The aim of this diploma thesis will be to create a plan for maintenance and repairs of the building on the street of the brothers Čapků in Brno. The first part will describe the individual concepts dealing with the life of individual structural elements, maintenance and repair and revisions of dedicated technical equipment. The second part will be focused on practice. A plan for maintenance and repairs of the building structures and a plan for revisions and inspections on the dedicated technical equipment will be prepared for the given building. The proposed plan will be designed to spread the funds over the whole period considered.

Postup opravy ropovodu při odstávce / Procedure of repair pipeline at dead plate

This thesis is focused on a crudeoil pipeline repair technology at the dead plate. The thesis is devided into 4 basic parts (chapters). The openning chapter describes reasons for performing regular pipeline inspections regarding pipeline's defects and technology of defects repair. The second chapter is devoted to the particular repair technology - cut out of a damaged pipeline part and its replacement by a new pipeline piece - with a detail description of preliminary and consenquential assembly activities (safety and environmental aspects). This chapter also comprises a list of documents confirming technical qualification and readiness for repairs execution. The third chapter is aimed at proposition and verification of the welding method by means of non-destructive and destructive testing of a test circumferential weld according to the proposed weld procedure specification – WPS. There are presented the results of particular tests and complex evaluation of the welding process design by the inspection organization. The last, fourth chapter deals with a calculation of general costs of repair execution which is an important part of a final pricing.

We studied the treebanks included in HamleDT and partially unified their label sets. Afterwards, we used a method based on variation n-grams to automatically detect errors in morphological and dependency annotation. Then we used the output of a part-of-speech tagger / dependency parser trained on each treebank to correct the detected errors. The performance of both the detection and the correction of errors on both annotation levels was manually evaluated on a randomly selected samples of suspected errors from several treebanks. Abasic sites (Ap site, from apurinic/apyrimidinic) are one of the most common lesions generated in DNA by spontaneous base loss or DNA repair processes. There are two equilibrating forms of an Ap site - ring-open aldehyde and cyclic hemiacetal. Ring- opened aldehydes are reactive electrophilic groups capable of formation covalent adduct with nucleophilic sites in DNA. DNA interstrand cross-link (ICL) resulting from the Ap sites is formed spontaneously as a covalent bond between ring-open aldehyde and amin group of adenin residue in the opposite strand of double stranded DNA. ICLs block DNA replication and transcription. The formation of Ap site derived ICL is relatively long process taking several hours. We assume that the ring-opening of an abasic site is the rate-limiting step in the formation of the thermodynamic ICL. However, formation, stability and DNA repair of Ap-ICL are still poorly understood processes. Here, I have set up mechanistic in vitro experiments to reveal and calculate the probability of Ap-ICl formation in vivo. In more detail, I study the rates of formation of Ap-ICLs in the sequence context of neighbouring nucleotides of freshly formed covalent bond of ICL. Automatická oprava pravopisu / Natural Language Correction

The goal of this thesis is to explore the area of natural language correction and to design and implement neural network models for a range of tasks ranging from general grammar correction to the specific task of diacritization. The thesis opens with a description of existing approaches to natural language correction. Existing datasets are reviewed and two new datasets are introduced: a manually annotated dataset for grammatical error correction based on CzeSL (Czech as a Second Language) and an automatically created spelling correction dataset. The main part of the thesis then presents design and implementation of three models, and evaluates them on several natural language correction datasets. In comparison to existing statistical systems, the proposed models learn all knowledge from training data; therefore, they do not require an error model or a candidate generation mechanism to be manually set, neither they need any additional language information such as a part of speech tags. Our models significantly outperform existing systems on the diacritization task. Considering the spelling and basic grammar correction tasks for Czech, our models achieve the best results for two out of the three datasets. In this thesis I deal with the passportization of the building-technical condition of the family house in Vyškov, the creation of its new project documentation and the proposal of repairs including appraisal cost. In the theoretical part I describe possible faults and defects of buildings, also their lifetime and the possibilities of finding their construction-technical condition. I also discuss the theory and usefulness of pasportizations. In the practical part, I evaluate the construction and technical condition of the selected house and make a finding of faults and defects of the structures, including the proposal for their repair and the appraisal cost with the item budget.

Genetic information must be protected, maintained and copied from cell to daughter cells, from generation to generation. In plants, most of the cells contain complete genetic information, and many of these cells can regenerate to a whole new plant. Such a feature leads to the need for precise control of which genes will be active and which not because in growth and differentiation, only the activity of specific genes for the individual cells, tissues, organs are required. One of the mechanisms controlling the gene activity is RNA interference (RNAi), which down- regulates or blocks the expression of specific genes at the transcriptional or post-transcriptional level. The crucial part of the RNAi is guiding the RNAi machinery to the target. It is mediated via sequence complementarity of the target with a small RNA (sRNA), which is diced from a double- stranded RNA (dsRNA) precursor. The molecular mechanism of dsRNA and sRNA formation and also the target origin predestinates the subsequent silencing pathway. In transcriptional gene silencing (TGS), the gene expression is regulated through chromatin epigenetic modifications. in english Introduction: Composite restorations in the oral cavity are exposed to an aggressive environment and mechanical challenge that gradually impairs their physical and mechanical properties. This may result in an enhanced wear rate, loss of esthetic properties and an increased risk of a restoration fracture or its marginal failure with a negative impact on the restoration's durability. Worn or failed restorations are usually completely replaced, which increases the irreversible loss of dental hard tissues. Repair of composite restorations by their partial replacement is therefore a minimally invasive, preventive and less time-consuming alternative to their complete replacement and increases their longevity. In the oral cavity, the adhesive bond between the existing composite restoration and the repair composite resin is exposed to various chemical substances and mechanical stress, e.g. surfactants in toothpastes, which can initiate its degradation. By decreasing the surface tension, the penetration of water into the adhesive joint can be enhanced, accelerating the hydrolysis of the adhesive and reducing the composite repair strength. Master thesis deals with the management of tangible fixed assets in the company Ecolab Hygiene s.r.o. which is engaged in purchase and sale of cleaning, desinfectant and sanitazing detergents, rental and repairs of cleaning machines and equipement. Introductory part is focused on theoretical bases. Second part of the thesis is focused on analysis of tanglibe fixed assets in the company and property maintenance. Based on analysis established knowledge there are subsequently weaknesses identified. In the final part solutions are proposed including their economic aspect whose implementation should have possitive impact on the current status improvement.
