Návrh opravy lesní cesty s využitím recyklovaných materiálů

Abstract The master’s thesis is divided into two parts. In the first one, there is theoretically described the issue of currently used technologies of the maintenance and repairs of the forest roads and then the issue of the use of recycled materials into the forest road structures with a focus on asphalt recyclates is discussed in detail. The second part of the thesis deals with the practical design of the repair of the forest road with asphalt recycled material. For the design of the repair the project documentation is elaborated to the required extent for the construction announcement. The thesis combines the modern concept of using recycled material with a practical design. The current trend of constant increase in prices of non - renewable resources and landfill is an impulse for the demand for recycled materials. This means that recycled materials will be still more used - not only for road construction sites, but also in other sites with the corresponding features.

Common mistakes Czech students make in English represent the main topic of the presented master thesis. The theoretical part mainly deals with the presentation of various TEFL (Teaching English as Foreign Language) methods. Moreover, they are discussed from several perspectives, including techniques, strategies and principles while using the particular method, the role of the teacher, his or her relationship with students, and especially the response to errors within each of the methods. The purpose of presenting TEFL methods is to show also the connection between mistakes and methods themselves. Concerning the area of mistakes and their corrections, we focus on the classification of mistakes and their division. The practical part is based on various illustrations of mistakes from students' writing assignments. These mistakes are divided into four major classes, including grammar, spelling, wrong word and word order. The thesis also deals with the remedial treatment which may hopefully reduce the number of possible mistakes. The aim of the thesis is to compare the level of students' English language knowledge at the Secondary School of Tourism in Prague where the author works as an English teacher.

However, although integrity constraints are defined for the given data, it is a common phenomenon that data violate the predefined set of constraints. Therefore detection of these inconsistencies and consecutive repair has emerged. This work extends and refines recent approaches to repairing XML documents violating defined set of integrity constraints, specif- ically so-called functional dependencies. The work proposes the repair algorithm incorporating the weight model and also involve a user into the process of de- tection and subsequent application of appropriate repair of inconsistent XML documents. Sustaining the integrity of DNA throughout the lifetime is critical for every living organism. Therefore organisms evolved numerous ways to detect and repair different types of DNA damage caused by various endogenous and exogenous factors resulting in replication stress. Defects in these repair mechanisms can lead to severe human diseases such as neurological disorders, familial cancers or developmental syndromes. In presented master thesis, we investigated the function of a yeast protein named Wss1, a metalloprotease that participates in a recently discovered DNA repair pathway that proteolytically removes DNA-protein crosslinks. Wss1 shows strong negative interaction with another DNA repair protease, Ddi1, in which case was discovered, that double-deleted yeast strain lacking WSS1 and DDI1 is hypersensitive to hydroxyurea. Hydroxurea is a ribonucleotide reductase inhibitor that, in the end, arrests cells in the S-phase of cell-cycle. Based on previous studies, we performed rescue experiments with various deletions and single-site mutants of Wss1p to assess the involvement of particular yeast Wss1p domains in the replication stress response to hudroxyurea.

Maintaining the chromosome continuity and complete genetic information in human cells is crucial for cell survival and the whole organism. It prevents life-threatening pathologies and preserves genetic continuity. However, cellular DNA is exposed to both endogenous and exogenous stress damaging its content and integrity. This stress activates mechanisms involving detection and repair of these damaged sites (DDR). One of the most serious types of DNA damage double-stranded breaks (DSB) occuring when both strands are severed. DSBs trigger wave of PTMs that regulate protein interactions, nuclear localization and catalytic activity of hundreds of proteins. Such modifications include acetylation, methylation, SUMOylation, ubiquitinylation and especially phosphorylation. The most important kinases involved in DDR kinases are ATM, ATR and DNA-PK. These kinases are activated immediately after the detection of the damaged area. DAXX (Death-associated protein 6) is an adapter and predominantly nuclear protein, which is involved in chromatin remodeling, gene expression modulation, antiviral response and depositing histone H3.3 variants into chromatin or telomeres. This thesis deals with the boundary between technical improvement and repairs of fixed assets. This work aims to describe and evaluate the selected decisions of the Supreme Administrative Court, primarily in the area of technical improvements and repairs and on the basis of these decisions determine the principles and parameters that should be determinig for taxpayers when they are assessing the technical improvements and repair on fixed assets. The first part is theoretically focused on legislation of technical improvements and repairs. In the following part are described and evaluated selected decisions of the Supreme Administrative Court, on which are subsequently drawn rules and parameters for assessing the boundaries between technical improvement and repair.

The theoretical part of the thesis entitled "The Required Energy of a Flying Object to Damage a Windscreen" covers the historical development of glass as a raw material and its further use as a key material for the automotive industry. In the following chapters, the thesis provides an analysis of a windscreen in general including the material composition of various windscreen manufacturers, the distribution according to the accessories used in the windscreens, and the various options of placing a windscreen in a car bodywork. The homologation of a windscreen and the procedure for replacing or repairing a windscreen after a damage is also covered. Lastly, the experimental part of the thesis describes the process of testing the required energy of a flying object to damage a windscreen.

The diploma thesis convey the information about the creation of technological procedures leading to the optimal way of repairing damaged parts of the fixture and creating production procedures for the production of new parts of the assembly. The theoretical part is focused on the analysis of the issue, determination of material of damaged parts for future production and designed semi-finished product. Furthermore, the technology of production from the point of view of used technologies is discussed. This includes a description of the tool materials. The practical part of the thesis is focused on the design of technological procedures for repairing damaged parts, which must be repaired as soon as possible. For the future production of new parts, the second half of the practical part includes the manufacturing processes including their description. There are evaluated variants of repair and production of new parts at the end of the thesis.
