Povrchové plazmonové rezonance na koloidních nanočásticích / Surface Plasmon Resonances on Colloidal Nanoparticles

The presented diploma thesis is focused on the Localized Surface Plasmons (LSP). The far-field optical response of the colloidal solutions of gold nanoparticles caused by LSP was investigated and compared with the numerical calculations. For the simulations, the Discrete Dipole Approximation (DDA) and Finite-Difference Time Domain (FDTD) techniques were employed. In particular, the shape and size effects of spherical particles and nanorods were studied. The simulations performed by both methods are in a good agreement for the spheres. For the nanorods, the resonance was found to be affected markedly by their geometry. Also, broader resonance peaks were found. This effect was assigned to the sample size distribution and its influence is discussed by comparing the simulations with experiments. In addition, synthesis of nanorods was carried out as well. Finally, the results on the study of optical properties of silver clusters formed under equilibrium conditions are presented.

Interakce hyaluronanu a povrchově aktivních látek / Interactions between Hyaluronan and Surface Active Substances

Vliv hyaluronanu na micelizaci tenzidů byl studován různými fyzikálně-chemickými metodami. Byly zvoleny dva kationaktivní tenzidy, a to tetradecyltrimethylammonium bromid (TTAB) a cetyltrimethylammonium bromid (CTAB). Metoda izotermické titrační kalorimetrie byla využita pro stanovení entalpie micelizace, tenziometrie popisuje povrchové vlastnosti daných vzorků. Byl sledován také vliv různé molekulové hmotnosti použitého hyaluronanu a délka alkylového řetězce tenzidu na agregační chování daného systému. Výsledkem jsou hodnoty kritické micelární nebo agregační koncentrace tenzidu. V neposlední řadě se diskutuje využití agregátů hyaluronan-tenzid jako možné nosiče pro cílenou distribuci léčiv.

Essential part of the thesis is concerned with calculations and variant solutions of the object reconstruction and water treatment technology. Technical drawings of the water treatment plant are included. All the data was processed in accordance with valid laws and standards. The water treatment plant will be restored to its functional state on the grounds of the designed solution.Therefore permanent high quality of drinking water will be guaranteed for the inhabitants of Kateřinice.

Theranostické systémy v sonografii / Theranostic systems in sonography

This work deals with preparation of microbubble suspension from a mixture of phospholipids, palmitic acid and polyethylene glycol. Properties of prepared systems were studied using bubble tensiometry and dynamic light scattering method and were compared with commercial contrast agent SonoVue®. Suspensions were prepared in various conditions including different atmosphere and increased temperature in some steps of preparation and different solution. Effect of polyethylene glycol addition on surface activity of the system was studied. Surface activity of phospholipids was insignificant. Surface tension decreased with increasing concentration and molecular weight of polyethylene glycol in the system. Effect of different atmosphere and increased temperature showed no substantial trend. It emerged that dynamic light scattering is not suitable for this type of samples because of high polydispersity and phase separation of the system.

Vliv povrchových rýh na přechod ke smíšenému mazání / The effect of surface grooves on transition to mixed lubrication

The purpose of this diploma thesis is to describe the influence of artificially produced surface grooves on transition from full EHL regime to mixed lubrication in point contact using optical interferometry. This article focuses on influences of grooves orientation, length in contact area, elastic properties of contact surfaces and different types of oil defined by pressure-viscosity coefficient. Empirical prediction model of groove influence on film thickness was established based on experimental data. The groove influence was incorporated into lubrication parameter. This modified definition was compared to classic one. It showed that avoiding groove influence in classic definition lead to significant underestimation. Influence of groove on friction coefficient is also studied.

The presented master’s thesis deals with antennas transmitting energy along conductive materials. Surface waves are used for energy transmission. According to its properties, the horn antenna was chosen for the problem solution. This thesis includes the theoretical analysis of surface waves, waveguides and horn antennas. The concept of antennas includes the comparison of two different materials and two different frequencies. From the set of 8GHz antennas, the structure of two antennas where one wearing the other was chosen as the most suitable. The FoamClad material was also selected due to achievement of better parameters and properties then material CuClad. This structure was designed by two identical horn antennas where the energy between them is transmitted via slot. The suitable feeding and parallel plates were designed, and further the current structure was modified to SIW structure. The designed antenna was manufactured and its properties were measured for further comparison with simulation results. Furthermore, achieved differences between simulated and real antenna are discussed.

