The relation of adult attachment security to changes in maternal parenting behaviors : a parenting intervention study

The goal of the current study was to examine the impact of mothers’ attachment classification on their ability to change their parenting beliefs and behaviors over the course of a parenting intervention program. Results indicated that in large part, this study did not support the idea that secure mothers would benefit more from a parenting intervention program than insecure mothers. However, treatment group placement was found to moderate the extent to which attachment security and time interact on level of permissiveness. Specifically, insecure mothers in the seminar plus hands-on condition significantly decreased in their permissiveness over time. Thus, insecure individuals benefit from parenting intervention programs when they have the opportunity to practice as well as learn the material presented to them. / text

Pochopení tolerance korupce: záleží na osobnostech? / Understanding Corruption Tolerance: Does Personality Matter?

Attitudes towards corruption have been attributed to a broad range of macro-level and, to a limited extent, micro-level antecedents. Neglected is academic research on the effect of individual-level psychological factors on the attitudes toward corruption. To fill this gap, this article aims to explore the impact of individuals' personality traits on their willingness to justify corrupt acts, drawing on data from World Values Survey Wave 6. By applying fixed effects models, the study finds that two of five personality traits - consciousness and agreeableness are significantly and negatively associated with individuals' willingness to justify corrupt exchanges. Additionally, the study results show that the association between the personality traits and corruption tolerance varies from country to country: openness to experience, for instance, was significantly and negatively associated with corruption tolerance in Germany, whereas this trend was not observed in the Dutch sample. Among the control variables, age and sex were significant predictors of corruption tolerance as well as the Dutch were less willing to justify corrupt acts than Germans. The study provides empirical and practical implications as well as suggestion for future research.

Religious Affiliation and Sexual Permissiveness Over Time

In this study, I analyze the relationship between sexual permissiveness and affiliation with a fundamentalist religion and how this relationship has changed over time. I first consider previous research that reviews how religious affiliation, religiosity, and religious fundamentalism shapes sex attitudes and, therefore sexual permissiveness. I then review existing studies that discuss what factors influence permissiveness toward different sexual behaviors. Next, I discuss the mechanisms of religious institutions that influence sexual permissiveness. Prior literature motivates my research question as there is a lack of studies that explore sexual permissiveness across religious affiliations. This study fills a void in the existing literature by exploring the gap in sexual permissiveness between religious affiliations and how that gap has changed over time. After considering the current literature, I introduce a hypothesis exploring the relationship between sexual permissiveness and affiliation with fundamentalist religion. This study performs OLS regression using secondary data from the General Social Survey (GSS) that describes respondents' religiosity, religious affiliation, and attitudes towards sex. The study's findings show that affiliation with more fundamentalist groups is significantly correlated with more conservative sex attitudes reflecting lower levels of sexual permissiveness. The results also suggest that the gap in sexual permissiveness between those that affiliate with liberal, moderate, and fundamentalist religions has become larger over time.

