate the morphological characteristics that allow the sexual differentiation of C. flavosfaciatus and to evaluate the attractiveness of C. flavosfaciatus to the synthetic compounds that make up the pheromone of this species. To verify the sexual dimorphism of C. flavofasciatus, a geometric morphometry analysis was first made using 80 adult individuals of each sex. There were 27 landmarks in the ventral region in each individual. The anatomical landmarks 2, 9, 10, 11, 18, 21 were the ones that contributed most to the distinction between males and females, which represent the region of the rostrum, the final portion of the abdomen and the end of the thorax. For sex confirmation, a withdrawal of the genitalia of both sexes was also performed. In females it was possible to recognize the following structures: sternite VIII and bursa copulatrix. In males, it was possible to observe the gastric spicule and the aedeagus with a pair of parammero. The Procrustes Anova showed significant differences between the sexes (P<0.001) and the size (centroid) in relation to sex (P <0.001), but there was no difference in shape in relation to size (P = 0.229) and the form in relation to sex in conjunction with the size (P = 0.707). Principal component analysis demonstrated an evident differentiation between the genders based on the ventral region of the species. The first two main components explained 51.909% (PC1 + PC2: 39.563% + 12.346%) of the shape variation. The discriminant analysis indicated a significant difference (permutation test = 10,000 permutations; P < 0.001) for the sexual dimorphism of the ventral region. The determination of the adult sex of C. flavofasciatus through the last abdominal sternum is as accurate as the dissection of the genitalia. The behavioral response of C. flavofasciatus to synthetic pheromonal compounds (a mixture of 2-((1R, 2S)-1-methyl-2-(prop-1-en-2-yl)cyclobutil)ethanol (grandisol), (E)-2-(3,3-dimethyl-cyclohexylidene)ethanol (alcohol E) and (Z)-2-(3,3-dimethyl-cyclohexylidene)ethanol (alcohol Z)) and for their host plants Cordia curassavica (Maria-Preta) and Citrus sinensis (Orange Tree) were evaluated by olfactory tests performed on a Y-tube olfactometer. Both sexes were significantly more attracted to C. sinensis stalks (P <0.05) than to filtered air. Tests with C. curassavica stalk attracted only males (P = 0.02). When the insects had the opportunity to choose between C. curassavica stalks and C. sinensis stalks, males preferred those from C. curassavica (P = 0.01), whereas females showed no significant preference (P> 0.05). The combination of the C. curassavica and C. sinensis volatiles associated with the synthetic pheromone at 100ppm concentration was more attractive for both sexes (P <0.05) than for the control (hexane together with the host plant). On the other hand, at the concentration of 10ppm, only the association with C. curassavica attracted males (P = 0.009). These results can be used as a basis for field testing, contributing to the development of an environmentally safe and effective method for monitoring orange tree borer. / O conhecimento sobre a biologia e o comportamento animal é fundamental para compreensão de como determinado organismo interage com o seu ambiente. A identificação do sexo dos indivíduos é um passo essencial na execução de estudos comportamentais. Um ponto integrante do comportamento animal é a comunicação, a qual pode ser intermediada por feromônios. Dentre os vários grupos, os insetos são os animais que mais utilizam os feromônios para desempenhar suas atividades fundamentais. Cratosomus flavofasciatus, um coleóptero da família Curculionidae, popularmente chamado de broca-da-laranjeira, é considerado uma das principais pragas do citros, apresentando grande ocorrência em pomares sergipanos. O presente trabalho teve como objetivo avaliar características morfológicas que permitam a diferenciação sexual de C. flavosfaciatus e avaliar a atratividade de C. flavosfaciatus aos compostos sintéticos que compõem o feromônio de agregação dessa espécie e à suas plantas hospedeiras. Para verificar o dimorfismo sexual de C. flavofasciatus, primeiramente foi feito uma análise de morfometria geométrica utilizando 80 indivíduos adultos de cada sexo. Foram feitos 27 marcos anatômicos (landmarks) na região ventral em cada indivíduo. Os marcos anatômicos 2, 9, 10, 11, 18 e 21 foram os que mais contribuíram para a distinção entre machos e fêmeas, os quais representam a região do rostro, a porção final do abdômen e o final do tórax. Para confirmação do sexo, foi realizada também a retirada da genitália de ambos os sexos. Nas fêmeas foi possível o reconhecimento das seguintes estruturas: esternito VIII e bursa copulatrix. Já nos machos foi possível observar a espícula gastral e o edeago com par de parâmeros. A Anova de Procrustes demonstrou diferença significativa da forma (P < 0,001) e do tamanho (centróide) (P < 0,001) entre os sexos, porém não houve diferença da forma em relação ao tamanho (P = 0,229) bem como da forma em relação ao sexo em conjunto com o tamanho (P = 0,707). A análise de componentes principais demonstrou uma evidente diferenciação entre os sexos com base na região ventral da espécie. Os dois primeiros componentes principais explicaram 51,91% (PC1 + PC2: 39,56% + 12,35%) da variação da forma. A análise de discriminantes indicou diferença significativa (teste de permutação = 10000 permutações; P < 0,001) quanto ao dimorfismo sexual da forma da região ventral. A determinação do sexo em