The genus Candida includes several opportunistically pathogenic species which are common causative agents of the yeast infections in humans. Although current medical research is focused mostly on cancer, AIDS or Alzheimer disease, the problem of systemic candidiases cannot be neglected. These infections represent a real threat to the immunocompromissed patients, they are connected with a high mortality rate and expensive medication with poor prognosis. Pseudomonas aeruginosa could be an inspiration in a way of how to eliminate the pathogenic yeasts. The bacterium can inhibit growth of the most common yeast species of the genus Candida, C. albicans. This effect is based on production of toxic substances by the bacterium and on interaction of the bacterium with the C. albicans cell wall, which leads to the lysis of the yeast cells and which is not fully understood. Nevertheless, coexistence of these microorganisms is also possible and their relationship is affected by various factors. Knowledge of these inter- microbial interactions was obtained from studies of diseases and pathologies, during which C. albicans + P. aeruginosa coinfections occur. Binding of sperm to the oocyte is mediated by complementary molecules on the surface of both gametes and involves the interaction of sperm protein receptors with the zona pellucida (ZP) saccharide structures. It has been shown that many proteins receptors are involved in the sperm-ZP interaction, and potential primary sperm receptors for ZP glycoproteins have been investigated in various mammals. The majority of proteins with identified sperm-ZP binding activity belong to the plasma membrane proteins. However, the exact methods for isolation of sperm membrane proteins are still to be standardized. This study is focused on investigating how employment of various isolation protocols leads to acquisition of various protein mixtures. Further in the work, two possible approaches towards identification of potential ZP-binding partners are implemented, in order to disclose novel primary ZP-binding receptor candidates. Sperm proteins of ejaculated and in vitro capacitated boar sperms were isolated by: Triton X-100, Triton X-114, acetic acid, sodium dodecyl sulphate (SDS), N-octyl-β-D- glucopyranoside (OBG), rehydration buffer (RHB), and finally by freezing-thawing extraction and they were characterized by 1-D, 2-D protein profiles, glycoprotein staining and substrate zymographic methods. Studium horninových struktur v Českém ráji pomocí seismického šumu / Ambient noise investigation of rock structures in Bohemian Paradise

We carry out non-invasive ambient noise investigation of rock structures in Bohemian Paradise (Bohemian Cretaceous Basin, Czech Republic). The study is focused on two key topics: 1) An in-situ elastic moduli estimate of competent, horizontally deposited sandstone layers. This is done by performing an ambient noise array measurement. The recording is processed with f-k array analysis, from which frequency-dependent Love and Rayleigh wave dispersion curves as well as the Rayleigh wave ellipticity are retrieved. The data are inverted for P- and S-wave velocity profiles, from which the Young's and shear modulus are successfully estimated. 2) Study of local response of Kapelník rock tower. We analyse a dataset of ambient noise recordings from the top of the tower and from its foot. Information about tower oscillation frequencies and directions, together with amplification ratios, are retrieved from particle motion polarisation analysis and from site-to-reference spectral ratios. The Euler-Bernoulli beam theory is finally used to interpret the measured data using the elastic moduli estimated from the noise array measurement. Method: The research for this thesis involved non-experimental pre-research and quasiexperiments. The thesis' theoretical hypothesis was developed on the basis of previous studies that had looked at the effect of brain damage on patients' upper limb function. A total of twenty patients between the ages of 20 - 59 were selected to participate in the empirical study reported in this thesis. All participating patients had suffered damage to their cerebral cortex. The causes of their brain damage included strokes, trauma and tumor. The participants were divided into two groups of ten using the Nottingham Sensory Assessment. Participants with sensory disorders were allocated to the experimental group. Participants without sensory disorders were placed in the control group.
