Goal of this diploma thesis is to create discrete simulation model of a buffer for sorting cars by their color in order to create bigger color blocks in front of upper paint area in paintshop of ŠKODA AUTO a.s. in Mladá Boleslav. Simulation model is created in Plant Simulation software produced by Siemens PLM Software company. Section of this diploma thesis is paid to system analysis of conveyors used in solved area and to suggestion of planned modifications. It is tested by simulation model whether the planned modifications would have positive effect on production characteristic in concern area. Conclusions are deduced from mentioned experiments.

This diploma thesis is based on creation of a simulation model of paint shop in the company ŠKODA AUTO a.s. The model was created in the simulation software Tecnomatix Plant Simulation. It is tested by simulation model whether the planned modifications in a buffer for sorting cars by their color would have positive effect on Perlenkette principle. The Perlenkette principle will help to stabilize supplier processes and reduction of running production times. Conclusions are deduced from carried out simulation experiments.

The output evaluation is make of experiments and comparison of influence of an old logic of choosing a colored block with a new control logic for car body with influence on the correct shifting of car body according to a sequence number. The containers for car body including all other workplaces of the painting area are created in the Plant Simulation program.

One of the main reasons for protein clustering is prediction of structure, function and evolution. Many of current tools have disadvantage of high computational complexity due to all-to-all sequence alignment. If any tool works faster, it does not reach accuracy as other tools. Further disadvantage is processing on higher rate of similarity but homologous proteins can be similar with less identity. The process of clustering often ends when reach the condition which does not reflect sufficient quality of clusters. Master's thesis describes the design and implementation of new tool for clustering of protein sequences. New tool should not be computationally demanding but it should preserve required accuracy and produce better clusters. The thesis also describes testing of designed tool, evaluation of results and possibilities of its further development.

This document describes implementation of photo slideshow editor. This editor can be used to create dynamic sequences from taken photos. Image taken by camera is projected onto three-dimensional sphere and drawn to screen. Then it is posible to rotate and zoom camera over image. Visual output of this editor makes moving over image more real. If panoramic image is used, user is able to free look around. This document describes method which can translate two-dimensional coordinates to three-dimensional space. Shows how to use transformations for moving camera and methods to implement camera zoom. Contains description of user interface and also mentions extensions that were implemented in editor.

Tubulin is the basic building block of microtubules which ensure intracellular transport, morphological changes, the connection of proteins of metabolic pathways, the segregation of chromosomes during mitosis and many other essential processes in plant cells. The localization of tubulin regarding nucleus is strictly controlled and during the interphase, tubulin is usually not present in the nucleus. As we previously showed, one of the exceptions of the presence of tubulin in the nucleus is the cold stress condition. Also, we discovered several plant-specific nuclear export sequences (NES) in tubulin molecules: one in α-tubulin and two in β-tubulin. In this work I found that double mutation of both functional β-tubulin NESes cause slight accumulation of the protein in the nuclei. However, the phenotype of the transgenic plants expressing β-tubulin with mutations is probably not affected by the presence of the mutated protein. The effect of the expression of the mutated β-tubulins was observed in Nicotiana tabacum BY-2 cells as well. The expression of β-tubulin carrying a single NES3 mutation or double mutation of both NESes bring changes of the division activity of the cells, while the NES2 mutation does not have any effect. Monika Švajková Abstract The thesis introduces authors view on the evolution of fairy tales in Europe and its transformation since 19th century in literature, visual arts and related field of knowledge. Its purpose was to create an art project that would connect cultures of Czech Republic and Finland through fairy tales and fine art. Resulting didactical tasks develop fairy tales of one country through contemporary art of the other. The tasks apply to several conceptions of teaching as the implementation took place at the primary art school, high school and lappish art gallery. The art project closes with authors art etudes that expand theme of the forest as an area for updating of fairy tales and simultaneously captures the authors view on the theme of fairy tales. The outcomes and benefits of the thesis were verification of the didactical tasks that would in contemporary art update the classic children literature in context of these two countries.

An infinitesimal fraction of the practically infinite sequence space has achieved enormous functional diversity of proteins during evolution. Intrinsically disordered proteins (IDPs) which lack a fully defined three-dimensional structure are the most likely precursors to today's proteins because of their flexible conformation and functional diversity. But how have these proteins evolved into often rigid and highly specialized protein structures? This evolutionary trajectory has the greatest support in the theory of induced fold whereby the development of the structure was mediated by the interaction and coevolution of primordial unstructured proteins with different cofactors or RNA molecules. Although some random sequences from the sequence space which is not used by nature are also able to form folded proteins the more suitable candidates for evolution of structure and function appear to be random sequences with a high content of disordered which have low aggregation propensity. The selected random protein sequences with high disorder content have been structurally characterized in this work for their further use in evolutionary studies.
