Molecular systematic studies in the genus Ribes (Grossulariaceae)

Infrageneric classification in Ribes has previously relied on a few, often conflicting, morphological markers, such as spines, glands, and inflorescence morphology. Suggestions that hybridization drives the evolution of the genus have not been tested using phylogenetic methods. To assess the validity of infrageneric classifications and the importance of hybridization to the evolution of the group, and to develop an explicit phylogenetic hypothesis, I surveyed exemplars from all subgenera for restriction site variation in two cpDNA regions. Parsimony analysis shows that red currants, European alpine currants, golden currants, true gooseberries, and western gooseberries appear on separate clades. A less well supported Glade includes the western North American ornamental currant group and a portion of the dwarf currants. The presence of distinct lineages in Ribes is strongly supported by characters with very high consistency, suggesting that hybridization among infrageneric groups is not common in the genus. Unexpectedly, spiny currants and true gooseberries are united, suggesting either a sister group relationship or the possibility that one of these groups arose by reticulate evolution. The four black currant species examined exhibit surprisingly high divergence, and are not monophyletic in the analysis. Maximum likelihood analysis supports these results. Basal relationships of these lineages are not well resolved. A similar analysis of a portion of the nuclear ribosomal repeat produced very few characters. Although these data are highly homoplasious, their analysis bears some resemblance to that of the chloroplast DNA. Most prominently, the ornamental currant Glade has identical membership. Brief reviews of the scattered palynological and paleontological literature concerning Ribes are presented. / Graduation date: 1995

Įvertinus 12 juodųjų serbentų (Ribes nigrum L.) veislių ir klonų derlių nulemiančių svarbiausių komponentų įtaką: atsparumą šalčiui, grybinėms ligoms, serbentinei (pumpurinei) erkutei, lapų žiotelių parametrus ir nustačius jų tarpusavio ryšį – išskirtos veislės tinkamos mechanizuotam derliaus nuėmimui, vertingi agronominiu požiūriu donorai, naujoms veislėms kurti. Veislė Tines yra vėlyvo žydėjimo. Veislės Titania, Tiben ir klonas 98 – 279 – 25 darnaus uogų sunokimo donoras. Mechanizuotam uogų skynimui tinkamiausios veislės Tines, Ruben ir Dailiai. Daugiakiakiškumu išsiskyrė veislės Ores, Ceres, Ruben ir klonas 98 – 279 – 25. Mažiausias sėklų procentas uogoje veislių Titania ir Tisel. Veislės Tines, Bona, Ruben, Dailiai ir klonas 98 – 279 – 25 yra stambiauogiškumo, o klonas 93 – 157 – 1 yra tiek stambiauogiškumo, tiek produktyvumo donoras. Vaiskrūmiai buvo santykinai atsparūs miltligei, vidutiniškai atsparūs šviesmargei ir deguliams bei skirtingo atsparumo serbentinei (pumpurinei) erkutei. Žiotelių ilgis susijąs su krūmų pažeidimu grybinėmis ligomis. Krūmų pašalimas susijąs su pažeidimu deguliais ir serbentine (pumpurine) erkute. / The aim of the research: to investigate and derive the varieties suitable for the machine harvesting valuable as agronomic donors for the development of new varieties. For this reason 12 different blackcurrant varieties and clones were investigated to determine the most important factors influencing their yield: frost , fungal diseases, currant gall mite, leaf stomata parameters and the relationship between them. Tines is a variety of late – flowering. Varieties Titania, Tiben and a clone 98 – 279 – 25 are donors for a sustainable fruit production. The research revealed that the varieties Tines, Ruben and the Dailiai are the most suitable for the machine harvesting. Varieties of Ores, Ceres, Ruben and clone 98 – 279 – 25 proved to produce maximum number of bunches. The minimum quantity of seeds was found in berries of Titania and Tisel varieties. Varieties Tines, Bona, Ruben, Dailiai and clone 98 – 279 – 25 proved to be a large – berry producing variety, whereas a clone 93 – 157 – 1 was determined to be the most productive and large- berry producing variety. Blackcurrant berry shrubs were relatively resistant to powdery mildew, the average resistance was revealed to leaf septoria and stem canker, and the cultivars demonstrated different resistance to the gall mite. According to the research, the stomata length is associated with the damage of the cultivars by fungal diseases. Application of Plant Extracts for the Prevention of Dental Erosion: An in situ/in vitro Study

