Organohalogenové sloučeniny - kontaminanty životního prostředí / Organohalogenic compounds - contaminants of environment

Polybrominated diphenyl ethers belong to group of brominated flame retardants. Important property of these substances is their ability to reduce flammability or to prevent ignition of many products of daily use, such us televisions, carpets, mattresses and many others. Polybrominated diphenyl ethers however are characterized by high persistence and lipophility, leading to their accumulation in the environment. In recent years, concerns about the effects of organic halogen compounds on human health, suggesting their inclusion in the list of persistent organic pollutants in 2009. These contaminants are capable of long-range transport and accumulation in the environment. Therefore, the presence of polybrominated diphenyl ethers is systematically observed in the abiotic and biotic components of the environment worldwide. Their presence has been demonstrated in areas that are clearly very distant from sources of contamination. This thesis is focused on the use of gas chromatography to assess the level of contamination of selected components of the environment, especially of soil. For these purposes was used method of gas chromatography with electron capture detection mass spectrometry.

This diploma thesis deals with reducing the time of sucking water into the rotating impeller of the centrifugal pump as used in firesport. The suction is carried out by the ejector, which is powered by combustion engine exhaust gases. A mathematical model was created, which shows the flow in the ejector. It was also used as a basis for the creation of a new ejector, which has an increased airflow when getting closer to atmospheric pressure.

EN The thesis is focused on a speciation analysis of arsenic with use of the hydride generation - cryotrapping - gas chromatography - atomic absorption spectrometry. The aim of this project was the development of a method and instrumentation of the speciation analysis of arsenic based on combination of selective generation of substituted hydrides with a detection by atomic absorption spectrometry and with use of the cryotrapping of generated arsenic species and their separation by the gas chromatography. In the first part of this work the effects of individual parameters which influence the separation in the gas chromatograph were studied. Especially, time of injection from the cryotrap into GC, temperature program, carrier gas flow and carrier gas flow through a "deanswitch". Optimization of these parameters is an attempt to achieve a satisfactory resolution of the individual peaks of arsenic species separated by gas chromatography. Furthermore, an adequate sensitivity of this method is required to be achieved in comparison to other methods that use AAS detection such as a combination of hydride generation with cryotrapping. Vývoj miniaturizované extrakční metody pro screening netěkavých nitrososloučenin ve sladu pomocí GC-NCD / Development of miniaturized extraction method used for GC-NCD screening of non-volatile nitroso compounds in malt

The aim of this diploma thesis was to develop a miniaturized extraction method for a fast screening of non-volatile nitroso compounds using gas chromatography with a nitroso specific chemiluminescence detection. According to a final methodology, the samples were prepared by extraction of grinded malt using a mixture of pyridine and acetonitrile in ratio 60:40 (v/v). To enhance volatility of the determined analytes, the two-step derivatization using hexamethyldisalazane and N,O-bis(trimethyl)-trifluoroacetamide was used. The total volume of the sample was 200 l and the preparation time after optimization was in total 80 min. The extraction method was connected to a classification method, which can divide chromatographic peaks into the groups of N-nitroso and C-nitroso compounds, and interfering substances. After application of the methods mentioned above to real malt samples, the specific chromatographic peaks of C-nitroso and N-nitroso compounds were selected. Alkyl derivatives of mercury, especially methylmercury, bring a significant risk to the environment and the human population. Methylmercury is highly toxic compound, which have the potential to bioaccumulate and biomagnify in tissues of aquatic organisms. This diploma thesis deals with issues of alkyl derivatives of mercury in biotic matrices. The goal of this thesis was to analyze fish tissues. Fish were caught from the river Svratka at the inflow and at the outflow of wastewater treatment plant. Methylmercury was determined in fish muscles and skin. Total mercury was determined in fish muscles. The extraction based on acidic digestion in hydrochloric acid and subsequent extraction with toluene was used for isolation of methylmercury from fish tissues. There was used gas chromatography with electron capture microdetector for determination of methylmercury. Total mercury was determined by special analyzer AMA 254.