adultos de C. flavofasciatus através do último esternito abdominal é tão precisa quanto à dissecação da genitália. A resposta comportamental de C. flavofasciatus aos compostos feromonais sintéticos (mistura de 2-((1R, 2S)-1-metil-2-(prop-1-en-2-il)ciclobutil)etanol (grandisol), (E)-2-(3,3-dimetilciclohexilideno)etanol (álcool E) e (Z)-2-(3,3-dimetilciclohexilideno)etanol (álcool Z)) e para suas plantas hospedeiras Cordia curassavica (Maria-Preta) e Citrus sinensis (Laranjeira) foi avaliada por meio de testes olfativos realizados em um olfatômetro em “Y”. Ambos os sexos foram significativamente mais atraídos para os caules de C. sinensis (P < 0,05) que para o ar filtrado. Já os testes com caules de C. curassavica atraíram significativamente somente os machos (P = 0,02). Quando os insetos tiveram a oportunidade de escolher entre caules de C. curassavica e caules de C. sinensis, os machos preferiram os de C. curassavica (P = 0,01), já as fêmeas não apresentaram preferência significativa (P > 0,05). A combinação dos voláteis de C. curassavica e C. sinensis associadas ao feromônio sintético na concentração de 100ppm foi mais atrativa para ambos os sexos (P < 0,05) que para o controle (hexano juntamente com a planta hospedeira). Por outro lado, na concentração de 10ppm, somente a associação com C. curassavica atraiu apenas os machos (P = 0,009). Tais resultados podem ser utilizados como base para testes de campo, contribuindo para o desenvolvimento de um método ambientalmente seguro e eficaz para o monitoramento da broca-da-laranjeira.

Most insects use chemical, visual, or acoustic signals in interspecific communication. Sex pheromones are the main signals involved in mating behavior. In this sense, this work aimed to describe the mating behavior of G. spegazziniana, to verify the attractiveness between males and females in Y olfactometer bioassays, and to identify the sex pheromone of this species. To describe the mating behavior virgin couples were placed in cages containing a yerba mate seedling. The insect behavior during the photophase and scotophase was observed. For behavioral tests in Y-olfactometer, virgin males or virgin females and a branch of yerba mate were used as odor source. To verify the presence of a sexual pheromone, body extracts of virgin males or virgin females were obtained and were analyzed by using gas chromatograph coupled to mass spectrometer (GC-MS). Mating occurred only during the photophase, in the scotophase the couples remained immobile in the abaxial surface of the leaves. The mating sequence showed that the female walks on the new leaves and the male goes to meet her, standing face to face with her. After a brief antennal touch between the couple, the male turns the abdomen and attaches to the female. As a result of Y olfactometer bioassays, males and females were attracted to females in the olfactometer but were not attracted to males, suggesting a presence of volatiles released by females for sexual attraction. There was a presence of a single female-specific compound, identified as 2-nonadecanone, that may act as sex pheromone.The results suggest that G. spegazziniana females release sex pheromone to attract males for mating. The compounds released by the females can interact with the volatiles emitted by the yerba mate, resulting in the attractiveness of G. spegazziniana males. / Gyropsylla spegazziniana (Hemiptera: Psyllidae) é um psílideo considerado uma das principais pragas da cultura da erva-mate (Ilex paraguariensis), causando grandes prejuízos econômicos. A maioria dos insetos utiliza sinais químicos, visuais ou acústicos na comunicação interespecífica. Os feromônios sexuais são os principais sinais envolvidos no acasalamento. Neste sentido, este trabalho teve como objetivo descrever o comportamento de acasalamento de G. spegazziniana, verificar a atratividade entre machos e fêmeas em bioensaios com olfatômetro em Y e identificar o feromônio sexual da espécie. Para a descrição do acasalamento, casais virgens foram colocados em gaiolas contendo uma muda de erva-mate. Foi observado o comportamento dos casais durante a fotofase e escotofase. Para os ensaios de atratividade em olfatômetro em Y, foram utilizados insetos vivos (machos e fêmeas virgens) e um ramo de erva-mate como fonte de odor. Para analisar a presença do feromônio sexual foram feitos extratos do corpo de machos e fêmeas virgens, que foram analisados em cromatógrafo gasoso acoplado à espectrômetro de massas (CG-MS). Foi verificado que a cópula ocorreu somente durante a fotofase, sendo que na escotofase os casais permaneciam imóveis na parte abaxial da folhas. A sequência de acasalamento demonstrou que a fêmea caminha principalmente pela brotação e o macho vai ao seu encontro, ficando frente a frente com a mesma; após um breve toque de antenas entre o casal, o macho vira o abdome e rapidamente ocorre a cópula. Nos bioensaios de atratividade, machos e fêmeas foram atraídos para fêmeas vivas no olfatômetro, porém não foram atraídos para machos vivos, o que sugere a presença de voláteis liberados pelas fêmeas para atração sexual. Foi constatada a presença de um composto fêmea-específico, identificado como 2-nonadecanona, com o possível papel de feromônio sexual. De acordo com os resultados, sugere-se que as fêmeas de G. spegazziniana liberam feromônios sexuais para atrair machos para o acasalamento. Os compostos liberados pelas fêmeas podem interagir com os voláteis emitidos pela erva-mate, resultando na atratividade dos machos de G. spegazziniana.
