Stanovení mikroflóry osiva vybraných druhů zeleniny a možnosti ochrany

The aim of this diploma thesis is assessing the determination and evaluation of pathogens species spectrum in carrot (Daucus carota), parsley (Petroselinum crispum) and pepper (Capsicum annum) seeds. The microflora was determined in laboratory conditions. The surface disinfected seeds and non-disinfected seeds were cultivated on culture medium PDA and filter paper. The level of bacterial and yeast contamination was low. The major part of pathogenswere fungi pathogens. The most often found pathogens were species from Alternaria and Cladosporium genus. The species spectrum was not different between individual varieties of carrot, parsley and pepper. In most cases, non-disinfected seeds had significantly higher amount of pathogens than the disinfected seeds.

The diploma thesis is focused on relationships between winter wheat pathogens occurrence, the weather and the microclimate in wheat stand. Attention was aimed at the incidence of Blumeria graminis, Mycosphaerella graminicola, Puccinia recondita f. sp. tritici, Pyrenophora tritici - repentis, pathogens from Fusarium genus and pathogens causing diseases of stalk bases. The evaluation was carried out in research station Žabčice during main part of growing season in the year 2015. Pathogens were determined and evaluated by degree of pathogen infection on five selected fungicidal untreated winter wheat varieties. At the same time the progress of vertical stratification of air and soil temperature in or under wheat canopy was evaluated, respectively. Those measurements were compared with temperature at the two closest climatological stations - in Žabčice and Brno - Tuřany.

This thesis is devoted to the specification of individual hazards that are most frequently encountered in catering . Attention is focused on biological hazards, mainly bacteria, which can become a deterioration of foods and their pathogenicity. In order to prevent the hazard, it is necessary to manage the associated legislation. Sanitation process is validated by microbiological analyzes. Cultivation swabs were focused on the detection of Enterobacteriaceae, the total number of microorganisms, fungi and yeasts groups coliforms, Listeria monocytogenes, Salmonella ssp. The methods of detection were cultivated on nutrient media according to ISO standards for the detection of coliform group, Listeria monocytogenes and Salmonella ssp were used for pathogen detection kits ( Path- Chek Hygiene pathogen detection). Using these methods, errors have been repeatedly detected in finding sanitation such as coliform bacteria, the family Enterobackteriaceae, and Listeria monocytogenes. Statistically, the most risky areas of the kitchen in terms of the total number of microorganisms was a kitchen mixer. In terms of frequency of occurrence of the family Enterobacteriaceae the kitchen mixer was the most risky area as well. Possible contamination routes have been the working surfaces of kitchen sponges and cloths.