This dissertation thesis deals with the diagnotics of power oil transformer. It consists of the theoretical and practical part At its beginning, the theoretical part deals with an general description of the transformer and its connection. Focus is given to aspects of the life of the transformer insulation system (oilpaper). Further, the technical diagnostics is analyzed, with description of quantities of gasliquid chromatography, the oil condition and condition of conductor insulation. Another important part of the thesis consists of an expert system, which is described generally from the past to the present. Characteristics, types and structure of expert systems are mentioned here, with its advantages and disadvantages. The practical part starts with a description of the data file, which is further used in the thesis. Main practical part is divided into three parts - the results of gas-liguid chromatography, selected results and expert systems. In the section entitled "Results of gasliquid chromatography", there is an analysis of selected properties of time series and temperature dependency of gases. Moreover, the methods of the ratios of gases such as Duval Triangle method are described. The second part entitled "Selected other results" shows the time dependency of variables and the neutralization number, along with the breakdown voltage dependency, and time of dependency, gas and moisture content in insulating oils. There is also a temperature dependency of moisture content dissolved in insulating oil discussed, as well as results of selected correlations, such as the correlation values of gas chromatography with state variables insulation of wires. The last part, "Expert systems", provides specific information about developed expert system. It describes the environment chosen for its development, followed by a description, features and verification of the expert system. Examples of the consultation results are included. All achievements of the individual parts are always summarized at the end of each section. The thesis contains chapters "Objectives of the work" and "Conclusion". The most important chapter is "The contribution of the work." It contains original work results and the results applicable in industry.

Charakterizace interakcí fluorované stacionární fáze Rtx-200MS s analyty metodou inverzní plynové chromatografie / Characterization of interactions between Rtx-200MS fluorinated stationary phase and analytes by inverse gas chromatography

Fluorinated stationary phase in Rtx-200MS column have been characterized by determination of system constants of Abraham equation. Retention on this phase is driven by dispersive and orientation/induction forces. Significant interaction contribution of lone pair electrons or π- electrons provides unique selectivity for analytes with excess of electron density. Unusual behavior of this phase have been determined by study of separation mechanism of polar and nonpolar analytes, in comparison of their separation on polar and nonpolar phases. This behavior is due to medium polarity of the phase (system constant s), which is not so pronounced to cancel separation of nonpolar analytes due to induction forces. In some cases contribution of lone pair electrons or π-electrons can contribute to this separations. This Master's thesis deals with determination of nucleobases in formamide samples (pure or catalyzed form) after their initiation by high-power density energy events. The theoretical part states the reader to the problems of prebiotic chemistry and acquaints him with the various theories of the origin of the nucleobases. The experimental part deals with the analysis of the samples initiated by Laser System PALS, which was used for the simulation high-power density energy events. For the identification and determination of the final products of laser plasma initiated chemical reactions in the gaseous phase of the samples was used the Fourier Transform Infrared Spectroscopy. The liquid part of the samples and the nucleobases in this part were analyzed by the gas chromatography with the mass detection.

This thesis deals with the Czech Presidency of the Council of the European Union in the first half of 2009 in terms of energy and energy security. In the introductory theoretical part the notion of "energy security" is defined and the chapter analyzes energy in the EU primary law too, including jurisdiction of the issue to a particular Union's institutions. In the practical part of the thesis the priorities of the EU in energy and energy security are described and steps that CZ PRES made to increase EU energy security are analyzed. Particular emphasis is placed on the gas crisis and the general supply of gas, which played during the Czech Presidency a significant role. Final chapters focus on the shift in the EU's energy security during the forthcoming Swedish and the Spanish Presidency.

Bumblebees are important pollinators, commercially used in large-scale plant growing in greenhouses. Their males produce marking pheromones for mating, which attract young bumblebee queens. These pheromones are often a complicated mixture of chemicals, which is produced in the male labial gland, and the mixture itself is specific for each bumblebee species. The regulation of bumblebee sexual pheromone biosynthesis is largely unknown, and this Master's thesis is focused on the analysis of the mechanisms which lead to the regulation of the Bombus terrestris male pheromone's fat-acid and terpenes biosynthesis, specifically on stereospecific enzymatic reduction of double bond of farnesol. This thesis studies the influence of potential neurohormones on a specific enzymatic mixture, which is involved in the pheromone biosynthesis. Methods used in this project include biochemical, analytic and molecular-biology methods.