Coxiella burnetii é a bactéria intracelular causadora da Febre Q, capaz de subverter funções celulares e evadir o reconhecimento do sistema imune da célula hospedeira permitindo o estabelecimento do seu nicho replicativo nas células-alvo: macrófagos e monócitos. É um patógeno altamente virulento, sendo necessário poucos organismos para desencadear a doença, e é considerado como um potencial agente de bioterrorismo da categoria B. Entre os danos econômicos causados por C. burnetii destaca-se a infecção de animais de gado, seu reservatório natural, pois causa aborto espontâneo dos filhotes e, consequentemente, prejuízo aos produtores. Seres humanos também adquirem a infecção, por inalação de partículas contaminadas. Hospedeiros imunocompetentes são capazes de restringir a infecção por C. burnetii apesar dos diversos mecanismos de evasão da resposta imune do hospedeiro, como interação com vias de sinalização celular e utilização de efetores bacterianos. No entanto, em hospedeiros não competentes a infecção pode evoluir para casos de Febre Q crônica e levá-los à morte. Modelos murinos são frequentemente utilizados para entender as interações patógeno-hospedeiro nas infecções por essa bactéria. A diferença na suscetibilidade de macrófagos de diferentes linhagens murinas e de diferentes tipos celulares à infecção por C. burnetii ainda é pouco conhecida. No entanto, sabe-se que C. burnetii fase II sucumbe a macrófagos derivados da medula óssea (BMDMs) de camundongos C57BL/6, enquanto células das linhagens BALB/c e A/J são suscetíveis à infecção. Nesse contexto, considerando a relevância biomédica de C. burnetii, faz-se necessário a determinação de um modelo relevante para melhor compreender as relações patógeno-hospedeiro nas infecções por essa bactéria. Neste trabalho, caracterizamos um novo modelo de estudo com macrófagos primários que permite avaliar a infecção com C. burnetii fase II in vitro, além de elucidar os mecanismos relacionados à suscetibilidade das células à bactéria. Por meio de quantificação do DNA genômico bacteriano por qPCR verificamos que macrófagos alveolares (AMs) murinos são altamente suscetíveis à replicação da bactéria, mesmo no fundo gênico restritivo C57BL/6. Caracterizamos a replicação da bactéria em AMs por microscopia de fluorescência e microscopia eletrônica de transmissão e demonstramos que essa replicação ocorre dentro dos vacúolos parasitóforos típicos. Pela análise de expressão gênica por RT-PCR efenotipagem de marcadores de superfície por FACS identificamos que a alta suscetibilidade dos AMs se deve a uma polarização dessas células para um padrão M2. Por fim, validamos a relevância desse modelo pela análise da suscetibilidade de AMs murinos deficientes para NOS2, IFN-? e IL-4 como prova de princípio. Verificamos que a suscetibilidade de AMs é comparável à de células Vero e células THP-1, modelos celulares imortalizados descritos como altamente suscetíveis à replicação de C. burnetii, e que AMs podem ser utilizados para estudos utilizando células derivadas de camundongos com fundo gênico C57BL/6. Dessa forma, nosso trabalho caracterizou os AMs murinos como um relevante modelo celular primário altamente suscetível para o estudo das interações patógeno-hospedeiro frente à infecção in vitro por C. burnetii fase II, além de elucidar os mecanismos por trás dessa suscetiblidade / Coxiella burnetii is the intracellular bacterium that causes Q fever, capable of subverting cellular functions and evading recognition by the host cell immune system, thus allowing the establishment of its replicative niche in its the target cells: macrophages and monocytes. It\'as a highly virulent pathogen, with few organisms being sufficient to cause the disease, and considered a potential class B bioterrorism agent. Among the economic damages caused by C. burnetii are infection of cattle animals, its natural reservoir, that leads to spontaneous abortion of the offspring and financial loss to the producers. Human beings also acquire the infection, through inhalation of contaminated air particles. Immunocompetent hosts are able to restrict infection by C. burnetii despite the several evasion mechanisms from the host immune system, which includes interaction with cell signaling pathways and utilization of bacterial effectors. However, in non-competent hosts the infection can evolve to chronic Q Fever and lead to death. Murine models are frequently used to understand the host-pathogen interactions during infections by this bacterium. The difference in the susceptibility among macrophages from different murine strains, as well as different cell types, still poorly known. However, it is known that C. burnetii phase II succumbs to bone-marrow derived macrophages (BMDMs) from C57BL/6 mice, whereas cells from BALB/c and A/J strains are susceptible to infection. In this context, considering the biomedical relevance of C. burnetii, it\'s necessary to establish a relevant study model to better understand the host-pathogen relations in infections by C. burnetii. In this work, we characterized a new study model with primary macrophages to assess the infection by C. burnetii phase II in vitro and elucidated the mechanisms underlying the susceptibility to C. burnetii. By using qPCR for quantification of bacterial genomic DNA, we saw that murine alveolar macrophages (AMs) are highly susceptible to C. burnetii replication, even in the usually restrictive C57BL/6 backgound. We characterized the bacterium replication in murine AMs by florescence microscopy and transmission electron microscopy, showing that this replication occurred inside the typical C. burnetii-containing vacuole. We also identified, through the gene expression by RT-PCR and the phenotypic profile of surface markers by FACS, that the increased susceptibility of murine AMs were due to a polarization of these cells into a M2 profile. To finish, we validated the relevance of thismodel through the analysis of the susceptibility of murine AMs deficient to NOS2, IFN-? and IL-4 as a proof of principle. We verified that the susceptibility of AMs is comparable to Vero cells and THP-1 cells, immortalized cell models described as highly susceptible to C. burnetii replication, and that AMs can be used to studies using cells derived from mice with the C57BL/6 background. In this way, our work characterized murine AMs as a relevant primary cell model highly susceptible to studies among the host-pathogen interactions in infections with C. burnetii phase II in vitro, and we also elucidate the mechanisms underlying this susceptibility

