Využití pokročilých statistických metod pro zpracování obrazu fluorescenční emise rostlin ovlivněných lokálním biotickým stresem / Utilization of advanced statistical methods for processing of florescence emission of plants affected by local biotic stress

Chlorophyll fluorescence imaging is noninvasive technique often used in plant physiology, molecular biology and precision farming. Captured sequences of images record the dynamic of chlorophyll fluorescence emission which contain the information about spatial and time changes of photosynthetic activity of plant. The goal of this Ph.D. thesis is to contribute to the development of chlorophyll fluorescence imaging by application of advanced statistical techniques. Methods of statistical pattern recognition allow to identify images in the captured sequence that are reach for information about observed biotic stress and to find small subsets of fluorescence images suitable for following analysis. I utilized only methods for identification of small sets of images providing high performance with realistic time consumptions.

The work deals with nature prefigurations, which can be used in architecture. The first part is concerned with the bionics in architecture in general. There are introduced all available pieces of knowledge falling within this branch. The second part is focused on the nature prefiguration – tree. The tree was analyzed from different points of view. Suitable principles, which were found, were transformed to the field of architecture. On the basis of children's drawings the changing approach to “tree as a home” during adolescence was charted. The work can serve as a comprehensive supporting document for study of bionics for students of architecture and architects and also for the general public. The work should point out that nature is our great standard for any activity.

This diploma thesis is focused on the creation of model tasks (by means of an experiment) for the study of biology at secondary schools and concerns the effects of temperature on plant growth. The selected tasks utilize the new didactic approach by means of the so called inquiry based education. This method of teaching contributes to the development of individual deduction reasoning and encourages the students to actively think about the subject matter. The thesis provides the necessary theoretical background for the topic together with the description of methods and material needed for the conducting the experiments. Furthermore, the thesis describes the necessary instructions and procedures to conduct the experiment and its use in the secondary school teaching practice and presents the requisite methodical guidelines and worksheets for them. Simultaneously, the thesis verifies the didactic applicability of the experiment and the worked out materials through a questionnaire survey and monitored application of the model tasks at secondary schools. Massive parallel sequencing (MPS) data analysis tasks are often computationally demanding and their execution time would take too long using standard computing machines. Thus there is a need for parallelization of this tasks and ability to execute them on a sufficiently powerful computing machines. In the first chapter we describe a newly created platform for resequencing analysis of MPS data - MOLDIMED and novel annotation tool, which is ready to deploy on HPC infrastructure. The second chapter describes MPS approaches in Diamond-Blackfan anaemia (DBA), which is predominantly underlined by mutations in genes encoding ribosomal proteins (RP); however, its etiology remains unexplained in approximately 25% of patients. We performed panel sequencing of all ribosomal genes in DBA patient without previously known molecular pathology. A novel heterozygous RPS7 mutation coding RPS7 p.V134F was found in one female patient and subsequently confirmed in two asymptomatic family members, in whom mild anemia were detected on further examination. Subsequently, we performed whole transcriptome analysis in all family members and patient with RPS7 mutation in comparison with healthy control group and with DBA patients with known mutation in RPS19. This work monitors the response of plants to water deficit and subsequent restoration of water availability with the aim to expand our knowledge about plant behaviour during and after drought stress from the perspective of two plant species with a different type of photosynthesis. A particular attention was paid to the intraspecific differences and their possible changes during stress and post-stress periods. The plant response was monitored as the changes of selected photosynthetic, morphological and developmental characteristics caused by the cessation of watering for six days followed by a six-day period of renewed optimal water supply. The experiments were carried out on plants with different types of photosynthesis - C3 plant Vicia faba L and C4 plant Zea mays L.; for both plant species, three varieties / genotypes were monitored. The response of both plant species to insufficient water supply differed by the extent of changes in relative water content (RWC), chlorophyll fluorescence parameters, the dry mass of individual leaves (developed during the stress period), the ratio between the aboveground dry mass and dry mass of roots. Terrestrial plants and insects account for the majority of the Earth's biodiversity today, and almost half of all insect species are herbivores. Thus, insects and plants share ancient associations date back for more than 400 million years. However, investigation of their interactions stands largely at the beginning in Western Europe. Nearly 4300 plant remains showing various kinds of feeding damages are available for the present study. These trace fossils are classified as so-called functional feeding-groups supply an outstanding example of the complexity, the structure and the changes of terrestrial ecosystems, are not well known during this interval. In Europe, the Neogene is characterized by palaeogeographic re- organization due to the collision of the African with the Eurasian plates. The Neogene plant record in Europe is rich and diverse offering a profound large-scale understanding of the floristic and vegetational development. A database of fossil traces from the Most Basin was compiled and analyzed by various statistical methods in terms of the diversity and intensity of palaeo-herbivory. This work monitors the response of plants to the application of a solution of synthetic brassinosteroid (5-fluor-3α,17β-dihydroxy-5α-pregnan-6-on) under drought stress conditions and subsequent restoration of water availability. The response of two plant species was monitored under six-day period of cessation of watering and under six-day period of renewed optimal water supply. The experiments were carried out on plants with different type of photosynthesis - C3 plant Vicia faba L. and C4 plant Zea mays L.. For both plant species, three varieties/genotypes were used. Photosynthetic, morphological and developmental characteristics were monitored. The brassinosteroid treatment influenced more the morphological than the photosynthetic characteristics. The content of photosynthetic pigments was influenced more than the chlorophyll fluorescence parameters. The differences in morphological characteristics between individual varieties/genotypes were observed. The response to the application of brassinosteroid was observed especially for drought-sensitive variety/genotype. Stanovištní nároky a kompetice mezi původním a invazními druhy Impatiens / Habitat requirements and competition between native and invasive Impatiens species

Three of Impatiens species are widespread in the Czech republic, one is native, two of them invasive. Because all species occur in relatively similar habitats, the question is, whether the invasive species can negatively affect the native species, or even the original can can be displaced. The aim of the thesis was: (i) to compare habitat requirments of the Impatiens species, (ii) to determine if the coexistence of species is possible, (iii) to assess the impact of invasive species to the original species. The investigation was carried out using two sets of permanent plots in 5 localities. In the first set with a total of 84 plots (combinations of occurrence of the three species) site characteristics as tree cover, soil humidity, slope and bare land cover were directly measured and nutrient, light, humidity and soil reaction characteristic assessed using Ellenberg indicator values. The second set contained a total of 45 plots with one of the native-invasive congener pair being removed and the intact controls. Number, cover and height of the plants were assesed. The thesis deals with the issue of compliance in the context of international treaties on protection of endangered species. Two important treaties are subject of the analysis - the 1973 Convention on International Trade in Endangered Species of Wild Fauna and Flora (known as CITES) and the 1946 International Convention for the Regulation of Whaling. My research focuses on the question whether member states comply with their obligations arising from the treaties. If they do not, then I look for a response to the violation such as use of compliance instruments (e. g. sanctions) and its effect on the breaching state, mainly if the response (if there was any) was successful in making the state come back into compliance. Finally, based on experience with CITES, ICRW and other multilateral environmental treaties, I attempt to design some of the new ICRW compliance mechanism features in order to increase its effectiveness.

This diploma thesis is divided into nine chapters. The introductory chapters are focused mainly on currently highly discussed issues concerning criminal acts connected with drug abuse and historical roots and development of contemporary Czech legal regulation. The main part of this diploma thesis consists of the description of every single crime regulated by sections 283-287 of Act No. 40/2009, Criminal Code. The last chapter of this diploma thesis afterwards presents the description of the other crimes which are according to the author the most drug abuse related.
