Optimalizace MRI měření slepičích embryí / Optimization of the MRI chicken embryos measurements

This thesis deals with optimization of MRI measuring of phantoms containing the chicken embryo. The theoretical part of thesis is dedicated to the MRI fundamentals and description of pulse sequences. In the next part the options of keeping and monitoring the temperature are being discussed. The thesis also deals with design and realization of phantoms which are used for measuring. A browser of images captured by MRI was programmed in Matlab environment. The program allows users to look through the images with different settings of pulses sequences or kind of phantom. The final part of thesis describes the anatomical structures of chicken embryo and discussion of collected results.

Gene detection in DNA sequences is one of the most difficult problems, which have been currently solved in bioinformatics. This thesis deals with gene detection in DNA sequences with methods using Hidden Markov Models. It contains a brief description of the fundamental principles of molecular biology, explains how genetic information is stored in DNA sequences, as well as the theoretical basis of the Hidden Markov Models. Further is described subsequent approach in the design of specific Hidden Markov Models for solving the problem of gene detection in DNA sequences. Is designed and implemented application, which uses previously designed Hidden Markov model for gene detection. This application is tested on the real data, results of these tests are discussed in the end of the thesis, as well as the possible extension and continuation of the project.

This thesis deals with moving objects detection in video sequences. The principal aim of such detection is to detect and locate motion in the image, separate individual objects, and track these objects. Subsequently, to eliminate shadows, the paper introduces method of motion detection based on Local Binary Patterns together with differential method above the HSV color space. The proposed method provides rapid and accurate movement detection in video sequences.

The subject of my thesis was to genetically modify a potato for increased resistance against its pathogens and pests. In developing a resistant plant, it is quite common to use the same type of molecules that plants use themselves in their defense reactions. In this work I used the gene SPI-2 originating from a honeycomb moth (Galleria mellonella). The protein SPI-2 is a member of serine protease inhibitors. Since the previous attempts of the team to detect the protein in transformed plants haven't been successful, the basic form of the gene was modified by adding a Kozak sequence near the start codon, which should have increased the translation initiation and hence increase the level of the protein. Two constructs were prepared for the transformation: SPI-2-T a SPI-2-Y. They differ by one amino acid, which slightly changes their inhibitory activity. First, the construct SPI-2-T was used for a transient transformation of tobacco Nicotiana benthamiana by agroinfiltration of its leaves. Then both constructs were used for a stable transformation of Solanum tuberosum cv. Desireé. The detection of the protein has not been successful, although the inserted gene was transcribed and his sequence was verified by sequencing. It is therefore most likely that the protein has a low stability in the cytoplasm....

User interface elements and the order in which they are organized and perceived by the user are creating particular narrative - visual narrative. This narrative, which is interpreted by users, should serve as a visual tool to guide users through the process of interaction. Meaning of the interface is created through visual relationships and their hierarchy. Visual narrative can guide the user through the necessary steps of interaction by using an appropriate spacing and enhancing of interfaces elements. Presentational diploma thesis demonstrates that structure of visual narrative has a particular impact on a quality of human-computer interaction. The case study represents application of constructed principles on the selected web pages of airline reservation systems.

This thesis is a part of the project aimed at sequencing the genome and transcriptome of the bank vole (Clethrionomys glareolus). The role of natural selection in the evolution of mitochondrial DNA (mtDNA) has been subject to much discussion; while some studies did not provide evidence that selection affected the phylogeography of the studied species, other considered adaptive evolution important. The bank vole is the key model we use to study the adaptation to climate change. As with other species, the phylogeography of the bank vole has been based on the variation of a small part of mtDNA. The goal of the thesis was to sequence the entire mitochondrial genome for representatives of all main mtDNA lineages of the bank vole using the Sanger and Illumina technologies, and to assess the role of selection and adaptation in the evolution and phylogeography of this species. The adaptive evolution in mtDNA probably was not the main driving force during the postlacial colonization of Europe. However, signatures of adaptive evolution have been found - an amino acid change with possible functional consequences in one gene and an excess of radical changes in physical- chemical properties of amino acids in populations at the latitudinal (northern and southern) extremes of the bank vole distribution. Význam fyzioterapie v prognóze pacientů s Pierre Robinovou sekvencí / The importance of physical therapy in prognosis in patients with Pierre Robin sequence

Clefts of orofacial komplex are the most common birth development defects. One of them is Pierre Robin sequence, which is characterized by presence of micrognathia, glossoptosis and upper airway obstruction. Because of these symptoms, isolated cleft palate "U" or "V" shaped can be part of the diagnose. Other clinical manifestations are feeding difficulties, obstructive sleep apnea syndrome and / or gastroesophageal reflux disease. The aim of the treatment is to secure the airway and the child's thriving after birth by using conservative or invasive treatment methods. Choosing of methods and procedures should be established by multidisciplinary cooperation. The other part of diploma thesis is trying to find out, which clinical manifestation are the most common in patients from the Czech Republic compare to foreign patients and the differences in management of the treatment. Furthermore we are trying to investigate, whether there is higher percentage of patients with delayed neuromotor development among PRS patients, which are under physical therapy. The last part is trying to find out the relationship between the poor posture and PRS diagnose in children age 4 - 6 years.