Para verificar a permissividade de células de origem hematopoiéticas à infecção por HAdV-41, foram infectados PBMC e IEL de voluntários. Os ensaios foram comparados com células HEK-293 infectadas. Foram analisadas as expressões dos genes virais E1A, E1B (55K), E3 (14K), VARNA, hexon e fibra curta (FC) e do gene celular GAPDH. O mRNA foi detectado por RT-PCR em tempo real e a produção de proteínas foi visualizada por IFI. Em HEK-293, a transcrição dos genes E1A, E1B e E3 iniciou-se às 11h p.i, hexon,às 13h pi.,VARNA e FC às 14h p.i. Em PBMC a transcrição de E1A, E1B e VARNA iniciou-se 17h p.i e a expressão dos genes hexon e fibra curta foi detectada 18h p.i e 20 h p.i. respectivamente. O nível de expressão dos genes virais em HEK-293 foi quase 200 vezes maior em relação à PBMC. Os IELs também mostraram-se permissivos à infecção pelo HAdV-41 como mostrado pela expressão dos genes virais. Essa é a primeira evidência de que este vírus possa infectar tais células. Os resultados obtidos ajudam a elucidar os mecanismos de interação do vírus com a célula-hospedeira. / In order to verify the permissiveness of hematopoietic cells to HAdV-41 infection, PBMC and IEL from volunteers were infected. PRION CHARACTERIZATION USING CELL BASED APPROACHES

Prions are the causative agents of a group of lethal, neurodegenerative conditions that include sheep scrapie, bovine spongiform encephalopathy (BSE), and human Creutzfeldt-Jakob disease (CJD). Prions are derived from the conversion of a normal, primarily alpha-helical, cellular prion protein (PrPC), to an infectious, beta sheet-rich conformer (PrPSc). Many unresolved issues surround the process of PrP conversion, and we know very little about cellular responses to these unique pathogens. Our lack of knowledge relates, in part, to the difficulty of infecting cells in vitro with prions. While expression of PrPC is an absolute requirement for prion propagation, I show here that not all cells that express PrPC are capable of propagating PrPSc. The goal of this thesis is to understand the role that host factors play in sustaining prion infection and to develop systems in which the cellular response to prion infection can be assessed. We hypothesize that cellular permissiveness to prion infectivity is co-dependent on unidentified additional cellular factors. To study the role of PrPC expression in susceptibility to prion infectivity, and identify these cofactors in cell culture, we utilized cells which fail to express endogenous PrPC, but become susceptible to prions following stable expression of PrPC. Following transfection of a species specific PrP expression construct and isolation of single cell clones, we assessed PrP expression and susceptibility to prion infectivity by measuring accumulation of protease resistant PrPSc. Differential gene expression studies suggest significant transcriptional differences between susceptible and resistant clones. Using three independent gene expression databases our analyses suggest that the resistant transcriptional profile favors cell division/cycle and chromosomal regulation pathways, while the sensitive transcriptional profile is involved in protein homeostasis and quality control. The results of these studies will not only lead to a greater understanding of PrP cell biology and the mechanisms of prion pathogenesis, but should ultimately lead to sensitive and expedient methods for detecting and characterizing prion infectivity from a wide range of sources.

Contemporary attitudes in affluent Western societies are characterised by a growing emphasis on individual freedom. What, then, does this commitment to liberty entail for our openness to diversity; and ultimately for liberal democracy? Previous research on popular attitudes, for example by Ronald Inglehart, tends to assume that valuing freedom entails an encouragement of a plurality of life-styles. This thesis, by contrast, argues that there are several ideals of freedom in public opinion; ideals that may have opposing consequences for our permissiveness towards ways of life that differ from our own. The introductory essay in this book suggests that Isaiah Berlin’s theory of positive and negative freedom provides a fruitful analytical framework, which helps theorise and empirically nuance our picture of popular ideals of freedom. Essay I goes on to present a novel, psychological, interpretation of Berlin’s Two Concepts of Liberty. This essay also suggests that Berlin was critical not only of enlightened ideals of positive liberty, but also of romantic ones, which might be even more widespread today. Essay II then applies Berlin’s framework to contemporary survey data. Through confirmatory factor and regression analyses, this essay demonstrates that Berlin’s negative-positive distinction does in fact hold also in popular opinion; and that the two dimensions have rather different effects on moral and legal permissiveness. Essay III, finally, revisits a recent example of disrespect in the name of liberty: the Danish cartoon controversy. This essay develops the concept of ‘romantic liberalism’, thereby deepening our knowledge of romantic ideals of positive liberty, and their particularly disrespectful tendencies. Drawing on Isaiah Berlin, and his critique of positive liberty, the essays in this thesis together suggest that it is crucial for liberal democracy to recognise the existence of treacherous liberties: ideals that lead their supporters to ridicule, condemn, or even prohibit ways of life that differ from their own – all in the name of liberty. / The Impact of Religion