Objectives: Antiadherent and antibacterial effects of certain plant extracts have been proven to be beneficial in preventive dentistry. In the present in situ/in vitro crossover study, the impact of plant extracts rich in polyphenols on the erosion-protective properties of the in situ pellicle was evaluated. Methods: Individual splints were prepared for 12 subjects for intraoral exposure of bovine enamel specimens. Following formation of a 1-min pellicle, watery plant extracts(leaves of the wild form of Ribes nigrum , the wild form of Origanum as well as a combination of both) were administered for 10 min in situ. Alternatively, a mouth rinse with fluorides (Elmex Kariesschutz) was performed for 1 min. After further oral exposure for 19/28 min, respectively, slabs were removed and incubated with HCl in vitro over 120 s (pH 2, 2.3, 3). The resulting calcium and phosphate release was quantified photometrically. Slabs with and without a 30-min in situ pellicle served as controls. The modification of pellicle ultrastructure was evaluated by transmission electron microscopy (TEM). Results: Plant extracts modulated the erosion-protective properties of the native in situ pellicle in all test groups in a pH-dependent manner. The combination of R. nigrum leaves and Origanum enhanced the protective properties of the pellicle at all pH values; the administration of this preparation was comparable, yet superior, to the effect of the fluoridated mouth rinse. TEM images indicated that rinsing with R. nigrum leaves /Origanum yielded a distinctly thicker and more electron-dense pellicle. Conclusion: The combination of certain plant extracts offers a novel approach to the complementary prevention of dental erosion.

Investigation Of Chemopreventive And Apoptotic Characteristics Of Turkish Medicinal Plant Rheum Ribes

Rheum species are medicinally important plants due to the presence of anthracene derivatives and in this study antioxidative, cytotoxic, apoptotic and chemopreventive characteristics of R. ribes extracts were evaluated. R. ribes shoot and root dry powder samples were prepared and extracted with ethyl acetate, ethanol and water. The extracts were revealed to be a potential scavenger of DPPH radicals and the chemical composition of the extracts was quantified by colorimetric determination of total phenol (GAE) and flavonoid (CAE) contents. HL&ndash / 60 cells were cultured in the presence of various concentrations of extracts up to 72 hr. R. ribes inhibited the surviv al of HL-60 cells in a concentration- and time-dependent manner, shown by trypan blue and XTT. R. ribes caused HL-60 cells apoptosis via formation of phosphatidylserine externalization, as evidenced by flow cytometry. Exposure of HL-60 cells to higher concentrations of extracts for 72 h resulted in a shift of 87% of the cell population from normal to the early/late apoptotic stage. The R. ribes induced apoptosis may be partially attributed to the activation of caspase-3 and up-regulation of caspase-3 expression was detected in western blot. The significant release of cytochrome c from the mitochondria into the cytosol was observed. The mRNA expression ratio of Bax/Bcl-2 was increased. The apoptosis was also demonstrated by DNA ladder and TUNEL. Chemopreventive effects of R.ribes were investigated at the gene level of CYP1B1 and CYP1A1, and GST enzyme activity against cDNB and concluded that R.ribes modulated activities of these enzymes generally at a time dependent level. T h ese findings suggest that Rheum ribes exhibits potential antioxidant and anticancer properties by inducing caspase-dependent cell death in HL-60 cells.

Establishing Organic Blackcurrants in Atlantic Canada

A study on Prince Edward Island was initiated to assess the impact of organic fertility amendment rate and timing treatments and deflowering on the growth, yield, and berry size and soluble solids, and plant and soil available nutrients of blackcurrants (Ribes nigrum L cv. Titania.). Plants at the site with lower leaf P and K showed lower growth and yield (492-2540 kg ha-1) than the other site (3935-5016 kg ha-1No significant differences were found in final size or 2011 yield at the site with larger bushes, while at the other site the medium spring fertility treatment gave the greatest growth and yield, followed by the high spring fertility treatment. Deflowering increased yield but not growth at the site with recommended ranges of leaf P and K; at the site with P and K deficiencies, growth increased in 2010 and 2011. There was no interaction between deflowering and amendment timing.