This study sought to articulate the constructs corruption and governance, to support research to adopt governance practices contributes in the fight against corruption in sport. Having as object the athletics, athletes and coaches were asked about how effective these measures would be, in your opinion, if adopted by the national body sport administration to which they are affiliated. In addition to this question at the operational level, it was also prepared a checklist, based on the survey of Marques and Costa (2009) and documentary analysis of the organization, which was confronted by the administrative direction of the organization, representing the strategic level of athletics. At the same time, it was also asked to athletes and coaches, their degree of permissiveness to corrupt actions as Lavena (2013) protocol. The study used the qualitative method at a stage in advance, exploratory and quantitative in the following, descriptive character. The collected data were treated by analysis of variance to find significance in the effectiveness of the actions and are described and presented in the results, which showed that between governance dimensions, only three of them (ownership, board of directors and independent audit) figured degree of statistical significance needed to be considered effective in combating certain corrupt actions in the sport, according to the procedure used, in the opinion of the athletes and coaches of athletics in the sample under the conditions tested in this study. / Comportamentos éticos são de grande importância na condução de negócios entre quaisquer empresas e são essenciais à própria interação entre os indivíduos na sociedade, em que ocorrem os comportamentos corruptos, que podem ser assim considerados não legítimos do ponto de vista ético e por isso, combatidos, inclusive no esporte. Esse trabalho procurou articular os construtos corrupção e governança, para apoiar a investigação se a adoção de práticas de governança contribui no combate à corrupção no esporte. Tendo como objeto o atletismo, atletas e técnicos foram inquiridos sobre quão efetivas seriam essas medidas, em sua opinião, se adotadas pela entidade nacional de administração do esporte a que estão filiados. Além desse questionamento ao nível operacional, também foi elaborado um checklist, baseado no levantamento de Marques e Costa (2009) e análise documental da entidade, que foi confrontado pela direção administrativa da entidade, representando o nível estratégico do atletismo. Paralelamente, também foi inquirido aos atletas e técnicos, seu grau de permissividade a ações corruptas, conforme protocolo de Lavena (2013). O estudo utilizou o método qualitativo numa fase a priori, exploratória e o quantitativo na seguinte, de caráter descritivo. Os dados levantados foram tratados por análise de variância para buscar significância na efetividade das ações e são descritos e apresentados nos resultados, que demonstraram que entre as dimensões de governança, apenas três delas (propriedade, Conselho de Administração e auditoria independente) figuraram com grau de significância estatístico necessário para serem consideradas efetivas no combate a determinadas ações corruptas no esporte, de acordo com o procedimento utilizado, na opinião dos atletas e técnicos de atletismo que compuseram a amostra, nas condições testadas neste estudo.

Western society has undergone a major developments in human values, social norms and the degree of tolerance to various manifestations of human behavior after the World War II. and this development naturally continues until today. With this is connected a permissive society, society with so free normative structure, that it tolerates almost all kinds of social behavior. For some people is exaggerated liberality harmful and threatens normative system and values asociated with moral decline. The thesis will be about permissiveness and tolerance to some kinds of people's action in Czech republic. At the very beginning of the theoretical part I will explain the basic concepts. Then I'll make a probe into the past and explain social revolution which led to formation of permissive society with all important causes. After that I'll focuse on main symptoms of permissive society. Theoretical part will be ended by historical look at the conditions for permissiveness in post-war Czech. In analytical part I will use data from European values study, my based on that quantitative research from year 2014 and other available data. There will be general anaylisis about development of permissivenes since 1991 till today. My 2014 data will be also used in more detailed analysis using sociodemographic characteristics. Powered...
